Potency-related effects of smoked cannabis on simulated driving performance: a randomized, controlled crossover trial.

Given the increasing availability of high-potency Δ9-tetrahydrocannabinol (Δ9-THC) products, understanding potency-related effects of Δ9-THC on driving performance is an important public safety issue. This randomized, double-blind, placebo-controlled, within-subjects trial examined the effects of smoked cannabis with varying Δ9-THC concentrations on simulated driving. Adults aged 19–45 who regularly used cannabis and held valid driver’s licenses completed simulated driving tasks after smoking placebo, or cannabis containing low (6.25%/47 mg Δ9-THC), medium (12.5%/94 mg Δ9-THC), or high (22%/16

Physical Performance Impairment with Cannabis Consumption in Adults Over 12 Hours.

Assessing cannabis-induced impairment in physical performance is critical for safety-sensitive occupations. This study evaluated the magnitude and duration of impairment following acute cannabis use. Twenty-eight adult users completed baseline and follow-up assessments at 1, 6, and 12 h after smoking a standardized cannabis cigarette. Measures included subjective intoxication, vital signs, psychomotor performance, muscle strength, balance, and endurance. Blood Δ9-tetrahydrocannabinol (THC) concentrations peaked at one hour and declined by six hours (p < 0.001), while Carboxy-THC showed minimal

Acute metabolic effects of cannabinoid receptor modulators during sequential hyperglycemic, euglycemic-hyperinsulinemic clamps in healthy individuals.

The endocannabinoid (eCB) system is involved in a vast array of physiological processes including regulating metabolism through insulin secretion and action. In this physiological study of 21 healthy men, we investigated the acute effects of a nonspecific cannabinoid receptor agonist nabilone (NAB 2 mg) and a specific cannabinoid receptor 1 (CB1R) antagonist CP-945,598 at two concentrations (low dose CP, LDCP 15 mg and high dose CP, HDCP 45 mg) on insulin secretion, insulin action, and glucose disposal-all compared with placebo (PL). One of the following compounds-NAB, LDCP, HDCP, or PL-was ad

Pharmacokinetic Differences Between Fast-Acting, Standard, and Placebo Cannabis Edibles.

INTRODUCTION: Edibles have become the second-most used cannabis product in legal U.S. states, wherein 64% of cannabis consumers reported using edibles within the past year. Among expansions to the legal cannabis industry are the newly marketed "fast-acting" edible compounds, which may address many of the issues associated with edible use related to overdose and dose management. The study hypotheses were that fast-acting edibles would reach peak concentration significantly faster than standard edibles and placebo edibles. MATERIALS AND METHODS: Twenty participants completed three arms within-s

Psilocybin restores behavioral and neuroplastic deficits induced by chronic stress in rats.

Psychedelics have emerged as a promising novel therapeutic approach for major depressive disorder (MDD). Altered activity and structural atrophy of the prefrontal cortex, hippocampus, and limbic structures are associated with depressive disorders. Psilocybin may reverse the loss of synaptic connections and restore the function of these brain regions. In this study, we investigated the effects of psilocybin on rat behavior, hippocampal neurogenesis, expression level of brain-derived neurotrophic factor (BDNF) and hypothalamic-pituitary-adrenal (HPA) axis activity. Psilocybin administered in two

Brain-body integromics of the ayahuasca experience.

Ayahuasca is a psychoactive brew containing N,N-dimethyltryptamine (DMT) and β-carboline alkaloids that induces marked alterations in perception, emotion and self-referential processing. However, the multiscale biological organization linking peripheral metabolism, brain network dynamics, neurochemistry, and subjective experience in humans remains poorly understood. Here, we applied an integrative, within-subject, multiblock partial least squares framework to model coordinated changes across four complementary biological and phenotypic layers: plasma psychoactive alkaloids, targeted metabolomi

Acute dose-dependent effects of 4-bromo-2,5-dimethoxyphenethylamine (2C-B) compared with 3,4-methylenedioxymethamphetamine (MDMA) and psilocybin in a double-blind, placebo-controlled study in healthy participants.

