Causal Links of Type 2 Diabetes and Its Complications With Cortical Modification: A Mendelian Randomization and Mediation Analysis.

BACKGROUND: Type 2 diabetes (T2D) and its complications are linked to cognitive decline over time, accompanied by cortical abnormalities. The causality between T2D and cortical modifications, however, remains elusive. METHODS: Applying two-sample Mendelian randomization (MR), we probed causal relationships between T2D and its complications and cortical modifications. Genetic associations were elucidated via linkage disequilibrium score regression and Bayesian colocalization. An instrumental variable-guided protein-protein interaction (PPI) network was constructed and subjected to clustering a

Prevalence of Germline Pathogenic RET Variants in a Pan-Cancer Patient Population.

PURPOSE: Germline likely pathogenic or pathogenic variants (LP/PV) in the RET proto-oncogene account for approximately 25% of medullary thyroid cancers (MTCs). Depending on the variant, individuals with RET LP/PV can have > 70% lifetime risk for MTC or C-cell hyperplasia, an MTC precursor. We assessed prevalence and genotype-phenotype correlations of RET LP/PV in a pan-cancer population undergoing agnostic multigene germline cancer genetic testing. METHODS: Patients with cancer at Memorial Sloan Kettering Cancer Center provided informed consent to Memorial Sloan Kettering-Integrated Mutation

Safety evaluation of DHA canola high-fat meal: A 13-week dietary feeding study in rats.

Long-chain omega-3 fatty acids, including eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), are essential omega-3 fatty acids that play crucial roles in human and animal health. EPA and DHA, are mainly sourced from marine species, such as fish, but rising demand and environmental concerns have driven the development of sustainable and environmental friendly alternatives. DHA canola offers a solution by expressing seven transmembrane enzymes that enable long-chain omega-3 production in canola seed. It also contains the pat gene, providing resistance to phosphinothricin and glufosinate

First large-scale screening of Notch biallelic variants implicates novel candidate genes in congenital hypothyroidism.

CONTEXT: Congenital hypothyroidism (CH) is a major preventable cause of developmental and cognitive impairment. Current genetic testing identifies causative variants for only half of cases, leaving the majority without a molecular diagnosis. Although animal studies implicate Notch signaling in thyroid development, its role in human CH remains unexplored. OBJECTIVE: To investigate whether Notch pathway variants contribute to genetically unexplained CH. DESIGN: Genetic screening using whole-exome sequencing with zebrafish functional validation. SETTING: Endocrinology outpatient clinic at Shan

Long-term risk of hypothyroidism after thyroid abnormalities identified in pregnancy.

BACKGROUND: When thyroid analytes are assessed during pregnancy, subclinical hypothyroidism and thyroid peroxidase antibodies may be identified in otherwise asymptomatic individuals. However, the long-term risk of progression to overt hypothyroidism in this population remains poorly defined. Understanding this trajectory is essential for informing long-term thyroid surveillance strategies. OBJECTIVE: To determine the long-term incidence and timing of overt hypothyroidism among individuals with subclinical hypothyroidism, thyroid peroxidase antibodies, or both identified during pregnancy and t

A study on the differences in urinary iodine and serum iodine in relation to children's iodine nutrition and lipids.

The safe intake range for iodine is narrow, and abnormal nutritional status (deficiency or excess) may affect children's lipid metabolism by interfering with thyroid function. Following the widespread implementation of iodized salt in China, urinary iodine levels in children in some regions have exceeded the appropriate range. Nonetheless, the connection between iodine nutritional status and childhood dyslipidemia remains unclear. This study examined the associations between iodine status and lipid profiles among 6-17-year-old children and adolescents residing in regions of China where iodized
