Low Skeletal Muscle Index Predicts Neoadjuvant Immunochemotherapy Tolerance and Survival in Oesophageal Squamous Cell Carcinoma.

BACKGROUND: Low skeletal muscle index (SMI) may identify host vulnerability in patients receiving intensive cancer therapy. However, its association with treatment tolerance and outcomes during neoadjuvant immunochemotherapy (nICT) for oesophageal squamous cell carcinoma (OSCC) remains unclear. METHODS: This multicentre retrospective cohort included 461 patients with locally advanced OSCC who initiated nICT between 2020 and 2023; 404 underwent oesophagectomy. Pretreatment SMI at the third lumbar vertebra was measured by two blinded radiologists. Low SMI was defined using sex-specific outcome-

Activity over Abundance: ADAMTSL4 Unlocks Latent TGFβ1 in Cancer Cachexia.

Cancer cachexia is increasingly recognized as a coordinated, tumor-induced, unsustainable alteration in interorgan communication. Machado and colleagues identify a new molecular mechanism that promotes cachexia, in which tumor-secreted ADAMTSL4 drives tissue wasting by promoting local activation of latent TGFβ1, revealing post-secretory control of ligand activation as a potential therapeutic vulnerability in cancer cachexia. See related article by Machado et al., p. 2172.

Tumor-derived SAA1-TLR4 signaling drives tumor-to-muscle communication in pancreatic cancer cachexia.

Cancer cachexia limits treatment tolerance and survival in pancreatic ductal adenocarcinoma (PDAC), yet the tumor-derived signals driving tissue dysfunction remain poorly understood. Here, we identify serum amyloid A1 (SAA1) as a mediator of tumor- to-host communication acting through Toll-like receptor 4 (TLR4). Tumor-derived SAA1 was elevated in human PDAC and in a mouse PDAC model and disrupted both myofiber and muscle stem cell (MuSC) homeostasis. Genetic reduction of tumor-derived SAA1 uncoupled tumor progression from host wasting, preserving muscle mass and function and prolonging surviv

Clove oil-derived β-caryophyllene carried in a self-nanoemulsifying drug delivery system improves adjuvant-induced arthritis in rats.

OBJECTIVES: β-Caryophyllene (β-Cary) exhibits anti-inflammatory and antioxidant activities. It can be obtained from clove oil, which is rich in eugenol; following eugenol removal, β-Cary can be isolated as the major constituent. However, the high lipophilicity of β-Cary limits its oral bioavailability. The present study investigated the anti-arthritic effects of β-Cary formulated in a self-nanoemulsifying drug delivery system (β-SNEDDS) in Holtzman rats with complete Freund's adjuvant-induced arthritis, emphasizing articular inflammation and systemic oxidative stress. METHODS: Healthy and adj

Gilthead seabream (Sparus aurata) mucus core O-glycosylation changes in Enteromyxum leei parasitized intestine.

The gilthead seabream, Sparus aurata, is an important fish within the aquaculture industry. Enteromyxum leei causes chronic enteritis, resulting in reduced nutrient absorption and growth, cachexia and/or death of the fish. There are currently no applied treatments for this disease. To investigate the first barrier pathogens encounter, the mucus, we characterized the gilthead seabream intestinal mucus glycome from five control and five E. leei infected gilthead seabream using mass spectrometry and investigated the link between identified glycans and infection parameters, gene expression and mic

Fibro-Adipogenic Progenitor Ablation Triggers Muscle Atrophy Through Cell Death-Induced Inflammation.

BACKGROUND: Skeletal muscle mass maintenance involves coordination of myofibers with mononuclear cell populations, including satellite cells, resident macrophages and fibro-adipogenic progenitors (FAPs). FAPs, identified by surface expression of PDGFRα, are important contributors to muscle homeostasis, as genetic ablation of FAPs induces rapid muscle atrophy. However, mechanisms underlying this response remain poorly understood. METHODS: We utilized tamoxifen-inducible FAP specific diptheria toxin (DTA) mice (PdgfraCre-ERT2/+; Rosa26^DTA/+) to evaluate consequences of FAP deletion, measuring

Complete mitochondrial genome analyzes of four gerbil species (Rodentia: Gerbillinae) distributed in Türkiye.

BACKGROUND: The genus Meriones Illiger, 1811 is widely distributed in the Middle East, Northern Africa, and Central Asia. In spite of many studies evaluating the gerbil phylogeny, the systematics of this genus is controversial because studies analyzing a single gene region yield inconsistent results. The presented research provides the first complete mitogenomes of four gerbil species (Meriones crassus, Meriones persicus, Meriones tristrami, and Meriones vinogradovi) distributed in Türkiye and novel and strong results to clarify the phylogenetic relationships of these gerbils. METHODS: Next G

[Brain imaging in cognitive disorders - from diagnosis to treatment and monitoring].

