Mechanisms and treatments of lymphedema.

Lymphedema refers to the interstitial edema caused by obstruction of lymph drainage. The clinical symptoms of lymphedema include local swelling and pain, thickened and rough skin, limited walking and repeated infection. Genetic mutation, fat deposition, aging and lymphatic filariasis may contribute to the pathogenesis of lymphedema. CD4+T cells also promote the development of lymphedema. Cell junctions between adjacent lymphatic endothelial cells (LECs) are crucial to maintain the integrity of lymphatic vessels, and damage of these junctions increases the vessel permeability and induces lymphe

Modeling and Predicting Counts and Environmental Correlates of Mosquito Distributions in Haiti Using Surveillance Data from the Ouest Department.

Vector-borne diseases present significant public health challenges in Haiti, particularly dengue fever and lymphatic filariasis (LF), with Haiti hosting 90% of LF at-risk individuals in the Americas. Understanding mosquito vector distribution is crucial, especially as ongoing sociopolitical instability in Haiti limits vector surveillance and control measures. This study analyzed the spatial distribution and abundance patterns of key mosquito vectors using ecological modeling approaches. Mosquito surveillance was conducted at 19 sites from the communes of Carrefour, Genier, and Léogâne in the O

DNA barcoding and mitochondrial genetic diversity of mosquito vectors from central India.

BACKGROUND OBJECTIVES: Accurate discrimination of mosquito vectors is essential for entomological surveillance in India, where multiple Anopheles, Aedes and Culex species transmit malaria, lymphatic filariasis and arboviral diseases. However, routine species identifications are challenged by cryptic and morphologically similar taxa. Furthermore, population genetic information of major mosquito vectors remains elusive. METHODS: A total of 265 adult mosquitoes representing 21 species from four mosquito genera and one non-mosquito dipteran genus ( Chironomus ) used as an outgroup, were collected

Filarial Worm Infestation of the Nasolacrimal Duct: A Rare Orbital Presentation.

Filarial infestation of the nasolacrimal duct is exceptionally rare and often misdiagnosed as benign lacrimal pathology. We present a 12-year-old boy with progressive medial canthal swelling initially suspected to be a dermoid cyst or inflammatory lesion. Magnetic resonance imaging revealed a well-defined enhancing lesion at the nasolacrimal duct with preseptal edema. Histopathology demonstrated a parasite with a thick eosinophilic cuticular wall and inflammatory infiltrate rich in eosinophils and foreign-body giant cells, consistent with filarial infestation, possibly Onchocerca species. Ecological Dynamics of Culex quinquefasciatus Larval Habitats in Purulia, India.

Mosquito-borne diseases remain a major global public health concern, with Culex quinquefasciatus acting as an important vector of lymphatic filariasis. This study investigated ecological and physico-chemical characteristics of larval breeding habitats across 14 villages in the Ajodhya Hill Forest, Purulia, India, between November 2023 and September 2024. Per-dip larval density (PDLD) was highest in polluted drains near human habitations (38.4 ± 1.5 larvae/dip), compared to ponds (25.53 ± 0.70) and roadside ditches (10.46 ± 0.35). Aerobic heterotrophic bacterial counts ranged from 4.13 × 105 to

Effects of Chlorella sorokiniana KU.B2 microalga on the development of Culex quinquefasciatus (Diptera: Culicidae) larvae.

Culex quinquefasciatus (Say) is a significant vector of lymphatic filariasis and arboviruses. Current control strategies rely heavily on chemical insecticides, highlighting the need for alternative and environmentally sustainable approaches. This study evaluated the effects of the microalga Chlorella sorokiniana KU.B2 on the development of Cx. quinquefasciatus larvae. Larvae were reared on microalgal-rabbit food diets: a control (100% rabbit food) and four treatments with increasing proportions of C. sorokiniana suspension (25 microalgae [Al], 50Al, 75Al, and 100Al). Additional rabbit food-onl

In silico identification and prioritization of potential drug targets in Wuchereria bancrofti: Profilin as a promising therapeutic candidate.

