Antiretroviral raltegravir, a selective human immunodeficiency virus type 1 integrase inhibitor, shows anti-Alzheimer's potential against amyloid-beta1-42-induced neurodegeneration.

There is growing evidence supporting the potential role of microbial infections in the aetiology of Alzheimer's disease (AD) and the protective role of anti-viral therapies. However, not much is known about the molecular mechanisms underlying their effects. We aimed to explore the modulatory role of raltegravir on monomeric amyloid beta 1-42 (m-Aβ1-42)-induced molecular alterations in cellular models of AD. Here we show, using the flow cytometry technique combined with specific monoclonal antibodies, that raltegravir significantly abolishes the m-Aβ1-42-stimulating effect on p-tau 181, and tha

Prevalence of elevated liver enzymes and its associated factors among people living with HIV on dolutegravir-based ART at Debre Markos Comprehensive Specialized Hospital, Ethiopia.

Elevated liver enzymes remain a clinically important adverse effect of antiretroviral therapy (ART) in people living with HIV (PLHIV). Although dolutegravir (DTG)-based ART is recommended as first-line therapy in Ethiopia, evidence on the burden and factors associated with elevated liver enzymes among PLHIV receiving these regimens remains limited. To assess the prevalence and factors associated with elevated liver enzymes among PLHIV receiving DTG-based ART at Debre Markos Comprehensive Specialized Hospital (DMCSH) in Northwest Ethiopia. An institutional-based cross-sectional study was conduc

Emergence of newly reported HIV-1 recombinants CRF120_0107 and CRF149_01B in Guangxi Zhuang Autonomous Region, China: Molecular evidence and public health challenges.

The Guangxi Zhuang Autonomous Region (Guangxi), located at the junction of Southwestern and Southern China, is a region with a high prevalence of human immunodeficiency virus type 1 (HIV-1). Understanding the genomic features and transmission dynamics of circulating HIV-1 strains is crucial for effective epidemic control. This study aimed to analyze the near-full-length genomic (NFLG) characteristics, recombination patterns, and cross-provincial transmission relationships of the newly reported recombinants CRF120_0107 and CRF149_01B in Guangxi. We selected six near-full-length HIV-1 genomic se

Antiproliferative effects of HIV integrase strand transfer inhibitors in experimental models for cancer research: A narrative review.

A growing body of literature suggests that integrase strand transfer inhibitors (INSTIs), a cornerstone of antiretroviral therapy for the treatment of HIV infection, seem to possess antiproliferative properties against some types of cancer and/or anti-invasion effects. Preclinical studies describe as INSTIs, including dolutegravir, raltegravir, cabotegravir, and elvitegravir and some 1,2,3-triazole derivatives, can inhibit the growth of various cancer cell lines and some of them seem to also have direct inhibitory effects on human endogenous retrovirus reactivation. This review comprehensively

Deep vein thrombosis as a public health priority: wHO's agenda for Africa 2030.

Deep vein thrombosis (DVT) is a serious vascular condition characterized by the formation of blood clots in deep veins, predominantly in the lower limbs. Despite its significant morbidity and mortality worldwide, DVT remains an under-recognized public health issue in Africa, where limited awareness, diagnostic challenges, and resource constraints contribute to suboptimal prevention and management. The rising burden of risk factors such as obesity, HIV infection, infectious diseases, and sedentary lifestyles highlights the urgent need to prioritize DVT within the continent's healthcare agenda.

Immune checkpoint inhibitors in people living with HIV and cancer: a Kaplan-Meier-derived individual patient data meta-analysis.

INTRODUCTION: Antiretroviral therapy has enabled people living with HIV (PLWH) to achieve near-normal life expectancy, but non-AIDS-defining cancers are increasingly frequent. Immune checkpoint inhibitors (ICIs) may be effective in this population, although evidence remains limited because PLWH have often been excluded from trials. This study assessed the efficacy and safety of ICIs in PLWH. METHODS: We conducted a systematic review and meta-analysis using Medline, Scopus, Web of Science, and Cochrane Library. Studies reporting outcomes of PLWH with malignancies treated with ICIs were include

Five-Year Immune Recovery After ART Initiation Among PLWH Aged ≥15 Years with Baseline Immunosuppression - China, 2013-2024.

