Novel insights on carbon source influence on azomycin production by Streptomyces eurocidicus.

Azomycin (2-nitroimidazole), a compound synthesized by Streptomyces eurocidicus, is medically significant as a build block for the drug applied in treatment for drug-resistant tuberculosis and Chagas disease, as well as tumor hypoxia imaging. Despite its clinical value, the optimized bioprocess parameters for its production remain largely unexplored. This study evaluated the influence of various carbon and nitrogen sources (specifically glucose, starch, soybean oil, and casamino acids) on azomycin yields during submerged cultivation. Batch fermentations revealed that azomycin biosynthesis is p

Assessing diagnostic accuracy of onchocerciasis rapid diagnostic tests using Bayesian latent class models.

BACKGROUND: Onchocerciasis elimination programs increasingly rely on rapid diagnostic tests (RDTs) that detect antibodies to the Ov16 antigen, yet the performance of currently available rapid tests remains uncertain. The commercially available SD BIOLINE Ov16 RDT has shown inconsistent sensitivity when used on whole blood and does not consistently meet international performance thresholds for mapping or decisions to stop mass drug administration (MDA). Two novel onchocerciasis RDTs have been developed, DDTD RDT and GADx RDT, but their diagnostic accuracy has not been fully evaluated using meth

Frequency of Mutations Within P-Glycoprotein and Beta-Tubulin Associated With Ivermectin Treatment in Onchocerca volvulus Adult Female Worm Populations in Some Onchocerciasis Endemic Communities in Ghana.

BACKGROUND: Ivermectin is the drug of choice for the control of onchocerciasis because of its potent embryostatic and microfilaricidal effect. However, with long periods of mass drug administration, the effect of ivermectin on the Onchocerca volvulus worms is waning. These suboptimal responses have been linked to genes such as p-glycoprotein (P-gp) and beta-tubulin (B-tub). The purpose of this study was to define the proportion of adult female O. volvulus from some endemic communities in Ghana that have mutations in these genes. METHODS: O. volvulus adult female worms were obtained from 33 pa

Comparison of 6-Week Doxycycline vs 6-Week Rifampicin or 3 Weeks of Doxycycline Plus Rifampicin in the Treatment of Onchocerciasis: A Randomized, Placebo-Controlled, Double-Blind, Phase 2 Trial.

BACKGROUND: Depletion of Wolbachia has proven to be a validated curative treatment for filariasis. This trial investigated alternatives to the current treatment regimen for onchocerciasis: 6 weeks of doxycycline (DOX) at 200 mg daily. METHODS: This randomized, placebo-controlled, double-blind, phase 2a trial with 5 treatment regimens was conducted in Ghana to determine (1) whether treatment times could be shortened as compared with 6 weeks of DOX at 200 mg daily by using a combination of DOX and rifampicin (RIF) for 3 weeks or (2) whether a dosage reduction to 6 weeks of DOX at 100 mg or a ch

A Preliminary Investigation on the Utility of Embedding Fluency-Building Strategies within the TAGteach Procedure.

UNLABELLED: TAGteach is a trademarked intervention package designed to promote skill acquisition by combining the use of an audible stimulus as immediate feedback with several other behavior analytic procedures. Although TAGteach effectively improves the accuracy of sports performance, previous researchers have noted that TAGteach does not reliably promote synchronized, coordinated (i.e., fluent) sport movements. To date, no study has evaluated the efficacy of embedding fluency-building strategies within the standard TAGteach procedure. We used a concurrent multiple baseline across skills desi

Integrating the crowdsourced image-based morbidity hotspot surveillance for neglected tropical diseases (CIMS-NTDs) into Nigeria's healthcare system: a mixed methods study.

BACKGROUND: Traditional surveillance systems for neglected tropical diseases (NTDs) often suffer from underreporting, delays and limited reach, hindering effective disease control. This study describes the integration process of crowdsourced image-based morbidity hotspot surveillance for NTDs (CIMS-NTDs) into the government-led NTD program and assesses its operational performance and stakeholder experience. METHODS: A mixed methods study embedded in an implementation research framework was conducted in Ondo State, Nigeria. A quasi-experimental design was used to compare CIMS-NTDs with the exi

Pharmacokinetics, safety, and tolerability of an oxfendazole tablet formulation: a phase 1, randomized, placebo-controlled trial in healthy African volunteers.

