Abnormal left ventricular mass as a prognostic marker for outcomes in infants and children with cirrhosis.

BACKGROUND/AIMS: Cirrhotic cardiomyopathy (CCM), a clinically consequential comorbidity of cirrhosis, remains undefined and underappreciated in children. This study aimed to characterize myocardial structural, functional, morphometric, and histopathologic features in pediatric cirrhosis and objectively derive a reliable echocardiographic (2DE) metric to predict perioperative adverse events, including mortality. METHODS: 2DE, morphometry, histopathology, and vasoactive-inotropic score (VIS) were compared between children with (n=226) and without (n=160) cirrhosis listed for liver transplantati

Hepatoprotective action of Dendrobium officinale polysaccharide against MAFLD: modulation of gut microbiota-metabolite cascade controls TLR4/NF-κB inflammatory response.

INTRODUCTION: Metabolic dysfunction-associated fatty liver disease (MAFLD) threatens public health worldwide, and Dendrobium officinale polysaccharide (DOP) exhibits promising hepatoprotective potential, yet its regulatory mechanism targeting the gut microbiota-metabolite-TLR4/NF-κB inflammatory axis remains poorly defined. METHODS: In this study, in-vivo rat and in-vitro HepG2 cell MAFLD models were established. Male Sprague-Dawley (SD) rats received 12-week high-fat diet (HFD) feeding to induce MAFLD; HepG2 cells were exposed to a 1 mmol/L free fatty acid (FFA) mixture (oleic acid/palmitic

The impact of nephrectomy on liver and contralateral kidney volume in patients with ADPKD; a case series.

PURPOSE: A substantial number of patients with autosomal dominant polycystic kidney disease (ADPKD) undergo nephrectomy. A decrease in intra-abdominal pressure after nephrectomy might hypothetically accelerate the growth of other polycystic organs and accelerate the rate of eGFR decline of the remaining kidney. In this case-series, we investigated the change in kidney and liver volume in patients before and after nephrectomy, and in a control group without nephrectomy. METHODS: In this retrospective case series kidney and liver volumes were measured using an automated stereological approach (

Pleuroparenchymal fibroelastosis in a monogenic mitochondrial DNA depletion syndrome.

BACKGROUND: Pleuroparenchymal fibroelastosis (PPFE) is a rare, most often idiopathic fibrotic lung disease with poor prognosis, mainly affecting adults. Biallelic pathogenic variants in the deoxyguanosine kinase gene (DGUOK) cause a mitochondrial DNA (mtDNA) depletion syndrome, predominantly affecting infants with severe hepatic and neurological disease. Neither detailed pulmonary manifestations nor a pulmonary-dominant phenotype has been reported. METHODS: We describe nine patients with PPFE and DGUOK-associated mitochondriopathy. Clinical, radiological, histopathological, and genetic data w

Calpain-1 deficiency alleviates oxidative stress and hepatic injury in ApoE-deficient mice with MASH.

BACKGROUND: Metabolic dysfunction-associated steatohepatitis (MASH) is a critical stage of metabolic dysfunction-associated fatty liver disease (MAFLD) characterized by hepatocyte ballooning, inflammatory infiltration, and fibrosis, often accompanied by oxidative stress, lipotoxicity, and apoptosis. Calpain-1 is involved in the regulation of oxidative stress and inflammation in cellular stress responses, but its role in MASH remains unclear. METHODS: This study established MASH models in ApoE knockout (ApoE KO) and ApoE/calpain-1 double knockout (AxC DKO) mice. Serum lipids and liver function

Efficacy and safety of PCSK9 inhibitors combined with statins in acute ischemic stroke: a systematic review and meta-analysis.

BACKGROUND: Acute ischemic stroke (AIS) imposes a substantial global burden of mortality and long-term disability. As novel lipid-modulating agents, proprotein convertase subtilisin/kexin type 9 inhibitors (PCSK9i) have demonstrated therapeutic benefits across a wide range of cardiovascular patient groups, but dedicated evidence in AIS cohorts has not been quantitatively synthesized. We therefore conducted this meta-analysis to compare the efficacy and safety of PCSK9i combined with statin therapy versus statin monotherapy among patients with AIS. METHODS: We searched seven databases from inc

Spondweni virus infection in pregnant rhesus macaques causes placental pathology without apparent fetal harm.

UNLABELLED: The 2015-2016 Zika virus (ZIKV) epidemic revealed the potential of flaviviruses to emerge rapidly, cause severe disease, and affect pregnancy outcomes. In 2016, Spondweni virus (SPOV), the closest known relative of ZIKV, was detected in mosquitoes in Haiti, suggesting it may also have the potential to emerge in the Western Hemisphere. The risks that close relatives of ZIKV pose to pregnant individuals are not well understood. Previously, we showed that SPOV can cause fetal demise, placental pathology, and vertical transmission in a mouse model. Here we report SPOV's pathogenic pote

A20 promotes OPN ubiquitination and degradation to inhibit cell growth and invasion in human hepatocellular carcinoma.

