Concentration-dependent effect of delta-9-tetrahydrocannabinol on epigenetic DNA modifiers in human peripheral blood mononuclear cells.

Cannabis is among the most used illicit substances in the world, and approximately 10% of regular cannabis users are estimated to be susceptible to developing cannabis use disorder (CUD). We examined the effect of different concentrations of delta-9-tetrahydrocannabinol (THC) on the epigenetic DNA modifiers DNA methyltransferases (DNMTs) and ten-eleven translocation enzymes (TETs); cannabinoid CB1 and CB2 receptors; and the cytokines IL-1β, IL-6, IL-10, and TNF-α. We used two in vitro study designs on human peripheral blood mononuclear cells (PBMCs) collected from healthy donors: (a) repeated

Molecular brain imaging of psychedelic action.

Molecular brain imaging by positron emission tomography (PET) and single photon emission computer-tomography (SPECT) entails the mapping of the cerebral distribution of radiopharmaceuticals that track physiological processes such as blood perfusion and glucose metabolism, or the abundance in brain of specific molecular targets such as neuroreceptors. PET and SPECT emerged as useful in vivo research technologies in the 1980s, finding early application in the study of psychostimulant drugs. The past decade has seen growing use of molecular imaging methods in the study of psychedelic action, alth

Differential metabolic pathways underlie THC- and CBD-mediated inhibition of B-cell activation in both young and aged mice.

OBJECTIVE: B lymphocytes play a crucial role in immunity but also contribute to the pathogenesis of various diseases. Cannabis plants produce numerous biologically active compounds, including cannabinoids. The two most studied phytocannabinoids are Δ9-tetrahydrocannabinol (THC) and cannabidiol (CBD). These cannabinoids exert diverse and potent biological effects primarily through the endocannabinoid system (ECS), which also plays a key role in mature B-cell function. Both the immune system and the ECS undergo age-related changes that lead to a clinically significant decline in function. METHO

Evaluating the impact of cannabis oil for autistic children with and without concomitant medications: Insights from an open-label study.

BACKGROUND: Although only two drugs are FDA approved for autism spectrum disorder (ASD), clinical practice treatment includes off-label use of medications to address the troubling symptoms of ASD. Several trials showed the beneficial effects of medical cannabis for alleviating symptoms of ASD. However, data are lacking regarding its safety and effectiveness as a single agent compared to add-on therapy. AIMS: To compare the safety and effectiveness of medical cannabis as a monotherapy and add-on therapy in autistic children. METHODS: An open-label trial recruiting autistic children was perfor

Population pharmacokinetic-pharmacodynamic modeling of co-administered N,N-dimethyltryptamine and harmine in healthy subjects.

N,N-dimethyltryptamine (DMT) is a psychedelic compound commonly co-administered with the monoamine oxidase inhibitor harmine in ayahuasca-inspired formulations. However, the impact of harmine on DMT pharmacokinetics (PK) and pharmacodynamics (PD) remains insufficiently characterized. In this single-blind, randomized, two-arm, factorial, dose-finding study, 16 healthy participants (9 males, 7 females) received six combinations of buccal DMT (0-120 mg) and harmine (0-180 mg) via a microcarrier-based transmucosal delivery system. Hexahydrocannabinol: pharmacokinetics, systemic toxicity, and acute behavioral effects in Wistar rats.

BACKGROUND: Hexahydrocannabinol (HHC) is a new psychoactive substance known for its mind-altering effects and temporary legal status. It is widely used in parts of the Europe and United Kingdom as a legal alternative to ∆9-tetrahydrocannabinol, yet little research has explored its effects and safety. This study examined how HHC is processed in the body, its toxicity, and its impact on behavior in male Wistar rats. METHODS: A 1:1 mixture of (9R)-HHC and (9S)-HHC was administered via intragastric gavage at doses of 1, 5, and 10 mg/kg. Behavioral effects were assessed using the open field test a

The effect of MDMA administration on oxytocin concentration levels: systematic review and a multilevel meta-analysis in humans.

3,4-Methylenedioxmethamphetamine (MDMA) has been shown to enhance social cognition, partly through its effects on oxytocin, a neuropeptide involved in social cognition. Despite growing evidence linking MDMA to increased oxytocin levels, no study has systematically examined this relationship across different doses, times, and participant characteristics. This systematic review and meta-analysis evaluates the effects of MDMA administration on peripheral oxytocin levels in humans. A systematic search identified ten studies, comprising 39 effect size estimates. Multilevel meta-analyses were conduc

Effects of Δ9-tetrahydrocannabinol (THC), cannabidiol (CBD), and THC:CBD mixtures on behavioral and physiological signs of morphine withdrawal in rhesus monkeys.

