Effects of NGR1 on the Protective Efficacy and Functional Vision Profile of Retinal Photodamage.

Prolonged exposure to high-intensity light can harm macula vision, particularly affecting the function of Müller cells and cone photoreceptors. Panax notoginseng saponins (PNS) have valuable pharmacological effects on cerebrovascular, neurological, and microcirculatory health. Notoginsenoside R1 (NGR1), derived from PNS, shows potential for treating vascular or ischemia-reperfusion-related retinal issues; however, its impact on cone cells and the functional vision profile is not well understood. This study aimed to explore the effect and efficacy of NGR1 on retinal photodamage in vivo in mice.

P2X2/3 receptors as a potential target for cancer pain treatment.

In the process of tumor growth and metastasis, which can invade nerve tissue, sensitize peripheral receptors, enhance sensory information transmission and induce pain. Bone cancer pain is the most common type of cancer pain, caused by primary bone tumors or other types of cancer metastasis, destruction and dissolution of bone tissue, leading to pain generation. The pathological mechanism of cancer pain is complex and its treatment is a thorny problem at present. P2X2/3 receptors are mainly expressed in nociceptive sensory neurons. ATP released by cells is involved in nociceptive and pain signa

Effects of FGF4/HIF-1α axis-mediated neuronal glycolysis on neuropathic pain.

The pathogenesis of neuropathic pain is complex, and effective treatment methods are lacking in clinical practice. Recent studies have shown that glucose metabolism reprogramming may be involved in the process of neuropathic pain, but its role and molecular regulatory mechanisms in neuropathic pain are still unclear. In this study, a rat model of chronic constriction injury of the sciatic nerve (CCI) was established, and the pain threshold was evaluated through behavioural analysis. Morphological staining, transmission electron microscopy, transcriptome sequencing, Western blotting, immunofluo

A study on the network pharmacology of HQGZWWT that prevents CINP by inhibiting ferroptosis through regulation of the p38/c-FOS/NF-kB pathway.

ETHNOPHARMACOLOGICAL RELEVANCE: Chemotherapy-induced neuropathic pain (CINP) is a frequent side effect of chemotherapy, and the mechanisms by which Huangqi Guizhi Wuwu Tang (HQGZWWT) prevents and treats CINP remain to be further elucidated. BACKGROUND: Oxaliplatin is a widely used chemotherapy drug with proven efficacy against multiple malignancies; however, its significant side effects severely limit patient dosing. One such effect is CINP, which involves multiple factors. Ferroptosis, a form of cell death, plays a crucial role in maintaining chronic neuropathic pain. HQGZWWT effectively pre

Behavioral Thermoceptive Responses and Morphologic Correlates in Mouse Models of CMT1A, HNPP, and Aging.

BACKGROUND: Thermoceptive dysfunction is a frequent but understudied feature of peripheral neuropathies and aging. Patients often report abnormal heat perception, yet the underlying sensory mechanisms remain unclear. This study evaluated thermoceptive behavior and corresponding structural changes in mouse models of inherited dysmyelinating neuropathy and natural aging to identify shared and divergent mechanisms. METHODS: Thermal preference was assessed using a user-independent gradient apparatus spanning physiological to noxious temperatures, with automated quantification of time in zone, dis

Inhibition of Pain Signal Conduction in Dorsal Root Ganglion Neurons by Optogenetic Technique Mediated with Upconversion Nanoparticles.

Traditional analgesic strategies not only have limited efficacy but also tend to cause side effects such as drug dependence and gastrointestinal tract damage. As a popular nonpharmacological neural modulation approach, optogenetic technology has seen increasingly in-depth research in the field of pain regulation. The dorsal root ganglion (DRG), a crucial relay station for pain information transmission, serves as a key target for pain regulation. Optogenetic technology holds broad prospects for pain regulation at the DRG. However, the application of traditional optogenetic technology in pain re

Endoplasmic reticulum stress contributes to high-dose radiation-induced chronic mechanical and thermal neuropathic pain.

