Functional characterization of a novel ZIP8 variant causing impaired manganese homeostasis and congenital disorders of glycosylation.

ZIP8, encoded by SLC39A8, mediates cellular uptake of divalent metal ions, including manganese (Mn). Mutations in SLC39A8 cause a congenital disorder of glycosylation (CDG) associated with Mn deficiency (SLC39A8-CDG). Here, we report a novel case of SLC39A8-CDG harboring a previously unreported mutation (p.F206I) in the transmembrane domain 3 (TMD3). The patient showed markedly low serum Mn levels and abnormal glycosylation. To evaluate the functional consequences of this variant, we generated cells expressing hZIP8-F206I. Mn, cadmium, and zinc uptake levels in cells expressing hZIP8-F206I wer

Defining Reference Intervals for Complete Blood Count and Micronutrient Parameters in Urban Bangladeshi Population.

With a lack of population-specific reference intervals (RIs) in Bangladesh, this study aimed to determine RIs for complete blood count (CBC) and specific micronutrients, vitamin D (VitD) and zinc, and to assess the possible impact of prior history of SARS-CoV-2 infection on these parameters. Healthy participants (n = 1724) of both sexes aged ≥ 10 years from slum and non-slum areas of Dhaka city were sampled systematically. A total of 26 CBC parameters were determined using an automated hematology analyzer, whereas the micronutrients VitD and zinc were measured using automated immunoassay analy

Single-atom nanozyme-based wireless microfluidic sensing platform for noninvasive hyperuricemia diagnosis.

Hyperuricemia is characterized by abnormally elevated serum uric acid (UA) levels. Persistent hyperuricemia not only triggers gout but also causes damage to the kidneys and cardiovascular system. Therefore, utilizing non-invasive diagnostic methods for early and continuous monitoring is crucial for preventing disease progression. Herein, we reported a portable wireless electrochemical sensing platform constructed from SA-Pt/rGO/MOF catalysts prepared via photochemical deposition of platinum single atoms (Pt SAs) on reduced graphene oxide/metal-organic framework (rGO/MOF) composites. The design

Cadmium exposure as a contributing factor to hepato-renal injury in apparently healthy active cigarette smokers in buea-cameroon.

Cigarette smoke contains numerous toxicants, including cadmium, polycyclic aromatic hydrocarbons (PAHs), tobacco-specific nitrosamines (TSNAs), and volatile organic compounds, all of which may contribute to hepato-renal injury. This study, conducted in Buea (Southwest Region, Cameroon), aimed to assess cadmium inhalation exposure from cigarette smoking as a contributing factor to altered liver and kidney function biomarkers in apparently healthy smokers, while acknowledging that combined exposure to multiple co-toxicants may potentiate organ dysfunction. A survey was used to recruit smokers an

Sperm-Associated Mercury Species Levels Determine Its Impairment: Evidence from Paired Analyses of Whole Semen and Seminal Plasma.

There have been conflicting results regarding the relationship between mercury (Hg) and semen quality among previous studies, partly because most studies rely on indirect reproductive biomarkers (e.g., whole blood, hair) and no study tested Hg species to understand their specific associations with semen quality. In this study, we collected paired whole semen and seminal plasma samples from 250 male participants aged 26-63 years old in Hong Kong. Total mercury (THg) and methylmercury (MeHg) levels were quantified in both matrices, and inorganic mercury (iHg) level was calculated based on the di

Early-life low-dose lead exposure impairs synaptic development via epigenetic repression of the PI3K/AKT/mTOR signaling pathway.

BACKGROUND: High-dose lead (Pb) exposure is known to trigger neuroinflammation and glial activation; however, underlying the effects of chronic, low-level Pb exposure affects brain development remain poorly understood. METHODS: In this study, we investigated whether exposure to a sub-threshold concentration of Pb (50 ppm in drinking water), resulting in blood Pb levels (3.6 ± 1.6 ng/dL) well below current international safety standards, can impair neurodevelopment in mice using an integrative multi-omic and morphological approach. RESULTS: Chronic early-life exposure led to significant behav

Ginsenoside Rh1 Modulates GABAergic Inhibitory Homeostasis and Mitochondrial Quality Control During Lead (Pb)-Associated Neuronal Dysfunction.

Disruption of neuronal excitation-inhibition balance and mitochondrial quality control may contribute importantly to lead (Pb)-induced neurotoxicity, but nutritional modulators targeting these processes remain poorly characterized. This study investigated the protective effects and underlying mechanisms of ginsenoside Rh1, a ginseng-derived bioactive compound, in Pb-exposed mice and Pb-treated HT22 hippocampal cells. Chronic Pb exposure caused spatial recognition deficits, reduced exploratory activity, anxiety-like behaviors, and marked neuronal injury, accompanied by Pb accumulation in blood

Multi-level evidence for Pb-Cd co-exposure nephrotoxicity: An integrated analysis of NHANES, outcome-oriented network toxicology, and cell-based validation.

BACKGROUND: Lead (Pb) and cadmium (Cd) remain widespread environmental toxicants, and the kidney is a major target organ. However, reliance on a single biomarker matrix or renal endpoint may incompletely characterize Pb-Cd-associated renal abnormalities. Whether blood- and urine-based Pb/Cd biomarkers exhibit differential association profiles across renal outcomes, and whether outcome-oriented networks converge on shared candidate pathways, remains unclear. METHODS: We analyzed NHANES 2011-2018 data from 5049 adults. Blood and urinary Pb and Cd biomarkers were evaluated in relation to estimat

Co-exposure to arsenic and nickel induces oxidative stress and mineral imbalance, impairing male reproductive parameters in Wistar rats.

Simultaneous exposure to multiple toxic metals is a common scenario in environmental contamination. Among these metals, arsenic and nickel are widely distributed pollutants with well-established toxic effects. However, their combined impact on male reproductive physiology and micromineral homeostasis remains poorly understood. This study evaluated the effects of subchronic co-exposure to arsenic (1 mg L-1) and nickel (7 mg L-1) in drinking water for 70 days on the hypothalamus, testis, and epididymis of adult Wistar rats. Exposed rats showed arsenic and nickel retention in all analyzed tissues
