In spinal rats physostigmine failed to produce a pressor response even after treatment of the animals with hexamethonium, whereas noradrenaline, McN-A-343 (4-(m-chlorophenylcarbamoyloxy)-2-butynyltrimethylammonium chloride) and AHR602 (3-acetoxy-1-benzyl-1-methylpyrrolidinium bromide) all produced large pressor effects. In rats anaesthetized with urethane, hexamethonium completely abolished the pressor effect of dimethylphenylpiperazinium but only partially blocked the pressor response to physostigmine; pressor effects of noradrenaline, McN-A-343 and AHR602 were potentiated. Combined treatment

Treatment with phenoxybenzamine and dichloroisoprenaline prevented the rise of blood pressure, contraction of the nictitating membrane and increase in cardiac contractile force produced by intravenous injections of bretylium and guanethidine in anaesthetized or spinal cats. Treatment with cocaine, imipramine or reserpine reduced the sensitivity to bretylium and guanethidine of the spinal cat. In the spinal cat treated with reserpine, sensitivity to the drugs could be restored by an infusion of noradrenaline. Chlorpromazine also blocked the pressor and nictitating membrane responses to bretyliu

In the cat, the pressor actions of noradrenaline and of adrenaline were generally reduced and that of tyramine was increased during infusions either of noradrenaline or of adrenaline. The increase in the response to tyramine after an infusion of noradrenaline was prevented by cocaine, methyl phenidate and pipradrol. Pipradrol, unlike its isomer azacyclonol, increased the responses to catechol amines but reduced that to tyramine. Cocaine did not prevent the increase in the noradrenaline content of cat kidney and uterus after an infusion of noradrenaline. In the pithed rat, cocaine increased the

In the anaesthetized cat a single, large, intravenous dose of (-)-isoprenaline reversibly blocked the depressor properties of isoprenaline. Further large doses then caused monophasic rises in blood pressure, which were similar in rate of onset and duration to those produced by adrenaline and noradrenaline. This pressor effect was not due to an increase in cardiac output, but the main cause was vasoconstriction in skeletal muscle. The responses to isoprenaline, adrenaline and noradrenaline were compared, both before and after the blocking dose of isoprenaline. Isoprenaline prevented or antagoni

In anaesthetized cats 3-phenoxypropylguanidine caused a contracture of the nictitating membrane, a dilatation of the pupil and a fall followed by a rise in the arterial blood pressure. In spinal preparations of cats the initial fall in blood pressure was usually absent and the rise in blood pressure subsided to a steady level, which was about 10 mm Hg above the initial pressure. The pressor action and the contracture of the nictitating membrane were inhibited by phenoxybenzamine and by previous treatment with reserpine, but were not abolished by adrenalectomy and bretylium. 3-Phenoxypropylguan

PLATELETS IN NATIVE BLOOD ADHERE SPONTANEOUSLY TO GLASS INDEPENDENTLY OF TEMPERATURE: if adenosine diphosphate is added to the blood the adhesiveness of the platelets is increased and this effect is largely independent of temperature. The mono- and triphosphates decrease adhesiveness at 20 degrees C. and 37 degrees C. but have no effect at 0 degrees C.; cocaine inhibits adhesion at 37 degrees C. and at 0 degrees C. Aggregation and viscous metamorphosis of platelets in native plasma is induced at 37 degrees C. by adenosine diphosphate or by thrombin; these reactions do not occur at 0 degrees C.

Intravenous injection of phenoxybenzamine, choline 2,6-xylyl ether bromide (xylocholine, TM 10), piperoxane or dihydroergotamine increases the vasopressor activity of blood plasma, collected from cats under chloralose anaesthesia. The increased vasopressor activity that follows the administration of piperoxane is due to an increase of adrenaline and noradrenaline in the plasma. Cross-circulation experiments show that catechol amines are liberated from the spleen by piperoxane.

Several factors influencing the hypertensive effect of eserine in the rat were investigated. Pretreatment with reserpine regularly depressed or abolished the hypertensive response to eserine. The slow intravenous infusion of either noradrenaline, dihydroxyphenylalanine or 5-hydroxytryptamine only occasionally restored the hypertensive effect of eserine in reserpine-treated rats. Bretylium and choline 2,6-xylyl ether bromide significantly depressed or even abolished the hypertensive effect of eserine. The effect of bretylium was stronger than that of choline 2,6-xylyl ether bromide. Cocaine was

Tyramine is mainly a pressor agent in the rat under urethane, although the hypertensive effect of tyramine in some experiments is followed by a prolonged increase or depression of blood pressure. Bretylium, in doses up to 10 mg/kg, prolonged the response to tyramine, whereas larger doses depressed or blocked its effect. When the hypertensive effect of tyramine was blocked by bretylium, both noradrenaline and dihydroxyphenylalanine, when slowly infused, were found to restore it. The well-known block by cocaine of the hypertensive response to tyramine could also be reversed by intravenous infusi

Cocaine augments appreciably the effects of small doses of acetylcholine on the heart rate, blood pressure, and the nictitating membrane in intact, anesthetized animals. This phenomenon assigns cocaine, thus far only known as a potentiator of adrenergic stimulation, a more general role of potentiator of both adrenergic and cholinergic neurohumors.