Wuchereria bancrofti, a major causative agent of lymphatic filariasis, poses a significant public health burden in tropical and subtropical countries. Despite the availability of antifilarial drugs used in mass drug administration programs, issues such as non-compliance, low coverage, emerging resistance, and incomplete parasite clearance underscore the need for novel therapeutic targets. In this study, we analyzed potential drug targets identified from our previous subtractive proteomic analysis of W. bancrofti. Computational analysis of these targets highlighted Profilin as a potential candi

Utility of antifilarial antibodies for post-validation surveillance of lymphatic filariasis in Tonga.

Post-validation surveillance (PVS) is essential to detect resurgence of lymphatic filariasis (LF) following elimination as a public health problem. Testing for antifilarial antibodies (Ab) alongside antigen (Ag) may increase sensitivity for identifying persistent transmission. We evaluated the utility of Ab-based serosurveillance for PVS in Tonga, which eliminated LF in 2017. A multi-pronged, risk-stratified cross-sectional survey incorporating community, school, an outpatient clinic, and volunteer recruitment was conducted in May-July 2024. Blood samples were tested for Ag using Alere™ Filari

A next-generation human lymphatic filariasis vaccine candidate, rBmHAXT, for clinical development.

This study was conducted to develop a robust, scalable manufacturing process for the candidate vaccine rBmHAXT for human lymphatic filariasis (LF). During scale-up production, rBmHAXT showed significant antigen aggregation leading to a loss of purified vaccine antigens. This project aims to create an improved formulation suitable for industrial-scale production while maintaining robust protection. We generated three variants: (1) ∆Cys, in which all cysteinyl residues were mutated to serinyl. (2) GS, which has a flexible glycine-serine linker inserted between each of the component antigens, and

Research Priority Setting Exercises in Neglected Tropical Diseases: A Systematic Mapping Review.

BACKGROUND: Neglected tropical diseases (NTDs) affect more than 1.7 billion people worldwide, primarily in low- and middle-income countries. Despite their high burden, NTDs remain underfunded and receive fragmented research attention. Research priority-setting provides a systematic approach to align limited resources with the most pressing needs. While several priority setting exercises have been conducted for NTDs, their findings have not been synthesized. This study aimed to systematically map the published research priority-setting exercises for NTDs, synthesize their methodologies, stakeho

The psychosocial impact of disability and stigma among persons affected by leprosy and lymphatic filariasis in India.

Neglected tropical diseases (NTDs), such as leprosy and lymphatic filariasis (LF), can cause disabilities despite available treatment. Both diseases disproportionately affect India's poorest populations, with high numbers in Jharkhand and Uttar Pradesh. Those affected experience severe psychosocial consequences, including participation restrictions, depression, and poor mental health. This study investigates the extent to which stigma affects social participation and mental wellbeing among persons with leprosy- and LF-related disabilities in two Indian states. Monitoring vector-borne diseases: A study in Italy and Greece.

Rapid urbanisation and environmental changes are reshaping mosquito population dynamics and the transmission of vector-borne pathogens. Culex pipiens and Aedes albopictus have successfully adapted to anthropogenic environments, becoming key vectors of emerging and re-emerging arboviruses and filarial parasites. This study conducted a comparative entomological and pathogen surveillance between 2022 and 2023 in central Italy and Crete to assess species composition, population dynamics, and circulation of arboviruses and filarial nematodes across different ecological contexts. In Italy, a total o

Co-designing the 'LEAD' strategy: A theory-informed stakeholder-led behavioural intervention to improve mass drug administration compliance for lymphatic filariasis elimination in an urban block of Odisha, India.

INTRODUCTION: Sustained compliance with Mass Drug Administration (MDA) is essential for interrupting transmission of Lymphatic filariasis (LF). However, behavioural and system-level barriers persist to hinder its uptake. We aimed to explore determinants of MDA participation using behavioural frameworks and to co-design a theory-informed, stakeholder-led intervention to enhance MDA compliance. METHODS: A mixed-methods formative study was conducted between January and June 2025 in an LF-endemic block of Odisha, India. The quantitative component comprised a community-based KAP survey(n = 400) as
