BACKGROUND: After the closure of the African Programme for Onchocerciasis Control (APOC) in 2015, the Ministry of Public Health of Cameroon has continued implementing annual community-directed treatment with ivermectin (CDTI) in endemic areas. The World Health Organization has proposed that 12 countries be verified for elimination (interruption) of transmission by 2030. Using Rapid Epidemiological Mapping of Onchocerciasis, a baseline geostatistical map of nodule (onchocercoma) prevalence had been generated for APOC countries, indicating high initial endemicity in most regions of Cameroon. BACKGROUND: Historically, onchocerciasis has been recognised for its dermatological and ophthalmological manifestations, such as blindness. However, growing epidemiological evidence indicates that onchocerciasis is also associated with neurological complications, particularly onchocerciasis-associated epilepsy (OAE). These complications are not currently reflected in disease burden estimates and associated disability-adjusted life years (DALYs) for onchocerciasis.
MAIN TEXT: The most recent global burden of disease estimates for onchocerciasis in 2019 reported 1.23 million DALYs without accou

Onchocerca volvulus is the agent of onchocerciasis (river blindness) and targeted by WHO for elimination though mass drug administration with ivermectin. A small percentage of adult female worms develop pleomorphic neoplasms (PN) which occur more frequently after ivermectin treatment. Worms with PN have a lower life expectancy and improved understanding of proteins expressed in PN and their impact on different tissues could help elucidate the mechanisms of macrofilaricidal activity of ivermectin. Within paraffin embedded nodules removed after ivermectin treatment, we detected 24 (5.6%) O. volv

BACKGROUND: Onchocerciasis remains an important public health problem targeted for elimination in Tanzania. Ulanga District was hyperendemic for onchocerciasis before the intervention, underscoring the need for particularly high coverage in MDA efforts. The district has been implementing MDA through Community Directed Treatment with Ivermectin (CDTI) strategy since 1998. However, there is preliminary evidence of persistent onchocerciasis transmission, which likely sustains the observed high prevalence of Onchocerciasis in both human and vector species. This could be linked to poor treatment co

The World Health Organization (WHO) has proposed elimination of onchocerciasis transmission (EOT) in a third of endemic countries by 2030. This requires country-wide verification of EOT. Prior to the shift from morbidity control to EOT, interventions in Africa were mostly targeted at moderate- to high-transmission settings, where morbidity was most severe. Consequently, there remain numerous low transmission (hypoendemic) settings which have hitherto not received mass drug administration (MDA) with ivermectin. The WHO has prioritised the delineation of hypoendemic settings to ascertain treatme

In onchocerciasis-endemic communities, black fly vectors of Onchocerca volvulus cause annoyance. No vector control was performed in Cameroon to complement ivermectin treatment despite high fly densities in the savannah and forest areas. This study assessed the knowledge and practices of four onchocerciasis-endemic communities in Sudan and the Guinea savannah in Cameroon. We surveyed four communities using a structured questionnaire. We interviewed 452 individuals (Mawong: 136, Befang: 160, Soramboum: 88, and Galim: 68) aged 14-50 years. Most respondents (90%) knew about black flies being a nui

This study aimed to detect and identify emerging pathogenic yeasts in surface waters from two Lagoon Systems in Rio de Janeiro, Brazil, by polyphasic taxonomy. The monitoring of potentially pathogenic fungi in aquatic environments, especially in regions impacted by human actions, is highlighted in this study as one of the axes of the One Health approach. Water samples were collected, and after isolation, fungi were identified by polyphasic taxonomy, including MALDI-TOF-MS, which is also used for bacteria identification. Our results describe the physicochemical parameters of the surface water o

Ivermectin (IVM), an antiparasitic drug approved by the Food and Drug Administration (FDA), is widely used to treat several neglected tropical diseases, including onchocerciasis, helminthiases, and scabies. Additionally, IVM has shown potential as a potent inhibitor of certain RNA viruses, such as SARS-CoV-2. However, IVM is highly hydrophobic, essentially insoluble in water, which limits its bioavailability and therapeutic effectiveness. The use of liposomes as drug carriers offers several advantages, including enhanced solubility for lipophilic drugs, passive targeting of immune system cells

