Valproate-induced hormonal and histological alterations in PTZ-kindled female rats with a focus on 5HT1A receptors.

Valproic acid (VPA), a widely used antiseizure medication, has been associated with significant reproductive and hormonal side effects that may lead to infertility in both sexes. Although VPA increases serotonin availability, the contribution of specific serotonergic receptors to these alterations remains unclear. This study evaluated the involvement of 5-HT1A receptors in VPA-induced hormonal and ovarian changes in pentylenetetrazol (PTZ)-kindled female rats. Fifty adult female Wistar rats were assigned to five groups (n = 10): Control (saline), PTZ + saline, PTZ + VPA, PTZ + NAD-299 (5-HT1A

Clozapine and Mirabegron, a case report of drug-drug interaction with severe consequences in a geriatric patient.

We report a rare but clinically significant drug-drug interaction between clozapine and mirabegron in a 63-year-old man with schizophrenia. The patient presented with repeated falls, drowsiness and balance disorders following initiation of mirabegron for overactive bladder. Clozapine plasma levels progressively exceeded the therapeutic range. Dose reduction of clozapine restored concentrations to target levels and improved mobility and alertness, with only one fall reported after adjustment. This case highlights a probable pharmacokinetic interaction through mirabegron's moderate inhibition of

Toward needle-free lamotrigine monitoring: Proof-of-concept for saliva and dried saliva spot analysis by LC-MS/MS - a short communication.

Recent advancements in micro-sampling methods provide non-invasive options for collecting biological samples used in therapeutic drug monitoring (TDM). Lately, alternative matrices such as capillary blood and saliva/oral fluid in the form of dried saliva spots, have attracted considerable interest. These matrices represent a promising approach for TDM, as they could improve accessibility while preserving analytical precision. The study developed and validated an LC-MS method for lamotrigine quantitation in saliva and Dried Saliva Spots (DSS). Samples were prepared via protein precipitation (me

Clinical Impact of Adjusted Valproic Acid Level in Patients with Hypoalbuminemia: A Single-Center Cohort Study.

Valproic acid (VPA) is highly protein-bound, thereby impacting its free fraction and clearance in hypoalbuminemia. There is limited data on VPA use in such patients. Thus, this study evaluates the impact of adjusted VPA concentration (aVPAc) in predicting effectiveness and adverse effects compared to total VPA (tVPA) levels in hypoalbuminemic patients. A retrospective cohort study involved adult patients with seizures or epilepsy between January 1, 2016, and December 31, 2022. The levels of tVPA and aVPA (adjusted for albumin) were compared using receiver operating characteristic curves, and A

Three cases of drug-related death during treatment for stimulant-induced psychosis.

Long-term use of stimulant drugs can cause schizophrenia-like psychotic symptoms including hallucinations and delusions in the syndrome known as stimulant-induced psychosis. Therefore, treatment for stimulant-induced psychosis follows the approach for schizophrenia and primarily involves the use of antipsychotic drugs that antagonize dopamine release. However, multiple studies suggest that pharmacotherapy and psychosocial therapy for stimulant-induced psychosis have limited therapeutic efficacy, leading to poor long-term prognosis characterized by recurrence and chronicity of psychiatric sympt

Antipsychotic plasma concentration as predictor of movement disorders and cardiometabolic side-effects: A comparison with prescription dose.

The clinical evidence for antipsychotic (AP) therapeutic drug monitoring (TDM) in evaluating AP-related movement disorders and cardiometabolic side-effects remains inconsistent. This study evaluates how AP plasma concentrations associate with movement disorders and cardiometabolic side-effects over time, and compares its predictive value to prescription dose in first-episode psychosis (FEP) patients. We included 200 remitted FEP patients from the HAMLETT trial. AP plasma concentrations were standardized using robust z-scores to accommodate different AP types. The St. Hans Rating Scale and Barn

XPO6 mRNA as a Potential Biomarker for Major Depressive Disorder: Associations with Disease State and Treatment Response.

INTRODUCTION: Exportin-6 (XPO6) regulates nuclear export of actin and related proteins, processes essential for cytoskeletal integrity and synaptic function. Given the importance of these mechanisms in mood regulation, XPO6 dysregulation may contribute to the pathophysiology of major depressive disorder (MDD), yet its role remains largely unknown. METHOD: XPO6 mRNA was measured in the peripheral blood of participants with MDD in an acute depressive episode (acute MDD group), MDD in remission, and healthy controls (screened using Chinese health questionnaire-12 and semi-structured interviews t

Translational assessment of butyrylcholinesterase activity as a diagnostic biomarker for depression using a chemiluminescent probe.

Depressive episode is a globally prevalent psychiatric condition with a complex, multifactorial etiology involving genetic and environmental factors and despite advances in biomarker research, clinical applications remain limited due to methodological inconsistencies and population variability. Butyrylcholinesterase (BChE), an enzyme involved in cholinergic transmission, has been less studied in the context of depression compared to acetylcholinesterase. Utilizing a BChE-responsive chemiluminescent probe (BCC), BChE activity was measured in a translational approach: human serum, rat plasma, an

Modulation of the Kynurenine Pathway in Obese Mexican Navy Women Following a Structured Weight Loss Program: A Pre-Post-Intervention Study.

Background: Obesity is characterized by chronic low-grade inflammation and metabolic disturbances, including an altered tryptophan (Trp) catabolism through the kynurenine pathway (KP). Since the KP is closely linked to immune activity, energy metabolism, and hepatic function, modulating its flux through lifestyle interventions has gained interest as a potential therapeutic strategy. Objective: This exploratory study aimed to investigate the impact of a structured 12-week weight loss program (WLP) on serum KP metabolites in a sample of Mexican women with obesity. Methods: This study involved a

Prenatal Δ9-tetrahydrocannabinol exposure rewires glucocorticoid-hippocampal crosstalk and impairs memory under stress in adolescent rat offspring.

BACKGROUND: Stress is a major determinant of psychopathology, with coping and cognitive impairment arising from abnormal glucocorticoid activity and brain-derived neurotrophic factor (BDNF)-dependent neuroplasticity in the hippocampus. The endocannabinoid system modulates stress and hippocampal functioning since early development, thus its early-life disruption may program vulnerability to stress-related cognitive deficits. METHODS: We investigated whether prenatal exposure to tetrahydrocannabinol (pTHC; 2 mg/kg, gestational days 5-20) alters spatial learning and memory in adolescent offsprin