BACKGROUND: Cancer cachexia is observed in more than 50% of advanced cancer patients, and impairs quality of life and prognosis. A variety of pathways are likely to be dysregulated. Hence, a broad-spectrum understanding of the disease process is best achieved by a discovery based approach such as proteomics. RESULTS: More than 300 proteins were identified with > 95% confidence in correct sequence identification, of which 5-10% were significantly differentially expressed in cachectic tissues (p-value of 0.05; 27 proteins from gastrocnemius, 34 proteins from soleus and 24 proteins from heart).

B-Cell Lymphoma Following an Indolent Course Over 30 Years with Immunophenotypic Changes at Each Recurrence.

INTRODUCTION: Diffuse large B-cell lymphoma (DLBCL) comprises biologically distinct subtypes, but whether cell-of-origin-related features remain stable over long disease courses is unclear. CASE DESCRIPTION: A woman was initially diagnosed with CD20-positive B-cell lymphoma in 1988 and treated with cyclophosphamide, vincristine and prednisolone (COP) chemotherapy. She experienced a first relapse in 1998 and received cyclophosphamide, doxorubicin, vincristine and prednisolone (CHOP) chemotherapy. The second relapse occurred in 2008 and was managed with rituximab. At the third relapse in 2011,

Activin A-Targeted Therapy in Cancer: An Updated Review on Challenges and Opportunities in Clinical Translation.

Activin A (ActA) is a cytokine from the TGF-β superfamily that mediates a vast number of physiological mechanisms, mainly through the SMAD signaling pathway. Growing evidence indicates that ActA overexpression is also correlated with poor prognosis in cancer patients and several tumor characteristics, including cancer proliferation, metastasis, immunosuppression, drug resistance, cachexia, and cancer-associated fibroblast activation. As such, ActA-targeted therapy has been viewed as a potential adjuvant therapy alongside other anti-cancer modalities that may result in more efficient anti-cance

MEF2C: A Novel Transcription Factor Implicated in Human Malignant Tumors.

BACKGROUND: Myocyte enhancer factor 2C (MEF2C) is a pivotal transcription factor that is responsible for maintaining myocyte differentiation. MEF2C is multifunctional, participating in diverse biological processes, including cardiac morphogenesis, angiogenesis, neurogenesis, and cortical development. Emerging evidence has identified MEF2C as a novel oncogene with dual regulatory functions in tumorigenesis. However, the mechanisms by which MEF2C regulates the progression of various malignant tumors are unknown. Therefore, it is crucial to further investigate the multiple signaling pathways unde

NSUN2 promotes the growth, metastasis, and glutamine metabolism of non-small cell lung cancer.

OBJECTIVES: As a 5-methylcytosine (m5C) methyltransferase, increased NOP2/Sun RNA methyltransferase 2 (NSUN2) has been revealed to promote the progression of non-small cell lung cancer (NSCLC) through m5C modification. Herein, this study aimed to investigate the potential molecular mechanisms underlying the high NSUN2 expression in NSCLC, and the potential downstream m5C mRNAs of NSUN2 in promoting NSCLC progression. METHODS AND MEASUREMENTS: Functional analyses were conducted using in vitro 3- (4, 5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide (MTT), 5-ethynyl-2'-deoxyuridine, Trans

Combined Impact of Neoadjuvant Therapy and Preoperative Cachexia in Patients Undergoing Pancreatoduodenectomy: Is There a "Double Jeopardy"? A National Cohort Study Investigating the Association with Short- and Long-Term Outcomes.

BACKGROUND: Cachexia is associated with worse postoperative outcomes, but the added role of neoadjuvant therapy (NAT) is unclear. This study evaluated whether preoperative cachexia and NAT act as a "double jeopardy" after pancreatoduodenectomy. PATIENTS AND METHODS: A nationwide observational cohort study was conducted using the Norwegian NORGAST registry (2016-2023). Adults undergoing pancreatoduodenectomy for malignancy were included. Cachexia was defined by consensus weight-loss criteria. Modified Poisson and Cox models (with a cachexia and NAT interaction term) estimated adjusted risk rat

The OtR tumor recurrence and metastasis model reveals doxorubicin-induced tumor shrinkage doesn't guarantee prolonged survival.

BACKGROUND: In preclinical research, tumor growth inhibition in subcutaneous models is frequently employed to evaluate therapeutic efficacy; however, such models often lack clinical translatability. METHODS: To better approximate clinical reality, taking the case of doxorubicin treatment, we utilized an orthotopic transplant and resection (OtR) strategy to systematically assess the effects of neoadjuvant chemotherapy, adjuvant chemotherapy, and their combination on tumor growth, recurrence, and malignant progression. RESULTS: Surprisingly, none of the treatments improved mouse survival, with