INTRODUCTION: Although antiretroviral therapy (ART) suppresses human immunodeficiency virus (HIV) replication in most people living with HIV (PLWH), 10%-40% show inadequate CD4 reconstitution despite sustained virologic suppression, increasing risks of AIDS- and non-AIDS-related events. Prior studies have suggested that immunological nonresponse (INR) is particularly common among individuals starting ART with immunosuppression. In China, real-world evidence for long-term immune recovery in immunosuppressed PLWH remains limited. METHODS: Immunosuppression was defined as a baseline CD4<350 cell

A multicentre evaluation of cardiovascular risk prediction tools in people living with HIV across England (ECARTH study).

INTRODUCTION: It remains unclear which cardiovascular disease (CVD) risk scores are optimized for people living with treated HIV with high rates of viral suppression. We evaluated the performance of commonly used CVD risk scores in a population of primarily well-treated HIV patients in England. METHODS: A multi-centre retrospective cohort analysis was performed, including people living with HIV (PLWH) above the age of 40 with no prior major adverse CVD events (MACE), regularly attending 7 HIV clinics in England during 2014. Outcomes were MACE over the following 5 years: myocardial infarction,

Uptake of Influenza, Pneumococcal, and COVID-19 Vaccines Among People Living with HIV: Findings from the Ontario HIV Treatment Network Cohort Study.

People living with HIV are at higher risk of severe illness due to common respiratory pathogens. We estimated uptake of influenza, pneumococcal, and COVID-19 vaccines among people engaged in HIV care in Ontario, Canada, and identified factors associated with vaccine uptake. We conducted a cross-sectional analysis of interviewer-administered, annual questionnaires (2021-2023) linked to medical charts and laboratory data. We used modified Poisson regression to calculate adjusted prevalence ratios and 95% confidence intervals for self-reported uptake of influenza (yes vs. no, last year), pneumoco

Kaposi sarcoma herpesvirus-associated diseases in solid organ transplantation: Toward a coordinated national strategy in the United States.

Although Kaposi sarcoma-associated herpesvirus (KSHV)-associated disease (KAD) has long been recognized as a complication of transplantation, in the past 5 years, the incidence of KAD among solid organ transplant recipients in the United States has increased greater than 5-fold, exposing gaps in screening and monitoring protocols. The United States lacks a coordinated framework for KSHV risk assessment and posttransplant monitoring, primarily because serologic and molecular testing is not yet widely available. Current laboratory-developed tests are not standardized across laboratories and anal

Mother-to-child HIV exposure and its relation with neurodevelopmental disorders: a systematic review and meta-analysis.

BACKGROUND: Mother-to-child Exposure to HIV (MCE) is a growing concern. While antiretroviral therapy has reduced vertical transmission rates, the inflammatory response and adverse neurobehavioural outcomes in children are conflicting. Thus, this review aims to synthesise existing evidence between HIV exposure and neurodevelopmental outcomes in MCE compared to unexposed children. METHODS: A comprehensive search was conducted to screen systematic reviews until July of 2025 were investigated, investigating the relationship between HIV exposure and neurodevelopmental disorders. The PECO framework

Immune cells harbor their own microbiome-derived metabolome: a new layer of immunometabolic regulation.

Current models of microbiome, immune crosstalk center on extracellular receptor-mediated signaling, yet a critical observation challenges this paradigm: intracellular concentrations of gut-derived bacterial metabolites (GDBMs) in CD4+ T cells do not correlate with paired plasma levels, and it is intracellular, not circulating, GDBM burden that associates with metabolic pathway disruption and immune senescence. Here we propose the concept of an intracellular microbiome metabolome: a pool of aromatic GDBMs actively accumulated through carrier-mediated transport, retained through transcriptional

Acceptability and Effectiveness of Point-of-Care HCV Testing in Low- and Middle-Income Countries: A Systematic Review.

Hepatitis C virus (HCV) diagnosis in low- and middle-income countries (LMICs) relies on costly and technically demanding centralized laboratory services. This review aims to synthesize evidence on Point-of-Care (POC) HCV testing in LMICs to inform scale-up for HCV elimination. We searched Medline, Embase, PsycINFO, and the Cochrane Library for studies on decentralized POC HCV testing in LMICs between 2010 and 2022. Two reviewers independently conducted eligibility assessment and full text review. Testing uptake and linkage data were extracted using Excel, then descriptively synthesized to iden