IntroductionExposure to high doses of radiation can cause serious harm to human health and lead to long-term chronic pain. However, the mechanisms underlying pain caused by radiation overexposure remain largely unknown.MethodsIn this study, the double hind paws and tail of mice were exposed to 40Gy electron beams to establish a radiation-induced pain model.ResultsEven after the inflammation of the irradiated skin subsided, the mice continued to exhibit prolonged mechanical and thermal pain. The results indicated that the neuron injury marker activating transcription factor 3 (ATF3) was markedl

Reduced Mechanical Tactile Stimulation Under Space Microgravity Affects Synaptic Signaling and Contributes to Neuromuscular Aging in Caenorhabditis elegans.

Although space travel is becoming more accessible, our understanding of how the space environment and microgravity (μG) affect biology, physiology, and human health remains incomplete. This study examined the effects of μG on synaptic signaling and neuromuscular aging in Caenorhabditis elegans. The D01 cohort, consisting of L4 larvae to young adults raised in μG, exhibited a downregulation of genes linked to synaptic signaling, dopamine response, locomotion, cuticle development, and mitochondrial metabolism. This was accompanied by altered synapse dynamics, reduced motility, and shorter body l

Autonomic dysfunction in fibromyalgia syndrome: the role of small fiber damage.

OBJECTIVE: Cardiovascular autonomic symptoms, particularly orthostatic intolerance, are frequent in fibromyalgia syndrome (FMS) and markedly contribute to disability, yet their underlying mechanisms remain unclear. Small fiber pathology (SFP), detected in 30-50% of patients with FMS, may also involve postganglionic sympathetic fibers. This study examined the contribution of SFP to autonomic dysfunction and cardiovascular autonomic symptoms in FMS. METHODS: We prospectively enrolled 43 patients with FMS reporting orthostatic intolerance and 20 age- and sex-matched healthy controls. All partici

Concurrent Long Thoracic and Lateral Cutaneous Intercostal Nerve Entrapment After Chest Tube Thoracostomy Causing Scapular Winging and Chest Wall Pain: Diagnosis With Ultrasound and Treatment With Hydrodissection.

BACKGROUND: Chest tube thoracostomy may cause iatrogenic nerve injury, resulting in persistent pain and functional impairment. Long thoracic nerve (LTN) palsy after chest tube placement has been described, but concurrent involvement of the LTN and the lateral cutaneous branch of an intercostal nerve (LCIN) appears to be rarely reported. We describe a patient with persistent post-thoracostomy chest wall pain and scapular winging in whom musculoskeletal ultrasonography identified entrapment of both nerves. CASE PRESENTATION: A 43-year-old man presented with severe left shoulder and chest wall p

Feeding following cleft palate surgery: a prospective randomized clinical trial comparing suprazygomatic maxillary nerve block and greater palatine nerve block.

INTRODUCTION: In the context of cleft-lip and palate surgery, both suprazygomatic maxillary nerve blocks (SZMNB) and greater palatine nerve blocks (GPNB) have been shown to provide improved postoperative pain relief. In this context, both pain relief and early postoperative feeding are of great importance. However, these two techniques have so far not been properly compared with regard to these characteristics. METHODS: Children 1-10 years of age undergoing palate surgery were randomized to either SZMNB or GPNB. Primary aim was early (0-24 hour) postoperative feeding (%) in relation to full n

Heteronymous feedback from quadriceps onto soleus in stroke survivors.

BACKGROUND: Recent findings suggest increased excitatory heteronymous feedback from quadriceps onto soleus may contribute to abnormal coactivation of knee and ankle extensors after stroke. However, there is lack of consensus on whether persons post-stroke exhibit altered heteronymous reflexes and, when present, the origin of increased excitation (i.e. increased excitation alone and/or decreased inhibition). This study examined heteronymous excitation and inhibition from quadriceps onto soleus in paretic, nonparetic, and age-matched control limbs to determine whether increased excitation was du

Effects of obturator and femoral nerve blocks on pain and kinematic gait parameters in patients undergoing unicompartmental knee arthroplasty: a retrospective cohort study.

