Micropuncture techniques developed for the study of renal physiology have been adapted for investigation of the male reproductive tract. Ultramicro specimens were obtained in vivo from the tubules of the rat testis and epididymis. These samples were analyzed for sperm morphology and concentration. Patients scheduled for cystectomy for bladder carcinoma or exstrophy of the bladder were studied. The number of eggs per day passed in the urine preoperatively was compared with the number of worms recovered from the cystectomy specimen by dissection. An 18 year old phenotypic man is described with chromatin-positive Klinefelter's syndrome and undetectable peripheral human follicle stimulating hormone levels. The subject manifested chromosomal mosaicism consisting of three stem cell lines (45X; 46XY; and 47XXY). To date, we have studied 7 patients with X/XY mosaicism, one of whom showed an X/XY/XYY pattern. Four patients presented as newly born infants because of incomplete male development, ambiguity of external genitalia or Turner syndrome. Fourteen personal cases of agenesia of the vas deferens, bilateral in 12 and unilateral in 2, are described. In 13 of the cases the diagnosis was confirmed by surgical exploration. The role of the human testis in the production of 17beta-estradiol (E(2)) was investigated by determining the concentration of E(2) and testosterone in peripheral and spermatic vein plasma samples.