Based on its in vitro profile and preliminary evidence, 4-bromo-2,5-dimethoxyphenethylamine (2C-B) may have psychoactive properties that are similar to 3,4-methylenedioxymethamphetamine (MDMA) and psilocybin, which are investigated for the treatment of posttraumatic stress disorder and depressive disorders. We compared acute effects of 2C-B (10, 20, and 30 mg), 125 mg MDMA, and 25 mg psilocybin in 24 healthy participants (12 women, 12 men) using a double-blind, randomized, placebo-controlled, crossover design. Determination of THC and THC-COOH in Dried Capillary Blood Spots and Comparison to Venous Blood of Recreational Cannabis Consumers in a Pilot Study.

With increased sensitivity of instrumentation, more substances can be analysed using capillary dried blood spots (DBS), and therefore, minimal invasive sample collection can be performed. However, for cannabinoids such as (-)-trans-Δ9-tetrahydrocannabinol (THC), low extraction efficiencies have been reported in previous studies. In this study, a novel extraction method was developed comprising an extraction with dimethyl sulfoxide followed by methanolic extraction, yielding extraction efficiencies > 80% for both THC and its metabolite 11-nor-9-carboxy-Δ9-tetrahydrocannabinol (THC-COOH). Effect of cannabidiol, cannabinol and tetrahydrocannabivarin in managing inflammatory pain.

Current medications used to treat the inflammatory pain either have limited effectiveness or may be associated with serious side effects. Non-psychoactive phytocannabinoids may have a better safety profile and could be used as an alternative approach to treat this condition. The aim of this study was to test the potential of three phytocannabinoids, cannabinol (CBN), tetrahydrocannabivarin (THCV) and cannabidiol (CBD), in treating inflammatory pain. The inflammatory pain was triggered in male rats by a single intra-articular knee injection of the complete Freund's adjuvant (CFA). One week late

Differential alterations in peripheral tryptophan pathways in methamphetamine versus MDMA users are linked to their contrasting psychiatric symptoms.

Methamphetamine (METH, "Crystal Meth") and 3,4-methylenedioxymethamphetamine (MDMA, "Ecstasy") are two types of substituted amphetamines that share structural-chemical similarities but exhibit contrasting acute and chronic effects including addictive liability. Tryptophan (TRY) pathways are involved in pleiotropic physiological functions at the interface of brain-body connections. Preclinical evidence suggests that amphetamines may modulate these pathways and, thus, indirectly influence brain functions via persistent alterations of peripheral metabolites. However, little is known about alterat

Phase I Metabolism of Novel Phencyclidine Derivative 3-Cl-PCP: In Vitro Studies With Pooled Human Liver Microsomes and Investigation of a Post-Mortem Case.

Assessing fatalities linked to new psychoactive substances (NPS) often remains challenging. This study investigates a fatal intoxication involving the novel phencyclidine derivative 3-chloro-phencyclidine (3-Cl-PCP), analyzing its phase I metabolism aided by pooled human liver microsomes (pHLMs) and supported by in silico models. Postmortem samples (bile, urine, cardiac and femoral blood, and gastric contents) and three syringes were collected from a fatal intoxication case. Quantification was performed using liquid chromatography-tandem mass spectrometry (LC-MS/MS). In vitro metabolism studie

Effects of sub-chronic cannabis smoke exposure on inflammatory markers in serum and brain in younger and older mice.

Aging is associated with chronic low-grade inflammation, which is thought to contribute to both cognitive decline and various neurodegenerative diseases. Cannabinoids are reported to reduce levels of inflammatory markers; however, these effects have not been thoroughly assessed in older subjects. To address this gap, we evaluated the effects of sub-chronic cannabis smoke exposure on serum and brain inflammatory markers in younger and older mice. Younger (4 month old) and older (22 month old) C57Bl/6J mice were exposed to smoke from burning either cannabis (5.5-6.2% THC) or placebo (0% THC) cig

Global increases in brain glucose metabolism following acute N,N-dimethyltryptamine and harmine administration in healthy volunteers: A randomized [18F]FDG-PET study.

