Liver-Related Late Effects in Long-Term Survivors of High-Risk Neuroblastoma: Insights From a Comprehensive Prospective Follow-Up Study.

BACKGROUND: Therapeutic advances in pediatric oncology have significantly improved overall survival, with a higher incidence of long-term adverse effects and sequelae. Among pediatric cancer survivors, the highest incidence of hepatic lesions has been observed in those previously treated for high-risk neuroblastoma (HRNB). While hepatic nodules in these patients historically raised concern for tumor locations, recent attention has shifted toward non-tumoral, late-onset hepatic sequelae. METHODS: In this study, we retrospectively reviewed long-term liver outcomes from an institutional perspect

Steatosis grade and cardiometabolic burden as determinants of hepatocellular carcinoma risk after hepatitis C cure in patients with metabolic dysfunction-associated steatotic liver disease.

BACKGROUND: Coexistence of metabolic dysfunction-associated steatotic liver disease (MASLD) increases hepatocellular carcinoma (HCC) risk after HCV cure. OBJECTIVE: The specific impacts of steatosis grade and cardiometabolic burden on HCC risk among patients with MASLD remain unclear. DESIGN: We enrolled 700 patients who underwent biannual HCC surveillance after HCV cure. Steatosis was graded by vibration-controlled transient elastography using controlled attenuation parameter cut-offs of 248-267, 268-279 and ≥280 dB/m, corresponding to S1, S2 and S3, respectively. Cardiometabolic risk facto

Research progress of GPC3 and gastric cancer: Clinicopathologic characteristics and application prospects.

The identification of new biomarkers and druggable targets is critical for improving the management of gastric cancer. Glypican-3 (GPC3), a glycosylphosphatidylinositol-anchored heparan sulfate proteoglycan, has emerged as a molecule of substantial translational interest due to its well-documented roles in tumorigenesis and its growing validation as a therapeutic target. Current research has found that GPC3 is expressed in some gastric adenocarcinomas and serves as a highly sensitive biomarker for alpha-fetoprotein-producing gastric cancer (AFPGC). GPC3 positivity is significantly correlated w

A novel maternally inherited CDKN1C variant in a familial beckwith-wiedemann syndrome case: expanding the genotype-phenotype spectrum.

BACKGROUND: Cyclin-dependent kinase inhibitor 1 C (CDKN1C), encoding the p57KIP2 protein, serves as a critical negative regulator of cellular proliferation. Loss-of-function mutations in CDKN1C are responsible for 5–8% of sporadic and 40% of familial Beckwith-Wiedemann syndrome (BWS) cases, with 133 variants catalogued in the Human Gene Mutation Database (HGMD). We herein report a novel CDKN1C variant in a familial BWS case. A female neonate delivered preterm via cesarean section for severe preeclampsia manifested large for gestational age, macroglossia, omphalocele, and nevus flammeus. Immedi

ARF6 promotes the nuclear translocation of β-catenin to facilitate AFP deficiency-mediated metastasis in CTNNB1S33Y-mutant hepatocellular carcinoma.

The mortality rate associated with hepatocellular carcinoma (HCC) remains alarmingly high, positioning it as a primary lethal cancer worldwide, primarily driven by its high degree of malignancy, recurrence, and metastasis. The high heterogeneity of HCC necessitates the urgent identification of new therapeutic targets and molecular pathways. Our previous research observed that alpha-fetoprotein (AFP) deficiency led to pro-metastatic roles in HCC with specific genotypes. Here, we further investigated the underlying molecular mechanism. Through proteome microarray screening, we identified that AD

Alpha-Fetoprotein as a Biomarker in Pregnancy: From Genetic Disorders to Obstetric Complications.

Alpha-fetoprotein (AFP) is a glycoprotein primarily produced by the fetal liver and yolk sac during development. It is a multifaceted biomarker with significant applications in the prenatal screening of congenital abnormalities, cancer, and other disorders. The level of AFP in maternal blood may indicate several obstetric concerns and complications during pregnancy. Atypical AFP levels are commonly utilized as a biomarker for detecting fetal anomalies, placental complications, and other pregnancy-related issues. These findings raise concerns regarding the effectiveness of screening maternal se

Evaluating microRNA-720 and microRNA-1275 as Noninvasive Specific Test Compared With Maternal Serum Alpha-Fetoprotein and Three-Dimensional Ultrasound to Diagnose Fetal Neural Tube Defects.

BACKGROUND: Neural tube defects (NTDs) are common congenital anomalies of the central nervous system and are associated with substantial fetal and neonatal morbidity and mortality. Although maternal serum alpha-fetoprotein (MSAFP) and ultrasonography (US) are routinely used for prenatal screening, small or atypical lesions may be missed. Circulating microRNAs have emerged as promising noninvasive biomarkers in prenatal diagnosis. AIM: To calibrate the diagnostic utility of circulating microRNA-720 and microRNA-1275 for prenatal detection of fetal NTDs and to compare their performance with MSA

Integrated genomic and biochemical diagnosis of a novel homozygous start-loss variant in AKR1D1 associated with neonatal cholestasis.

INTRODUCTION: Congenital bile acid synthesis defects are rare autosomal recessive disorders that typically present in early infancy with cholestasis, progressive liver dysfunction, and, in severe cases, acute liver failure. These conditions may mimic other metabolic diseases detected in newborn screening, complicating early diagnosis. The AKR1D1 gene encodes Δ4-3-oxosteroid 5β-reductase, a key enzyme in primary bile acid synthesis, and pathogenic variants cause bile acid synthesis defect type 2 (OMIM #235555). CASE DESCRIPTION: We report a 3-month-old male infant with severe neonatal cholesta

Hepatocellular Carcinoma Arising from Ectopic Liver Tissue in the Left Triangular Ligament: A Case Report.

Accessory liver lobes represent an infrequent anatomical anomaly. The probability of hepatocellular carcinoma (HCC) developing in these accessory lobes remains exceptionally rare. Herein we report a case of a HCC arising from accessory liver lobe. A 63-year-old man came to our institution because of an abnormally elevated level of alpha-fetoprotein (AFP). There were no previous medical issues related to chronic liver disease or cirrhosis. Initially, MRI, PET-CT, and preventive interventions failed to detect tumor lesions. After conducting three follow-ups, it was concluded that the elliptical
