Primary evaluation of immune responses in mice after a single mRNA immunization with Echinococcus granulosus Eg95 and Coenurus cerebralis TM16.

Echinococcus granulosus and Taenia multiceps cause zoonotic cystic diseases. Studies have shown that Eg95 from E. granulosus is highly conserved, exhibits excellent immunogenicity, and serves as a critical candidate antigen. In recent years, the TM16 recombinant protein antigen has been one of the most studied antigens for preventing C. cerebralis and demonstrates optimal experimental immunogenicity. This study developed both recombinant protein and mRNA-LNP vaccines targeting the Eg95 and TM16 antigens. Immunization of BALB/c mice showed that both vaccines significantly increased specific ant

Comparative assessment of immunochromatographic test kits using low-molecular-weight antigens from cyst fluids of two different genotypes of Taenia solium for serodiagnosis of human cysticercosis.

Human cysticercosis is a serious zoonosis caused by infection with larvae (cysticerci) of the pork tapeworm, Taenia solium. Infection can involve the nervous system, causing chronic headache and intracranial hypertension, focal neurological deficits, epileptic seizures, and paralysis. The disease is found in developing countries, where porcine cysticercosis is prevalent and undercooked pork is habitually consumed. This study aimed to develop immunochromatography-based test (ICT) kits, using low-molecular-weight antigens purified from cyst fluids of Latin American and Asian genotypes of T. soli

Core-shell heterostructured nanozyme for alveolar echinococcosis treatment via targeted electron transfer and catalytic reactive oxygen species generation.

Alveolar echinococcosis (AE), a severe parasitic disease, poses a significant therapeutic challenge due to the limitations of surgery and albendazole chemotherapy, particularly the difficulty in achieving targeted drug delivery and disrupting the robust structure of the protoscoleces (PSCs). This study introduces a novel therapeutic strategy of "targeted binding-electron transfer (ET)-osmotic disruption" to address these challenges. We designed a core-shell heterostructure nanoplatform (FZ) composed of a magnetite (Fe3O4) nanozyme core and a zinc oxide (ZnO) semiconductor shell. This architect

Zoonotic Helminths in the Southern Peruvian Altiplano: A Four-Year Sero-Epidemiological Study and One Health Policy Implications.

We assessed the prevalence of three helminthic zoonoses-echinococcosis, fasciolosis and the taeniosis/cysticercosis complex-among residents of the Chucuito Health Network (Puno Health Region, Peru) over four years (2018-2021). Sera (n = 910) were analysed by ELISA to detect pathogen-specific antibodies, following national protocols. Echinococcosis predominated, whereas fasciolosis and taeniosis/cysticercosis occurred at comparatively low levels. Prevalence ranged from 4.4-9.2% for echinococcosis, 1.1-4.9% for fasciolosis, and 1.1-2.7% for taeniosis/cysticercosis across the four years. Prevalen

Comparison of Effectiveness Between Cysticidal and Surgical Treatments in Patients with Intraventricular Neurocysticercosis: A Single-Center Experience.

Neurocysticercosis (NCC) remains a major public health problem in endemic countries. Clinical manifestations and therapeutic strategies vary depending on the location of the parasite. While the benefits of cysticidal treatment are well established for parenchymal and subarachnoid NCC, the optimal management of intraventricular NCC (IVNCC) remains controversial. We conducted a retrospective study of 51 patients: 37 (72.54%) received cysticidal treatment as initial therapy and 14 (27.45%) underwent neurosurgical intervention. Although six months after treatment, the proportion of patients with i

MRI Characteristics of Intraspinal Sparganosis: A Case Report.

BACKGROUND: Intraspinal sparganosis constitutes an uncommon parasitic infection. The absence of distinct clinical manifestations and imaging characteristics frequently leads to its misdiagnosis as a tumor, cyst, or hematoma. CASE PRESENTATION: In this study, we present a case involving a 57-year-old female patient with a history of consuming raw or undercooked frog meat and pork. Imaging studies identified an intraspinal occupying lesion. The patient subsequently underwent surgical intervention, which resulted in a pathological diagnosis of intraspinal sparganosis. Following this diagnosis, a

Exploring the Potential Role of Interleukin-33 in Inducing Eosinophil-Mediated Fibrosis in Hepatic Alveolar Echinococcosis.

BACKGROUND: Alveolar echinococcosis (AE) is a serious zoonotic parasitic disease. This study aimed to investigate the mechanisms underlying the formation of the dense fibrotic band surrounding hepatic alveolar echinococcosis (HAE) lesions, which impedes chemotherapeutic drug penetration. Additionally, the roles of Interleukin-33 (IL-33) and eosinophils in the progression of fibrosis within this band were examined. METHODS: IL-33/suppression of tumorigenicity 2 (ST-2) expression levels were compared between patients with HAE and healthy controls, as well as between close to lesion tissues (CLT

Macrophage-derived S100A9 drives liver fibrosis in Echinococcus multilocularis infection by up-regulating secreted protein acidic and rich in cysteine (SPARC) in hepatic stellate cells.