Abnormal osteopontin (OPN) expression has been implicated in hepatocellular carcinoma (HCC) progression. In this study, we assessed the prognostic value and biological functions of OPN in HCC. We also identified that A20 interacts with OPN and promotes OPN ubiquitination and protein degradation through the ubiquitin-proteasome system. The reduction in OPN levels induced by A20 attenuates OPN-associated proliferative and migratory phenotypes in HCC cells. Notably, advanced HCC tissues exhibited elevated OPN levels, while A20 expression showed variable patterns in clinical samples. In the clinic

Sestrin1 Ameliorates Senescence and Mediates the Protective Effects of Exercise Against Hepatic Abnormality in C57BL/6J Mice.

Liver aging is characterized by structural remodeling and functional decline, frequently driven by mitochondrial dysfunction and senescence-associated secretory phenotype (SASP). Sestrins (SESNs) are a family of stress-inducible proteins, among which Sestrin1 and Sestrin2 are crucial for maintaining metabolic and redox homeostasis, yet their specific involvement in hepatic senescence remains unclear. In the present study, in vivo experiments were performed using WT mice at young, middle, and old ages, together with aged Sestrin1-/- and Sestrin2-/- mice. The aged mice displayed multifaceted hep

Cirrhotic Cardiomyopathy: Proportion and Correlation with Liver Fibrosis Severity.

BACKGROUND: Cirrhotic cardiomyopathy is characterized by an impaired systolic response to stress, diastolic dysfunction, and electrophysiological abnormalities, without underlying structural heart disease. Left ventricular diastolic dysfunction (LVDD) is the most common manifestation found in cirrhotic cardiomyopathy. Previous studies reported its correlation with a higher Child-Turcotte-Pugh (CTP) score in late-stage cirrhosis. To date, no studies have evaluated the correlation between liver fibrosis and LVDD parameters. This study aims to evaluate the correlation between liver fibrosis and L

ERα/AR imbalance underlies enhanced hepatic susceptibility to pentachlorophenol sodium in female mice.

sodium pentachlorophenol (PCP-Na) is one of the persistent organic pollutants (POPs) with high lipophilicity. chlorinated hydrocarbon that exhibits broad-spectrum systemic toxicity, accompanied by severe carcinogenic, teratogenic, and mutagenic risks; however, the molecular basis of its gender-specific pathogenesis remains unclear. In this study, mice were exposed to PCP-Na by intraperitoneal injection for 10 days (2.5, 5, and 10 mg/kg). To elucidate the gender-sensitive endocrine mechanism, we evaluated the expression of AR and ERα in the liver and conducted targeted hormone receptor interven

Characterization of posttranslational modification in metabolic dysfunction-associated steatohepatitis: acetylation, lactylation, and phosphorylation.

In metabolic dysfunction-associated steatohepatitis (MASH), abnormalities in posttranslational modification (PTM) are both a consequence of the pathological process and a driving force for disease progression. Therefore, this study intends to characterize the PTM features (acetylation, lactylation, and phosphorylation) of MASH, which will provide a crucial theoretical basis for the diagnosis and development of new targets for MASH. In this study, protein/modification profiles of clinical liver tissues from Normal and MASH were analyzed by LC-MS to identify differentially expressed proteins (DE

Smoking reshapes systemic metabolic coordination across lung cancer subtypes: a multicenter total-body 18F-FDG PET/CT study with tumor habitat imaging.

PURPOSE: Lung cancer (LC) is increasingly recognized as a systemic disease, and these systemic alterations may differ across histological subtypes. Smoking is a major risk factor for LC, yet its impact on systemic metabolism across LC subtypes remains underexplored. METHODS: We studied 125 LC patients and 200 healthy controls from three centers who underwent static and dynamic total-body 18F-FDG PET/CT. Thirty-seven regions of interest spanning the central nervous system and peripheral organs were analyzed. Regional standardized uptake values normalized by lean body mass and time-activity cur

A novel stop-loss variant in ISG15 causes C-terminal dysfunction and impairs USP18-mediated negative feedback.

INTRODUCTION: ISG15 is a type I interferon (IFN-I)-inducible ubiquitin-like protein that restrains IFN-I signaling. Classical ISG15 deficiency is thought to cause disease mainly by disrupting USP18 stabilization, which leads to sustained IFN-I signaling and interferon-driven autoinflammation. Whether patient-derived C-terminal ISG15 variants can impair IFN-I negative feedback through altered C-terminal functional presentation has remained unclear. METHODS: We studied a Chinese boy with early-onset recurrent fever, severe ulcerative skin lesions, and elevated liver enzymes. Whole-exome sequenc
