System-level clustering of testosterone-related biomarkers identifies high-risk aging profiles linked to inflammation and renal function.

BACKGROUND: Serum total testosterone (TT) interacts with multiple physiological systems and is implicated in heterogeneous aging processes in men. However, aging-related phenotypes associated with TT are unlikely to be captured by single biomarkers or conventional clinical categories. This study aims to identify data-driven aging phenotypes based on TT and related clinical biomarkers using an unsupervised analytical framework. METHODS: Clinical laboratory data from 5,877 Japanese male patients undergoing routine health evaluations are analyzed. After restricting the cohort to individuals with

Assay-Specific Diagnostic Thresholds for Adrenal Insufficiency: A Retrospective Comparison of Monoclonal Cortisol Measurements Using Roche Elecsys Cortisol II and Tosoh AIA-CL2400.

Background The diagnosis of adrenal insufficiency traditionally relies on peak cortisol values during stimulation testing, with a cut-off of 18 µg/dL established using polyclonal antibody assays. However, newer monoclonal antibody-based assays show reduced cross-reactivity with other glucocorticoids. In April 2022, our hospital changed the cortisol measurement method from Roche Elecsys Cortisol II to Tosoh AIA-CL2400, both using monoclonal antibodies but with potentially different measurement characteristics. In addition to stimulation test results, physicians initiate hydrocortisone replaceme

Reducing exposure to non-persistent endocrine-disrupting chemicals and their impact on human health: Evidence from dietary, behavioral, and residential interventions.

BACKGROUND: Endocrine-disrupting chemicals (EDCs) are widespread environmental contaminants that interfere with hormonal signaling and are associated with adverse reproductive, metabolic, thyroid, neurodevelopmental, and other endocrine-related health outcomes. Diet, consumer products, and indoor environments are major sources of exposure. While regulatory approaches are critical, practical interventions to reduce exposure and mitigate health effects are increasingly being explored. OBJECTIVE: This narrative review synthesizes evidence on interventions that reduce human exposure to EDCs. This

Chronic health effects of recurrent indoor Pyrethroid exposure in enclosed workplaces: an occupational exposome-based clinical review in the context of limited safety guidance.

Pyrethroid insecticides are widely used indoors in residential, commercial, and occupational pest-control settings and are generally regarded as safe because of their relatively low acute toxicity. This perception is largely based on short-term exposure data and application practices conducted under well-ventilated conditions. However, indoor pesticide application does not always occur in environments that allow for rapid residue clearance. In enclosed workplaces-particularly those with centralized heating, ventilation, and air-conditioning systems, limited fresh-air exchange, and dust-retaini

Balancing oncologic risk and fertility potential: a single-center study on Turner syndrome patients with Y chromosome material.

INTRODUCTION: Turner Syndrome (TS) is a chromosomal disorder characterized by partial or complete monosomy of the X chromosome. Approximately 5-10% of patients with TS harbor Y chromosome material, which is associated with an increased risk of gonadoblastoma and other gonadal malignancies. However, the indications and optimal timing of prophylactic gonadectomy remain controversial, particularly in asymptomatic individuals, because a subset of TS patients with Y chromosome material may retain residual gonadal function and fertility potential. This study aimed to determine the prevalence of Y ch

Low Energy Availability in Male Indian Athletes: A Cross-Sectional Study.

BACKGROUND: Low energy availability (LEA) is a mismatch between dietary energy intake and training-related expenditure that can impair performance, recovery, endocrine function, bone health, and overall athlete well-being; it is most often expected in endurance, weight-category, and physique-sensitive sports but can occur across disciplines. In male athletes, LEA is under-recognized because symptoms are subtle and non-specific and are frequently attributed to training fatigue. Because abrupt workload fluctuations may further strain recovery, the acute to chronic workload ratio (ACWR) provides

Water Fluoridation Risks or Benefits: Summary of the Symposium Presented During the Enamel 11 Meeting Held in Paris France.