BACKGROUND: Postoperative data indicate that 15-30% of patients undergoing unicompartmental knee arthroplasty (UKA) experience moderate-to-severe pain during the early recovery period, impeding rehabilitation. Due to the complex innervation in the knee, continuous femoral nerve block (FNB) is often administered but provides incomplete analgesia. Although the analgesic effects of nerve blocks are well studied, less is understood about their effects on postoperative rehabilitation and gait kinematics. Thus, in this study, we aimed to investigate the impact of ultrasound-guided obturator nerve bl

Which Patients With Dysfunctional Voiding Respond Well to Sacral Neuromodulation? ICI-RS 2025.

AIMS: Dysfunctional voiding (DV) is characterised by fluctuating or intermittent urinary flow during voiding in neurologically normal individuals. Given the different definitions used and heterogeneous pathophysiologies, outcomes following sacral neuromodulation/sacral nerve stimulation (SNM/SNS) are variably reported. The aim was to identify the areas of research required to be able to accurately predict response to SNM/SNS in adults with DV. METHODS: The relevant literature was reviewed by a multidisciplinary panel and the findings were discussed at the ICI-RS meeting held in 2025 in the UK

Aβ low threshold mechanoreceptors contribute to sensory abnormalities in fibromyalgia.

Fibromyalgia syndrome (FM) is characterized by widespread pain and fatigue. People living with FM also experience tactile allodynia, cold-evoked pain, paraesthesia and dysaesthesia. There is evidence of small fibre neuropathy and hyperexcitability of nociceptors in FM; however, the presence of other sensory abnormalities suggests involvement of large diameter sensory fibres. The passive transfer of FM IgG to mice causes cold and mechanical hyperalgesia associated with changes in A- and C-nociceptor function. However, whether FM IgG also confers sensitivity to light touch and whether large diam

Preserved finger flexion following high median nerve transection: a rare case report and review of literature.

BACKGROUND: High-level complete transection of the median nerve will impair the flexion function and sensation of the thumb and index finger, and will also result in weakened wrist flexion strength. In rare cases, atypical clinical manifestations may arise, potentially due to ulnar-to-median nerve anomalies, such as Marinacci communication (MC), or the function of muscles directly innervated by ulnar nerve branches. CASE PRESENTATION: A 52-year-old male sustained a chainsaw injury to his left elbow and forearm, resulting in complete transection of the median nerve. Despite this, he retained p

The effect of end-tidal carbon dioxide on optic nerve diameter in COPD patients: A cross-sectional study.

Chronic obstructive pulmonary disease (COPD) is a heterogeneous condition that causes persistent and often progressive airway obstruction with symptoms such as dyspnea, cough, expectoration, and/or exacerbations due to abnormalities in the airways and/or alveol. It is known that hypercarbia causes headaches by increasing intracranial pressure (ICP). Ultrasonographic measurement of optic nerve sheath diameter (ONSD) noninvasively has been shown to be as successful as invasive measurements in demonstrating increased intracranial pressure. The aim of this study was to determine the effect of the

A role for fibroblast and mural cell subsets in a nerve ligation model of neuropathic pain?

Neuropathic pain is a particularly intractable type of chronic pain that can result from physical nerve damage due to surgery or entrapment. Here, we present data which suggest that a particular subclass of fibroblast and mural cells may be implicated in the sensory neuron dysfunction that is characteristic of this pain state. In a mouse model of traumatic painful neuropathy, we used cell sorting, nerve tissue clearing and RNA sequencing to study stromal cells. With cell sorting (n = 4 mouse nerves) and tissue clearing (n = 5), we show that fibroblasts and mural cells positive for the platelet
