Computational Validation of Tecomella undulata Bark as a Source of Anti-Filarial Leads Targeting UDP-Galactopyranose Mutase.

Lymphatic filariasis remains a major neglected tropical disease and there is an urgent need for parasite-specific therapeutic targets. The enzyme UDP-galactopyranose mutase (UGM) is essential for galactofuranose biosynthesis in Brugia malayi and is absent in humans, which makes it a promising drug target. In this study, we investigated phytochemicals from the bark of Tecomella undulata to find potential drug candidates against B. malayi UGM using an in silico approach. The structure of the target protein was predicted using the Alphafold model, and the active site was predicted using the combi

Emergence of a novel zoonotic brugian filarial infection during post-validation surveillance for lymphatic filariasis in Sri Lanka.

BACKGROUND: Lymphatic filariasis (LF) affects approximately 120 million people globally and is caused by Wuchereria bancrofti, Brugia malayi and Brugia timori. Sri Lanka was historically endemic for both bancroftian and brugian filariasis, with B. malayi presumed eliminated by the late 1960s. The country achieved validation of LF elimination as a public health problem in 2016. However, sporadic Brugia infections have been detected in microfilaria surveys from the mid-2000s onwards; the sub-periodic periodicity of which is suggestive of a zoonotic origin and warrants further research. METHODS:

There Is No Association Between Loiasis and Malaria: Findings from a Secondary Analysis of a Cross-Sectional Survey in Rural Gabon.

Loiasis exists in regions where malaria is highly endemic, yet few studies have investigated their association as concomitant infectious diseases. Secondary data analysis from a cross-sectional survey conducted in Gabon (2015-2016) was performed to assess the association between malaria and loiasis. A total of 947 participants of all ages were enrolled in the original study. In crude analyses, malaria showed a seemingly protective association with loiasis, manifesting in an odds ratio (OR) of 0.67 (95% CI: 0.45 to 1.01; p = 0.0521). This borderline association disappeared completely after adju

Pharmacokinetics, safety, and tolerability of an oxfendazole tablet formulation: a phase 1, randomized, placebo-controlled trial in healthy African volunteers.

Oxfendazole, a registered veterinary drug, has demonstrated broad-spectrum activity against multiple nematode species. First-in-human studies in healthy, predominantly Caucasian adults receiving a liquid formulation of oxfendazole demonstrated a favorable pharmacokinetic and safety profile, supporting its potential as a drug candidate for treating human helminth infections. To further advance its clinical development, we conducted a phase I bioavailability study using a field-applicable, immediate-release oxfendazole tablet. This trial investigated the pharmacokinetics, safety, and tolerabilit

Autophagy is essential for anti-Wolbachia drug efficacy in Brugia malayi and insect cells.

INTRODUCTION: Onchocerciasis and lymphatic filariasis (LF) cause a significant global public health burden with more than 900 million individuals at risk and over 60 million people living with symptomatic manifestations caused by filarial diseases. Due to the importance of Wolbachia for the survival of adult filarial nematodes, anti-Wolbachia therapy has been validated as a safe macrofilaricidal treatment for LF and onchocerciasis. The A-WOL consortium was established with the goals of defining registered anti-Wolbachia antibiotics, as well as developing new drugs effective in a regimen of 7 d

Pulmonary Dirofilaria immitis Infection and Concurrent Pleural Lymphangiomatosis in a Young Woman from Costa Rica: A Diagnostic Challenge after 20 Years of Silence.

Human dirofilariasis is an underreported, vector-borne disease caused by the filarioid Dirofilaria immitis. A case of pulmonary dirofilariasis is reported in a woman from Costa Rica, manifesting respiratory distress, marked eosinophilia of 47%, and severe chylothorax. Pleural biopsies revealed a lymphoplasmacytic reaction with eosinophils, and bronchoalveolar fluid analysis revealed 28S and internal transcribed spacer 1 fragments that were 100% and 99.3% similar to D. immitis, respectively, and a coxA sequence identical to Wolbachia from D. immitis. Other infectious, autoimmune, and malignant

Surgical Intervention for Filariasis-Induced Lymphedema: A Systematic Review of Ablative and Physiological Approaches.

Advanced filariasis-induced lymphedema causes irreversible fibrosis, severe disability, recurrent infections, and major psychosocial burden, often with poor response to conservative therapy. Surgical management in this type of lymphedema is rarely addressed. We conducted a PRISMA 2020-guided systematic review evaluating surgical interventions versus conservative management or no intervention in adults with filarial lymphedema, involving various anatomical locations. Of 580 records identified, 29 studies were included for qualitative synthesis. The evidence base predominantly consisted of case

Molecular characterization of filarial infections and their associated risk factors in rural community of Southeastern Gabon.

BACKGROUND: Filarial infections continue to represent a considerable public health challenge in resource-limited communities in sub-Saharan Africa. In Gabon, the implementation of effective control strategies, such as mass drug administration (MDA), is hindered by a lack of contemporary data on the local distribution, molecular diversity, and associated risk factors of these infections, particularly in rural areas where Loa loa co-endemicity poses a risk of severe adverse events. This study aimed to molecularly characterize filarial species and determine the prevalence, associated risk factors

Ocular Loiasis in Morocco: A Case Report From a Non-endemic Area.

Loiasis is a parasitic infection caused by the filarial nematode Loa loa, endemic to Central and West Africa. Its occurrence in non-endemic regions such as Morocco is rare and may pose a diagnostic challenge. We report the case of a Moroccan patient presenting with an intermittent sensation of a mobile foreign body in the eye, associated with redness and photophobia. The patient reported no recent travel to endemic regions but had close contact with individuals originating from Cameroon, a loiasis-endemic area, raising questions regarding possible indirect or remote exposure. Following several

Sustained Drug Exposure Drives Efficacy in Mice: PK/PD Analysis of Corallopyronin A against Wolbachia using Physiologically Based Absorption Modeling.

Corallopyronin A (CorA) depletes essential Wolbachia endosymbionts of filarial nematodes and is therefore a promising candidate for treating the neglected tropical diseases lymphatic filariasis and onchocerciasis. To optimize anti-Wolbachia therapy, the pharmacokinetic/pharmacodynamic (PK/PD) relationship was investigated. Due to the intra-nematode and -cellular target, conventional minimal inhibitory concentration could not be determined. Instead, an iterative approach was used to define PD efficacy thresholds and compare them with IC₅₀/IC₉₀ values from infected cells. A physiologically based

Molecular and pathological characterization of a Dirofilaria sp. infection in a little egret (Egretta garzetta).

Dirofilaria spp. are mosquito-borne filarial nematodes that primarily infect mammals. Here, we describe a filarial nematode infection in a female little egret (Egretta garzetta) from Jeju, Korea. Necropsy revealed multiple slender nematodes (4-6 cm) in the subcutaneous tissues of both legs. Histopathological examination demonstrated granulomatous inflammation with intralesional parasites. Morphologically, the nematodes exhibited a thick multilayered cuticle with prominent longitudinal ridges, consistent with members of the genus Dirofilaria. Molecular analysis based on 18S rRNA and mitochondri
