Emergence of a novel zoonotic brugian filarial infection during post-validation surveillance for lymphatic filariasis in Sri Lanka.

BACKGROUND: Lymphatic filariasis (LF) affects approximately 120 million people globally and is caused by Wuchereria bancrofti, Brugia malayi and Brugia timori. Sri Lanka was historically endemic for both bancroftian and brugian filariasis, with B. malayi presumed eliminated by the late 1960s. The country achieved validation of LF elimination as a public health problem in 2016. However, sporadic Brugia infections have been detected in microfilaria surveys from the mid-2000s onwards; the sub-periodic periodicity of which is suggestive of a zoonotic origin and warrants further research. METHODS:

Illuminating the cytological intricacies of isolated filarial epitrochlear lymphadenitis: a case series of a seldom-seen clinicopathological occurrence with a review of the literature.

BACKGROUND: Lymphatic filariasis is a major parasitic disease caused by Wuchereria bancrofti, Brugia malayi, and Brugia timori, with India contributing a significant proportion of global cases. Although lymphadenitis commonly involves inguinal, axillary, or cervical nodes, isolated epitrochlear lymphadenopathy is exceptionally rare and can be mistaken for other infectious or neoplastic causes. Fine-needle aspiration cytology (FNAC) offers a rapid, cost-effective method for diagnosing filarial infections, especially when peripheral blood smears are negative. This study presents a series of isol

Diethylcarbamazine: A review of its modes of action.

Filariases are neglected tropical diseases caused by Clade III parasitic nematodes transmitted by biting insects as microfilaria which mature into female and male adults in the host. One example of filariasis is lymphatic filariasis or elephantiasis, caused by three species of filaria: Brugia malayi, Wuchereria bancrofti, and/or Brugia timori. Elephantiasis results from the occlusion of lymphatic vessels by adult parasites and subsequent swelling of the limbs. It is a disfiguring disease that occurs in 120 million people worldwide. Another filarial disease is onchocerciasis caused by Onchocerc

An Overview on Antifilarial Efficacy of Heterocyclic Motifs Encompassing Synthetic Strategies, SAR and Commercialized Medications.

Filariasis is one of the oldest, most dangerous, debilitating, disfiguring diseases and often ignores tropical disorders. It presents with a range of clinical symptoms, a low death rate, and a high morbidity rate, which contributes to social discrimination. This condition has major effects on people's socioeconomic circumstances. This illness is carried by mosquitoes that have spread malaria. Lymphatic filariasis, caused by Wuchereria bancrofti, Brugia malayi, and Brugia timori, is a crippling illness with serious social and economic consequences. The infection persisted despite therapy with c

Profiling the broad antibody diversity of lymphatic filariasis immune antibody repertoire by deep sequencing.

Lymphatic filariasis is caused by infections of thread-like filarial worms, namely Wuchereria bancrofti, Brugia Malayi and Brugia timori. However, in-depth analysis of the antibody repertoire against Lymphatic filariasis is lacking. Using high-throughput sequencing of antibody repertoires, immunome analysis of IgG (LG) and IgM (LM) repertoires were studied. Despite significant differences between LG and LM in V(D)J gene usage, IGHV4-34, IGHV6-1, IGHD3-10 and IGHJ4 were preferred in both repertoires. The CDR3 in the LG repertoire showed a longer length than LM. Higher SHM level were observed in

Potential Anti-Filarial Molecules Against ATP Binding Site of MurE Enzyme: A Molecular Docking and Dynamics Approach to Combat Lymphatic Filariasis.

Lymphatic filariasis (LF) is a mosquito-borne disease caused by parasitic nematodes Brugia malayi, Brugia timori, and Wuchereria bancrofti. The drugs available are effective in several cases, and the absence of vaccination is the crucial factor hindering the elimination of LF. The UDP-N-acetylmuramoyl-L-alanyl-D-glutamate-2,6-diaminopimelate ligase (MurE) plays an important role in the peptidoglycan biosynthesis of Wolbachia endosymbiont B. malayi, which are reported to be a vital drug target for bacterial and endosymbiotic hosts. Thus, we selected the ATP binding cavity of MurE as the potenti

Current perspectives in the epidemiology and control of lymphatic filariasis.

SUMMARYLymphatic filariasis (LF), a debilitating tropical disease caused by parasitic filarial worms, Wuchereria bancrofti, Brugia malayi, and Brugia timori, remains a significant public health challenge in tropical and subtropical settings where the disease is endemic. The disease affects millions worldwide, leading to severe disability and social stigma. Following the World Health Assembly resolution WHA50.29 in 1997 encouraging Member States to eliminate LF as a public health problem, the Global Programme to Eliminate Lymphatic Filariasis (GPELF) was established in 2000. The establishment o

Robust COVID-19 Vaccine Responses Despite Filarial Co-Infection: Insights from a Lymphatic Filariasis Cohort in Ghana.

BACKGROUND/OBJECTIVES: Although the COVID-19 pandemic has largely concluded, the varied trajectories it has followed in different regions of the world remain incompletely understood. Intensive research is needed to fully grasp its course and the implications for future global health challenges. Notably, the milder trajectory of the COVID-19 pandemic in Sub-Saharan Africa has defied initial predictions. An emerging body of evidence suggests that, in addition to the continent's younger average age and the lower prevalence of relevant comorbidities, co-infections with helminths may have also impr

In silico screening of flavonoid-based inhibitors targeting UDP-galactopyranose mutase of Brugia malayi for lymphatic filariasis drug design.

Lymphatic filariasis (LF) is a vector-borne disease caused by parasitic roundworms, such as Wuchereria bancrofti, Brugia malayi, and Brugia timori, transmitted through mosquito vectors. Drugs such as albendazole, ivermectin, and diethylcarbamazine are used to treat LF; however, they mainly act against the microfilarial stage and have limited effect on the adult worms. So, it is time to identify or design a more efficient drug against the LF. Targeting the UDP-galactopyranose mutase (UGM) enzyme offers an effective strategy for treating lymphatic filariasis, as this enzyme is absent in mammals.

Wolbachia and Lymphatic Filarial Nematodes and Their Implications in the Pathogenesis of the Disease.

Lymphatic filariasis (LF) is an infection of three closely related filarial worms such as Wuchereria bancrofti, Brugia malayi, and Brugia timori. These worms can cause a devastating disease that involves acute and chronic lymphoedema of the extremities, which can cause elephantiasis in both sexes and hydroceles in males. These important public health nematodes were found to have a mutualistic relationship with intracellular bacteria of the genus Wolbachia, which is essential for the development and survival of the nematode. The host's inflammatory response to parasites and possibly also to the