The aim of this study was to assess dosing errors when administering ivermectin (IVM) according to height, using a measuring cup with reference to the manufacturer's recommended dosage according to weight. For this purpose, a secondary analysis of the database of a community trial conducted in the health districts of Bougouni and Kolondièba was carried out. This made it possible to assess the proportions of participants who received underestimated doses, and to identify the factors associated with underestimation of IVM dose when using the toise. In all, 8.6% of participants (258/3011) receive

Cytoforms (cytospecies and cytotypes) of the Simulium damnosum complex are the major onchocerciasis vectors in Africa; but they differ in vectorial efficiency. Transmission elimination of onchocerciasis requires understanding the vector dynamics and delineation of transmission zones, for monitoring and evaluation, and for stop-treatment decisions. This study sought to understand the vector population dynamics and to delineate Operational Transmission Zones in the 3 administrative regions (Far North, North, and Adamawa) of northern Cameroon. Using precontrol and relief maps, larvae were sampled

Current guidelines for onchocerciasis elimination rely heavily upon assessment of the presence of Onchocerca volvulus in the vector Simulium damnosum (sensu lato). This entomological study was conducted over four years in several regions of Burkina Faso to determine the progress made towards interrupting onchocerciasis transmission. Larvae and adult blackflies were collected in eight river basins (Comoé, Léraba, Dienkoa, Mouhoun, Bougouriba, Bambassou, Nakambé, Nazinon and Sissili). Larvae were analyzed by cytotaxonomy, and the adult blackflies analyzed for the presence of infective larvae of

Diptera plays a pivotal role in forensic investigations, particularly in estimating the post-mortem interval. This can be achieved by analyzing the succession patterns of species on decomposing bodies, which requires detailed knowledge of the local fauna. In rural areas of the Rio de Janeiro state, Brazil, forensic entomological succession remains understudied. This study aimed to evaluate the ecological succession of adult and immature dipterans on decomposing pig carcasses in rural areas of Seropédica municipality during dry and rainy seasons, through the threshold indicator taxa analysis (T

Onchocerciasis (river blindness), a neglected tropical disease caused by the filarial nematode Onchocerca volvulus, impacts millions of people in sub-Saharan Africa. The WHO coordinates global efforts to eliminate onchocerciasis and has prioritized development of improved diagnostic tests to aid these efforts. To find new microfilarial-associated diagnostic targets to help identify active infections, we used laser capture microdissection to isolate embryonic stages from histologic sections of O. volvulus worms in subcutaneous nodules excised from onchocerciasis patients. INTRODUCTION: A 2018 study found high epilepsy prevalence and mortality in the Maridi onchocerciasis focus (South Sudan). From 2019 onward, onchocerciasis elimination measures were strengthened in Maridi, and in 2020, an epilepsy clinic was established at Maridi County Hospital with free provision of anti-seizure medication. We investigated the changes in epilepsy-related mortality following these interventions.
METHODS: Repeated house-to-house surveys (2018 and 2024) in three Maridi neighbourhoods (Kazana-1, Kazana-2, Hai-Gabat). The number of deaths that occurred during the two years preced

This work explores the potential of B12N12 and Al12N12 nanocages as carriers for 1-(phthalazin-1(2H)-one)[(pyridin-2-yl)ethylidene]hydrazone (APN). Density functional theory (DFT) calculations at M06/def2-SVP were conducted for energy minimization and at M06/def2-TZVP level for property calculations. Molecular electrostatic surface potential (ESP) analysis of APN identified two potential adsorption sites: the pyridine nitrogen (confA) and the azomethine nitrogen (confB). Thermochemical analysis indicates that the APN-nanocage complexes are energetically favorable, spontaneous and exothermic wi