The mu-opioid receptor agonists methadone and buprenorphine are effective for treating opioid use disorder (OUD); however, both drugs are diverted and misused and withdrawal signs can emerge when treatment is tapered or discontinued. Mu-opioid and cannabinoid type 1 receptors are colocalized in several brain regions, and cannabinoids have been proposed as potential treatments for opioid withdrawal. This study tested Δ9-tetrahydrocannabinol (THC) and cannabidiol (CBD), alone and in mixtures, for treating the behavioral and physiological signs of morphine withdrawal in 3 male rhesus monkeys. The

Influence of Carrier Oil, Sex, and Age on Pharmacokinetic and Acute Behavioral Effects of Vaporized Cannabis Extract in Mice.

The legalization of cannabis in several states across the United States has increased the need to better understand its effects on the body, brain, and behavior, particularly in different populations. Previous rodent studies have revealed age and sex differences in response to injected Δ9-tetrahydrocannabinol (THC). However, the pharmacokinetic and pharmacodynamic properties of THC administered through more translationally relevant routes of administration are less well known. Here, we addressed this gap by investigating age and sex differences in pharmacokinetics and the acute behavioral effe

A comparison of the prevalence of cannabis and alcohol use among drivers and passengers in British Columbia and Ontario, Canada.

BACKGROUND: Similar to drink driving, the prevalence of driving under the influence of cannabis (DUIC) is expected to depend on the availability and cost of cannabis which would impact cannabis use in both drivers and passengers, and factors that specifically target cannabis use in drivers such as the deterrent effect of traffic laws and driver's opinion about the risks and acceptability of DUIC. To disentangle these effects, we aimed to compare the prevalence of alcohol and tetrahydrocannabinol (THC) detection 1) in drivers vs. passengers involved in motor vehicle accidents and 2) in drivers

Acute effects of MDMA, MDA, lysine-MDMA, and lysine-MDA in a randomized, double-blind, placebo-controlled, crossover trial in healthy participants.

3,4-Methylenedioxymethamphetamine (MDMA) is used recreationally, as a research tool, and in MDMA-assisted therapy in patients with posttraumatic stress disorder. 3,4-Methylenedioxyamphetamine (MDA) is a psychoactive metabolite of MDMA. Acute effects of MDMA and MDA have never been directly compared in humans. Lysine-conjugated amphetamines slowly release active amphetamine once absorbed, suggesting pharmaceutical strategies to enhance tolerability and reduce abuse potential. Therefore, lysine-MDMA (Lys-MDMA) and lysine-MDA (Lys-MDA) were developed as prodrugs of MDMA and MDA, respectively. We

Tetrahydrocannabivarin (THCV) Dose Dependently Blocks or Substitutes for Tetrahydrocannabinol (THC) in a Drug Discrimination Task in Rats.

Delta-9-Tetrahydrocannabivarin (THCV), a naturally occurring cannabinoid and structural analog of THC, exhibits a dual pharmacological profile as a CB1 receptor agonist/antagonist and a partial CB2 agonist. This study evaluated the effects of THCV in a THC discrimination model in rats. Male Sprague-Dawley rats (n = 16, 300-340 g, PND60) were trained under a fixed ratio 20 (FR20) schedule to discriminate THC (3 mg/kg) from vehicle. Substitution tests were conducted with THC (0.325-3 mg/kg), THCV (0.75-6 mg/kg), and THC-THCV combinations. THCV produced an inverted U-shaped substitution curve, si

Psychedelic 5-HT2A receptor agonism alters neurovascular coupling and differentially affects neuronal and hemodynamic measures of brain function.

Human neuroimaging studies report that psychedelics induce serotonin-2A receptor-dependent changes in functional brain reorganization, presumably reflecting neuromodulation. However, these studies often overlook the potent vasoactive effects of serotonin. Here we identified psilocybin-induced alterations in hemodynamic response functions during human functional magnetic resonance imaging, suggesting potential disruptions in neurovascular coupling. We then used wide-field optical imaging in awake Thy1-jRGECO1a mice to determine whether psychedelic-induced changes in hemodynamics arise from neur