Oxfendazole, a registered veterinary drug, has demonstrated broad-spectrum activity against multiple nematode species. First-in-human studies in healthy, predominantly Caucasian adults receiving a liquid formulation of oxfendazole demonstrated a favorable pharmacokinetic and safety profile, supporting its potential as a drug candidate for treating human helminth infections. To further advance its clinical development, we conducted a phase I bioavailability study using a field-applicable, immediate-release oxfendazole tablet. This trial investigated the pharmacokinetics, safety, and tolerabilit

Cognitive, Behavioral, and Emotional Manifestations in Nodding Syndrome: A Neuropsychological Narrative Review.

BACKGROUND: Nodding syndrome is a childhood-onset epileptic encephalopathy described in onchocerciasis-endemic regions of sub-Saharan Africa. Although characterized by recurrent atonic seizures with repetitive head-nodding movements, increasing evidence suggests that the condition extends beyond a purely motor epilepsy and involves progressive cognitive, behavioral, and emotional impairment. METHODS: This study was designed as a narrative review with a specific neuropsychological focus. A literature search was conducted in PubMed, Scopus, and Google Scholar for publications between 2013 and 2

Niger: the first human onchocerciasis-endemic country in Africa to achieve elimination of infection transmission.

BACKGROUND: Aerial larviciding of Simulium breeding sites in Niger was conducted from 1976 to 1989, as onchocerciasis was a serious public health problem. In 2002, onchocerciasis was declared no longer of public health importance. Surveys to determine whether transmission of infection was still occurring began in 2014, including onchocerciasis elimination mapping in 2019-2020. METHODS: The endemic area encompassed 1575 communities across the basins of the Niger, Tapoa, Goroubi, Diamangou, Sirba and Mekrou rivers. From 2014 to 2018, flies caught from 55 sites were processed to detect infection

Transforming infectious disease control through social innovation, community engagement and intersectional gender research.

Community engagement and approaches that aim to change unequal power relations are essential for inclusive, relevant and sustainable health interventions. A people-centred approach to research and programme implementation can amplify the voices of disadvantaged and often forgotten people and move towards genuine partnership with the communities, ensuring that research and action meaningfully reflect the priorities and realities of those most affected.

Onchocerciasis overview: pathogenesis, clinical spectrum, and therapeutic challenges of a neglected tropical disease.

Onchocerciasis is still a major health burden in sub-Saharan Africa and Yemen, where many cases occur as control efforts face persistent obstacles, while smaller, well-defined transmission foci remain in Brazil and Venezuela. Caused by the filarial nematode Onchocerca volvulus and transmitted by blackflies, the disease causes progressive disability, economic loss, and hampers community development. Despite decades of mass drug administration with ivermectin, the inability of the drug to kill adult worms, increasing concerns over resistance, and the lack of effective vaccines continue to hinder

A GIS and field-based assessment of the ecological consequences of illegal mining (galamsey) on blackfly breeding sites in Ghana: implications for the sustainable development goals.

BACKGROUND: The Ofin River basin has historically supported Simulium blackfly breeding, vectors of Onchocerca, yet recent Programme reports have noted a sharp decline in monitored populations. With illegal artisanal mining (galamsey) expanding around the basin, this study assessed how associated ecological changes may influence habitat suitability for blackfly breeding across three riverine communities (Adwuman, Buabenso, and Kyekyewere). METHODS: Water discharge and quality were assessed through field measurements and laboratory analyses of pH, electrical conductivity, turbidity, colour, tot

Autophagy is essential for anti-Wolbachia drug efficacy in Brugia malayi and insect cells.

INTRODUCTION: Onchocerciasis and lymphatic filariasis (LF) cause a significant global public health burden with more than 900 million individuals at risk and over 60 million people living with symptomatic manifestations caused by filarial diseases. Due to the importance of Wolbachia for the survival of adult filarial nematodes, anti-Wolbachia therapy has been validated as a safe macrofilaricidal treatment for LF and onchocerciasis. The A-WOL consortium was established with the goals of defining registered anti-Wolbachia antibiotics, as well as developing new drugs effective in a regimen of 7 d
