Inflammatory bowel disease (IBD) is a chronic inflammatory disorder resulting from a combination of genetic susceptibility, environmental factors, and an abnormal immune response of the intestinal immune system to commensal microorganisms. The gut microbiota and its metabolites play a pivotal role in maintaining intestinal immune homeostasis. Recent advances indicate that dysbiosis of the microbiota is accompanied by alterations in its metabolic functions. Abnormal levels of key metabolites, particularly short-chain fatty acids (SCFAs), tryptophan derivatives, and secondary bile acids, are clo

Signal Transduction and Activation of Transcription 6 (STAT6) is a key molecule in the Janus Kinase/Signal Transducer and Activator of Transcription (JAK/STAT) signaling pathway. Primarily induced by Interleukin-4(IL-4) and Interleukin-13(IL-13), STAT6 undergoes phosphorylation and translocates to the nucleus, where it regulates gene transcription. While the abnormal activation and oncogenic functions of STAT6 in various solid tumors are well documented, its precise biological role in the hematopoietic system has not yet been systematically investigated. Compared with solid tumors, STAT6 activ

Metabolic regulation is central to the tumor suppressor function of p53. By analyzing the human patients with autoimmune diseases, we found that p53 expression was significantly reduced in Treg cells, negatively correlating with abnormally elevated BCL-6 levels. p53 loss causes dysregulated immune homeostasis and dampens Treg function in vitro and in vivo. Mechanistically, p53 transcriptionally activates ALDH6A1 expression and propionyl-CoA anabolism to upregulate functional Treg gene expression via histone propionylation. Treg-specific knockout of ALDH6A1 phenocopies the autoimmune responses

A growing consensus indicates that periodontitis (PD) adversely affects glycemic control in type 2 diabetes, though its underlying mechanisms remain unclear. Here, we report that PD, induced by long-term oral ligatures, significantly aggravated hyperglycemia and hepatic gluconeogenesis in high-fat diet-induced obese mice, with its impact on insulin resistance being more pronounced in the liver than in adipose or muscle tissue. Immunologically, PD increased systemic B cell abundance, promoted hepatic B2 cell mobilization, facilitated the expansion of Kupffer cells and adipose tissue macrophages

BACKGROUND: Disrupted sleep and shift work are linked to multiple adverse health outcomes, but their association with reproductive endocrine disorders (REDs) and underlying mechanisms remain unclear. Understanding this relationship is crucial for public health, given the growing prevalence of sleep disturbances and shift work in modern societies.
PURPOSE: To investigate the impact of sleep traits and shift work on RED risk in women, explore inflammatory mediation, and assess potential causality using Mendelian randomisation (MR).
METHODS: We analysed 244,561 women in the UK Biobank. Sleep du

INTRODUCTION: Sirolimus, also known as rapamycin, is an mTOR receptor inhibitor that suppresses cell proliferation and angiogenesis, demonstrating efficacy against multiple types of vascular anomalies. However, sub-therapeutic concentrations (below effective levels) and supra-therapeutic concentrations (leading to adverse reactions) of sirolimus may both negatively impact patient treatment outcomes. This study aimed to establish optimal models to predict the risk of sirolimus exposure using machine learning, ensure that sirolimus blood concentrations remain within the therapeutic range, and th

Phelan-McDermid syndrome (PMS) is a relatively frequent cause of syndromic intellectual disability (ID) and autism spectrum disorder (ASD). It is typically caused by genetic alterations in the 22q13 chromosomal region, most often involving heterozygous deletions or mutations in the SHANK3 gene. More than half of affected individuals exhibit functional impairments in speech, cognition, motor skills, and behavior. Despite multiple ongoing therapeutic programs, objective and scalable liquid biomarkers to support patient stratification and to monitor disease course or treatment response are still

INTRODUCTION: Idiosyncratic drug-induced liver injury (iDILI) is an unpredictable adverse drug event that often involves an abnormal immune response. iDILI is a challenge in the development of new drugs and the diagnosis in clinical practice. Hepatocyte-like cells generated from monocytes of iDILI patients are being developed as an ex vivo assay to enhance understanding of underlying molecular mechanisms and to assist in drug causality assessment, especially in cases involving multiple agents.