Fluoride is widely used in dentistry to prevent dental caries. Fluoride can be naturally present in ground waters, and as a public health measure, many countries have added fluoride to drinking water. Experimental studies show that fluoride can affect the cells of the developing tooth organ to result in dental fluorosis, and cellular effects of fluoride raise concerns about potential neurodevelopmental and endocrine effects. Evidence of increasing levels of fluorosis in populations, have led regulatory organizations to revise their recommendations for optimal doses. To review the scientific ev

The Oxidative Stress-Sensitive Ca2+ Channel, TRPM2, Contributes to Diabetes Progression Accompanied by ER Stress.

Beta cell dysfunction is a critical factor contributing to decreases in insulin secretion. A growing body of evidence suggests the involvement of endoplasmic reticulum (ER) stress in beta cell dysfunction, which leads to the development of diabetes. On the other hand, oxidative stress also plays a role in the development of diabetes with beta cell dysfunction. Transient receptor potential melastatin 2 (TRPM2) is a Ca2+-permeable channel activated by oxidative stress and is highly expressed in beta cells. Some studies have claimed that Ca2+ influxes via TRPM2 in response to oxidative stress are

Clinical and molecular features of ovarian stimulation in peripubertal girls with mosaic Turner's syndrome.

STUDY QUESTION: Do peripubertal girls with mosaic Turner's syndrome (TS) respond to ovarian stimulation (OS) for oocyte freezing as adult women with normal ovarian reserve? SUMMARY ANSWER: Clinical and molecular reproductive/endocrine features of OS in these patients are similar to those of adult females. WHAT IS KNOWN ALREADY: OS for oocyte freezing is quite a new concept in peripubertal and young adolescent girls with TS because ovarian tissue cryopreservation (OTC) does not have proven efficacy, likely due to already diminished ovarian reserve and accelerated follicle atresia. No data are

Karyotype-based evaluation of ovarian reserve before ovarian tissue cryopreservation in Turner syndrome patients.

BACKGROUND: Turner syndrome (TS) girls experience accelerated ovarian reserve depletion, exhibiting a strong demand for ovarian tissue cryopreservation (OTC) to preserve fertility and ovarian function. However, the current knowledge of OTC in TS is limited, and predicting follicular presence is insufficient. This study aims to investigate ovarian follicular reserve based on karyotypes, thereby contributing empirical data to informed decision-making in TS populations. METHODS: The retrospective case-control study comprised 26 TS patients and 52 age-matched non-TS (NTS) controls who underwent OT

Shared Genetic Architecture of Premenstrual Disorder and Postpartum Depression: Registry-Based and Genetic Evidence.

BACKGROUND: Premenstrual disorder (PMD) and postpartum depression (PPD) have a strong phenotypic link and echo women's hormone fluctuations. Yet, the extent to which they may be cross-inherited remains poorly understood. METHODS: Using the nationwide cohort of 907,841 women who born 1950-2007 and gave birth during 2001-2021 in Sweden, we estimated the cumulative incidence functions-based heritability and genetic correlation for PMD and PPD. We also analyzed genome-wide association study (GWAS) summary statistics from the largest European-ancestry cohorts for PMD (17,511 cases and 54,786 contr

Aberrations in Peripheral Blood Neuron-Specific Enolase Levels in Depression: A Systematic Review and Meta-Analysis.

PURPOSE: Neuron-specific enolase (NSE) is a type of glycolytic enzyme enolase. As NSE is predominantly localized in endocrine cells and neurons, it is frequently recognized as a specific biomarker for peripheral neuroendocrine cells and neurons. Recently, numerous clinical studies have investigated the correlation between peripheral blood NSE levels and depression; however, the findings remain inconsistent. The objective of this study is to systematically evaluate peripheral blood NSE levels in patients with various types of depression. METHODS: We conducted an extensive literature review usi

Myxedema Coma Despite Mild Biochemical Hypothyroidism: A Case of Profound Clinical Decompensation.

Myxedema coma is a rare endocrine emergency with high mortality that is typically associated with severe biochemical hypothyroidism. However, profound clinical decompensation may occur despite only modest abnormalities in thyroid function tests, making diagnosis challenging. We report the case of an 87-year-old woman with long-standing hypothyroidism, primary adrenal insufficiency, pituitary pathology, and ischemic heart disease who presented with an Escherichia coli urinary tract infection and subsequently developed profound hypothermia, severe bradycardia, hypotension, reduced consciousness,