Alveolar echinococcosis (AE), caused by Echinococcus multilocularis(E. multilocularis) infection, is a severe parasitic disease characterized by progressive liver fibrosis. Although the damage-associated molecular pattern molecule S100A9 has been implicated in some inflammatory and fibrotic diseases, its role in AE induced liver fibrosis remains unclear. This study aimed to elucidate the function and mechanism of S100A9 in E. multilocularis induced liver fibrogenesis. Using proteomic analysis, we identified significant up-regulation of S100A9 in the livers of infected mice. In a stem cell-base

First identification of adult Mesocestoides vogae in a domestic dog in Japan.

In recent years, several cases of Mesocestoides vogae infection have been reported in domestic dogs in Japan. These cases typically involved the detection of tetrathyridia and segmented worms with immature proglottids, but not mature adults with gravid proglottids. In this study, mature and gravid proglottids of M. vogae were identified in a domestic dog from Niigata Prefecture. Morphological examination confirmed the parasite to be a Mesocestoides species based on the presence of a well-developed parauterine organ. Further molecular analysis identified the parasite as M. vogae. This is the fi

Study of Interleukin 1β Gene Polymorphisms in Tunisian Paediatric Population With Cystic Echinococcosis.

Cystic echinococcosis (CE) is a highly common zoonosis worldwide and Tunisia remains the most endemic country in North Africa. The variability of clinical manifestations of CE depends on predisposing factors related to the interaction between host genetic and immunological factors. This study investigates the association between single nucleotide polymorphism (SNP) of IL-1β (rs16944 and rs1143634), an important pro-inflammatory cytokine in the CE immune response and susceptibility to CE in a Tunisian population. The case group included 152 paediatric patients diagnosed with CE, whereas the con

Cognitive sequelae associated with parenchymal and extraparenchymal neurocysticercosis.

BACKGROUND: While the incidence of neurocysticercosis (NC) with viable parasites has been declining in Mexico and some other countries, this trend is not observed globally, and the burden of patients who have previously suffered from NC remains considerable. Cognitive manifestations are often overlooked in the clinical management of NC. In routine practice, patients are generally considered cured once viable parasites are no longer detectable, despite the potential for persistent neurological sequelae. OBJECTIVES: To evaluate the frequency, characteristics, and severity of cognitive sequelae

Percutaneous vs surgical management of World Health Organization cystic echinococcosis 1 and 3a liver hydatid cysts.

BACKGROUND: Hydatid cyst disease of the liver remains a significant public health problem in endemic regions. While surgical treatment has traditionally been the mainstay of therapy, minimally invasive percutaneous approaches have emerged as safe and effective alternatives, especially for selected World Health Organization (WHO) cystic echinococcosis (CE) 1 and CE3a cysts. Comparative data on efficacy, complication rates, and clinical outcomes between the two methods are essential for guiding optimal treatment selection. AIM: To compare and evaluate the efficacy, safety, complication rates, a

The gut microbiota mediates depression-like behaviors in mice with chronic Echinococcus multilocularis infection.

Alveolar echinococcosis (AE), a chronic parasitic disease caused by Echinococcus multilocularis (E. multilocularis), remains poorly characterized with respect to central nervous system (CNS) involvement, and its long-term effects on mental health have not been systematically investigated. In this study, we established a BALB/c mouse model of chronic E. multilocularis infection and applied an integrative framework combining behavioral assessments, histomorphological analyses (hematoxylin-eosin staining, Nissl staining, and transmission electron microscopy), cytometric bead array (CBA), and mult

A new case of Echinococcus ortleppi infection diagnosed by next-generation sequencing in China.

Cystic echinococcosis is an important parasitic zoonosis infecting numerous humans with high morbidity and mortality. As one of the etiologic agents, Echinococcus ortleppi infection in humans has been very rare. In this study, a 27-year-old man was diagnosed with cystic echinococcosis in Wuhan City of China, a non-endemic area. Next-generation sequencing identified that the etiologic agent was E. ortleppi. Its complete mitochondrion sequence (13,600 bp) has 99.92% identity to E. ortleppi from cattle in Japan. This is the third reported E. ortleppi infection case in China. Although extensive ep

Impact of human adherence level to control measures on cystic echinococcosis transmission.

Cystic echinococcosis (CE) is a widespread zoonotic disease that poses a serious threat to both human health and livestock production. Its control effectiveness largely depends on human adherence. A game-theoretic epidemiological model is developed to couple disease transmission with human behavioral responses, aiming to investigate the effects of human adherence on CE transmission. The equilibria and threshold dynamics are derived, and numerical simulations are conducted to explore how key factors influence disease prevalence, behavioral strategy switching, and control effectiveness. The resu

Preclinical evaluation of Senna alata leaf extract against Hymenolepis diminuta: a promising natural anthelmintic solution.

BACKGROUND: Senna alata is widely recognized for its wide range of therapeutic benefits, including its antioxidant activity. Our previous research demonstrated its non-toxic nature in mice and its modulatory effects on the antioxidant system of the zoonotic parasite Hymenolepis diminuta. However, understanding the mode of action of S. alata during active parasitic infection is essential for evaluating its antiparasitic potential. This study aims to explore the in vivo efficacy of alcoholic leaf extract of S. alata against H. diminuta infection in a rodent model, focusing on both antiparasitic