INTRODUCTION: Solid tumors are characterized by densely packed extracellular matrices and limited vascularization, creating significant resistance to both diffusive and convective transport. Tumor growth depends on complex flow-structure interactions across multiple scales, while vascular abnormalities and enhanced permeability elevate interstitial pressure in the tumor microenvironment.
METHODS: In this study, we developed an integrated computational framework with a theoretical modeling framework that couples three phase, viscous-laminar, transient simulations of glycocalyx-patched tumor ve

Acute myeloid leukemia (AML) is an aggressive hematologic malignancy characterized by the uncontrolled expansion of undifferentiated myeloid precursors in the bone marrow. Hypomethylating agents (HMAs) such as azacitidine and decitabine can reverse abnormal DNA methylation, promote leukemic cell differentiation, and enhance immune recognition, yet relapse and therapeutic resistance remain major challenges. In this study, we found that long-term, low-dose HMA treatment upregulated CD70, NK receptor ligands, and CD1d on AML tumor cells, rendering them more susceptible to chimeric antigen recepto

Acute myocardial infarction is an essentially inflammatory disease. The inflammation is predictive of adverse clinical outcomes in coronary artery disease, and the Syntax Score was developed as an angiographic scoring system to anatomically classify the complexity of atherosclerosis plaques. The inflammatory response leads to an increase in mean platelet volume (MPV), neutrophil/lymphocyte ratio (NLR) and the appearance of nucleated red blood cells (NRBCs) in the peripheral blood of the patients with acute myocardial infarction. These hematological variables express systemic inflammation and h

Diabetic kidney disease (DKD) is a major microvascular complication of diabetes mellitus and the leading global cause of end-stage renal disease. While chronic hyperglycemia and classic metabolic pathways such as protein kinase C activation, advanced glycation end product (AGE) accumulation, and renin-angiotensin system overactivity elucidate many features of DKD, they do not fully account for its complex inflammatory pathology. Recent studies have highlighted neutrophil extracellular trap (NET) formation, or NETosis, as a critical mechanism linking metabolic stress to immune dysregulation in

The gut-brain axis is increasingly implicated in Alzheimer's disease (AD) pathogenesis, but the potential correlation between AD-associated gut microbiota and central inflammation remains largely unclear. This study aimed to explore their correlative link, with a focus on changes and involvement of Th17 cell-related factors in the gut-brain axis. Healthy C57BL/6J mice were pretreated with antibiotics for 1 week to deplete the indigenous gut microbiota, followed by 2 weeks of fecal microbiota transplantation (FMT) using feces from APP/PS1 AD model mice. Abnormal intestinal mucosal immunity plays a crucial role in ulcerative colitis (UC). Autotaxin (ATX) can promote T cell migration and was reported to have a regulatory effect on Th17 cells, while sphingosine-1-phosphate (S1P) and its receptors (S1PRs) modulate Th17/Treg balance and inflammation, with S1PR modulators approved for UC. ATX can catalyze sphingosylphosphorylcholine (SPC) to produce S1P; however, the relationship between ATX and S1P/S1PRs in UC is unclear. Understanding the role of ATX-S1P/S1PRs in intestinal immunity can provide new treatment strategies for intestinal inflammatory

Chemotherapy is one of the important means to improve the 5-year survival rate of nasopharyngeal carcinoma (NPC) patients, and commonly used chemotherapy drugs include cisplatin and gemcitabine. Previous studies have shown that chemotherapy can induce some tumor cells to exhibit a senescent phenotype, thereby altering the tumor microenvironment towards tumor immune suppression and tumor recurrence. However, the role and detailed mechanism of chemotherapy in the senescence of NPC cells are not yet clear, and further in-depth research is needed. This study focuses on the mechanism of cisplatin-i

NDUFA5 encodes a structural subunit of mitochondrial complex I (NADH:ubiquinone oxidoreductase) located in the peripheral arm of the enzyme complex. Complex I is the largest enzyme of the mitochondrial respiratory chain and is essential for oxidative phosphorylation. There are many well-characterized conditions associated with nuclear-encoded mitochondrial complex I dysfunction, including Leigh syndrome, leukoencephalopathy, lethal infantile mitochondrial disease, hypertrophic cardiomyopathy, and exercise intolerance. The vast majority of these nuclear-encoded mitochondrial complex I deficienc

Obesity involves abnormal adipose tissue remodeling, including increased fat cell number (hyperplasia) and size (hypertrophy), along with dysregulated adipogenesis, function, and turnover of white, beige, and brown fat cells. These changes cause chronic inflammation and insulin resistance, leading to metabolic diseases. Post-translational modifications (PTMs), especially ubiquitin and ubiquitin-like modifications (UBLs), regulate adipose development by attaching to proteins, affecting their activity, stability, and localization through a cascade involving E1, E2, and E3 enzymes.