Classical psychedelics such as N,N-dimethyltryptamine (DMT) modulate consciousness via serotonergic receptor agonism, and are increasingly investigated for their psychotherapeutic potential. When combined with the monoamine oxidase A (MAO-A) inhibitor harmine-mimicking the pharmacological profile of ayahuasca-oral DMT induces a psychedelic experience lasting 4-5 h. While some neuroimaging studies have characterized effects of DMT on functional connectivity and electroencephalography its impact on cerebral energy metabolism remains largely unexplored. We assessed the cerebral metabolic rate for

Acute and post-acute neurobehavioral responses to lysergic acid diethylamide in healthy subjects: a randomized controlled study.

Preclinical studies suggest that lysergic acid diethylamide (LSD) may induce lasting changes in brain function and learning ability, but evidence in humans is uncertain. Motor learning, in particular, has clinical relevance but has not been investigated in human studies of psychedelics. Forty-five healthy subjects (24 women) participated in this randomized crossover trial comparing 100 µg LSD with a placebo. For up to one week after dosing, we investigated LSD's post-acute neurophysiological effects using auditory tetanization with electroencephalography (EEG), paired associative stimulation (

A six-hour time series of the acute effects of ingested cannabis intoxication on a battery of cognitive-motor tasks.

The psychoactive component of cannabis, Δ9-tetrahydrocannabinol (THC), is known to alter cognitive-motor function; however, despite the high prevalence of cannabis use globally, the time course of the effects of ingesting THC is unclear. To allow future studies to accurately time the assessment of the impacts of cannabis on behaviour, this study aimed to establish a detailed time series of subjective intoxication and cognitive-motor performance following THC ingestion. Fifteen infrequent cannabis users (8 females; 29.4 ± 8.1 y) completed a testing battery before (baseline) and at 11 timepoints

Three-year review of tetrahydrocannabinol and alcohol blood concentrations and effects in impaired driving investigations in Ontario, Canada.

Driving under the influence of cannabis is a significant public safety issue. Tetrahydrocannabinol (THC), the main psychoactive compound in cannabis, can impair driving ability and when used in combination with alcohol, the level of impairment can increase. Case studies documenting blood concentrations of THC and associated observations of impairment in individuals suspected of impaired driving continue to provide important contributions to the scientific literature. Retrospective analysis was performed using data from impaired driving investigations in which THC alone, and in combination with

Bedtime tetrahydrocannabinol ingestion reduces morning orthostatic cardiovascular reactivity in adults: An exploratory study.

Acute cannabis use and its primary psychoactive constituent tetrahydrocannabinol (THC) can induce postural dizziness, indicating increased risk of orthostatic instability. However, it is unknown if vulnerabilities persist the morning after bedtime THC consumption. This study investigated the influence of bedtime THC consumption on cardiovascular variables during an orthostatic challenge. Nine individuals with no cannabis use and eight individuals who regularly use cannabis participated in a tilt table test associated with a 3-day in-laboratory stay. Participants underwent an acclimatization da

Illicit drug N-ethylpentylone potentiates platelet activation through 5-hydroxytryptamine2A receptor-mediated mitogen-activated protein kinase signaling.

BACKGROUND: Several illicit drugs have been reported to be associated with ischemic stroke and myocardial infarction. However, their underlying mechanisms remain poorly investigated. OBJECTIVES: In this study, we focused on N-ethylpentylone (NEP), a synthetic cathinone with potent hallucinogenic properties, and explored its role in platelet activation. METHODS: Platelet function assays and flow cytometry were used to evaluate the effects of NEP on platelet aggregation, spreading, clot retraction, and integrin αIIbβ3 activation. 5-HT1A receptor blockade potentiates the subjective effects of DMT.

INTRODUCTION: Serotonergic psychedelics are being investigated in the treatment of various disorders and in the improvement of well-being. Evidence suggests that their subjective effects may play a role in long-term behavioral outcomes. The subjective effects are mediated by 5-HT2A receptor agonism, but the 5-HT1A receptor may also play a role in the subjective effects. This study elucidates the effects of 5-HT1A receptor blockade using pindolol pre-treatment on the subjective effects induced by N,N-dimethyltryptamine (DMT).
