The acute cardiovascular response to dynamic exercise and recovery following cannabis use.

Despite clear benefits in the long term, exercise transiently increases cardiovascular risk during and immediately after exertion. Cannabis use has similarly been associated with cardiovascular events, but its effects on cardiovascular function and potential interaction with exercise remain poorly understood. We examined cardiovascular responses during and after exercise following cannabis use, to elucidate their combined effects. In a within-subject design, participants either inhaled cannabis with a high Δ-9-tetrahydrocannabinol (THC) content via smoking (S-THC) or vaporizing (V-THC), or vap

Pharmacodynamic effects and plasma pharmacokinetics of N, N-dimethyltryptamine after intranasal versus subcutaneous administration in male rats.

RATIONALE: There is growing interest in the therapeutic utility of psychedelic compounds that act as serotonin-2 A receptor (5-HT2A) agonists. N,N-dimethyltryptamine (DMT) is a 5-HT2A agonist that produces intense and short-lived psychedelic subjective effects, but the compound requires non-oral routes of administration that bypass gastrointestinal metabolism. OBJECTIVES: Intranasal (IN) delivery of DMT represents one potential non-oral route of administration, but the feasibility of using this route is not well studied. Here, we examined the pharmacodynamic effects and plasma pharmacokinetic

Cannabis use and cardiometabolic risk in schizophrenia.

PURPOSE: Metabolic syndrome (MetS) is common in schizophrenia and drives cardiovascular risk. While cannabis use and potency are increasing, the impact of cannabis on cardiometabolic health in schizophrenia remains unclear. This study assessed the association between objectively measured cannabis use and MetS prevalence in a large schizophrenia cohort. METHODS: We conducted a cross-sectional analysis of 988 participants with DSM-IV schizophrenia from the CATIE study. Cannabis use was measured via hair testing for tetrahydrocannabinol (THC), the gold standard for long-term use detection. MetS

Evaluation of THC-induced neurotoxicity via oxidative stress in undifferentiated SH-SY5Y cells.

The increasing global consumption of cannabis, particularly high-THC products, has raised public health concerns due to potential neurotoxic effects, although its association with oxidative stress remains a subject of debated. Some studies link THC-rich cannabis to increased oxidative damage, while others highlight antioxidant properties of cannabinoids. This study aimed to evaluate whether THC concentrations observed in real-world scenarios, specifically in the blood of drivers involved in traffic accidents, can induce neuronal damage through oxidative stress in vitro. Human undifferentiated

Health impacts of cannabis: focus on smoking vs. vaping effects on the respiratory and cardiovascular systems.

PURPOSE OF REVIEW: The legalization of recreational and medical cannabis has shifted consumption patterns, with vaping emerging as a common alternative to smoking. This review summarizes recent evidence comparing the respiratory and cardiovascular health effects of these two modes of administration. RECENT FINDINGS: Vaping reduces exposure to combustion-related toxicants and allows modulation of Δ 9 -tetrahydrocannabinol (THC) bioavailability compared to smoking cannabis. However, both modes of consumption produce comparable physiological effects, including acute increases in heart rate and b

Psilocybin induces sex- and context-specific recruitment of the stress axis.

Following decades of prohibition, psychedelic drugs have reemerged as promising therapeutics for stress-related conditions, including depression and post-traumatic stress disorder. Still, their impact on stress-related brain regions and the hypothalamic-pituitary-adrenal (HPA) axis remains unclear. This work explores the acute effects of psilocybin on the primary regulators of the HPA axis: corticotropin-releasing hormone neurons in the paraventricular nucleus of the hypothalamus (CRHPVN). Here, using blood plasma measurements and in vivo single-fiber photometry, we demonstrate that psilocybin

Subdiaphragmatic vagotomy reduces hypothalamic oxytocin expression and blood levels after oral MDMA administration in male rats.

3,4-Methylenedioxymethamphetamine (MDMA) is a widely recognized entactogen frequently used recreationally. It is known for its interaction with the serotonin and oxytocin systems, which underlie its entactogenic effects in humans. Recently, we demonstrated that the gut-brain axis, mediated by the subdiaphragmatic vagus nerve, contributes to MDMA-induced resilience enhancement in rodents. This study investigates whether subdiaphragmatic vagotomy (SDV) affects plasma oxytocin levels and the expression of oxytocin and c-Fos in the hypothalamus following a single oral dose of MDMA in rats. SDV sig

Early psilocybin intervention alleviates behavioral despair and cognitive impairment in stressed Wistar rats.

Chronic stress exerts profound effects on mental health, contributing to disorders such as depression, anxiety, and cognitive impairment. This study examines the potential of psilocybin to alleviate behavioral despair and cognitive deficits in a rodent model of chronic stress, focusing on the interplay between the Hypothalamic-Pituitary-Adrenal (HPA) axis and the Endocannabinoid System (ECS). Twenty-two male Wistar rats were divided into control and stress groups. Animals within the stress group were exposed to predator odor and chronic social instability to induce chronic stress, and were eit

Safety, tolerability and subjective effects of vaporized N,N-Dimethyltryptamine: A randomized double-blind clinical trial.

Vaporized N,N-Dimethyltryptamine (DMT) has a short duration and simple application, prompting this technique as a new approach for psychedelics' clinical use. Building on our initial dose-ascending studies and addressing potential confounding effects of dosing order, this study is the first to evaluate inhaled DMT in a randomized, placebo-controlled, double-blind design. Given DMT's therapeutic potential, we aimed to assess its safety, tolerability, and subjective effects compared to an active placebo. Twenty-five healthy participants completed two treatment sessions, receiving DMT (60 mg) or

The effect of methamphetamine and 3,4-methylenedioxymethamphetamine on peripheral endocannabinoid concentrations: a study in healthy adults.

RATIONALE: Stimulant drugs such as methamphetamine (MA) and 3,4-methylenedioxymethamphetamine (MDMA) can impact neurobiological systems implicated in stress, reward processing, and drug use. Although recent preclinical evidence implicates the endocannabinoid (eCB) system in these processes, little is known about the acute effects of stimulants on eCB levels in humans. OBJECTIVES: The aim of the present study was to investigate the effects of acute administration of the prototypical psychostimulant MA and the psychostimulant-empathogen MDMA on circulating eCB levels in healthy adults. Psilocybin in the treatment of eating disorders: a systematic review of the literature and registered clinical trials.

BACKGROUND: Fluoxetine remains the only pharmacological treatment approved for Bulimia Nervosa, and no other drugs have been approved for eating disorders (EDs). The rationale for exploring psilocybin as a treatment for EDs is compelling, both from biological and psychological perspectives. Moreover, its safety profile in healthy individuals appears favorable. This systematic review aims to examine original research articles and registered clinical trials to assess the current psilocybin's therapeutic potential in EDs. METHODS: Systematic review following the indications of the Preferred Repo
