Pseudohypoxia caused by germline genetic alterations in the VHL gene is associated with increased diabetes and cardiovascular risk: a UK biobank study.

BACKGROUND: Von Hippel-Lindau protein deficiency leads to cellular and tissue false sense of hypoxia (pseudohypoxia), driving erythropoiesis, angiogenesis, and dysglycemia. The impact of partial VHL protein deficiency, caused by heterozygous VHL gene alterations on diabetes and cardiovascular risk has not been investigated. Hence, in the current study we assessed a possible association between VHL genotype and cardiovascular risk based on a the UK Biobank genomic and clinical data. METHODS: Demographic, clinical and biochemical data were extracted, and exome analysis, focusing on the VHL gene

Loss of FUT8 impairs embryonic development by reducing cAMP production in granulosa cells.

PURPOSE: In cases of impaired fertility, the quality of oocytes and their subsequent embryonic development following fertilization are critical concerns in clinical practice. Core fucosylation, catalyzed by fucosyltransferase 8 (FUT8), is a common N-glycosylation modification that plays a key role in cell proliferation and signal transduction. This study examines the role of core fucosylation in follicular regulation during oocyte development. METHODS: Patients were categorized into two groups based on their embryo formation rates. Core fucosylation levels, catalyzed by FUT8, were assessed in

The prevalence of pathogenic variants in medically actionable genes among individuals with idiopathic hypogonadotropic hypogonadism/Kallmann syndrome.

PURPOSE: Idiopathic hypogonadotropic hypogonadism (IHH) and Kallmann syndrome (KS) are rare reproductive disorders with known genetic heterogeneity. Using exome sequencing, our group previously reported the prevalence of pathogenic and likely pathogenic (P/LP) variants in genes causing IHH/KS as the primary endpoint of our study. Here, we investigate the frequency of secondary findings (SF) to determine whether individuals with IHH/KS harbor an increased burden of P/LP variants in medically actionable genes (MAGs) defined by the American College of Medical Genetics and Genomics (ACMG). METHOD

STARD9 and CDK5RAP2-Novel Candidate Genes for 46,XY Complete Gonadal Dysgenesis.

46,XY gonadal dysgenesis, characterised by absent or defective testicular development in individuals with a 46,XY karyotype, results from disruptions in the genetic programme governing testis determination and differentiation during embryogenesis. While monogenic causes explain approximately 50% of cases, emerging evidence suggests an oligogenic basis in some patients. However, many cases remain without a definitive molecular diagnosis. In this study, we investigated a patient with 46,XY gonadal dysgenesis to explore the underlying genetic aetiology. Whole-exome sequencing in the patient did n

Acute exposure to antimony elicits endocrine disturbances, leading to PCOS and ovarian fibrosis in female zebrafish.

Antimony (Sb) is an estrogenic metal. Exogenous exposure to Sb can affect estrogen levels and their receptor expression in organisms, exerting estrogen-disrupting effects and even inducing polycystic ovary syndrome (PCOS), which is accompanied by the progression of ovarian fibrosis. To investigate the pathological mechanism of this reproductive damage caused by Sb exposure, we exposed female zebrafish to Sb solution for 18 days for acute toxicity experiments. The results showed that Sb exposure affected the changes of GnRH, FSH, LH, E2 and T levels on the HPG axis, which disrupted the balance

Olfactory Receptor Gene Deletions: A Novel Genetic Findings in Idiopathic Prepubertal Gynecomastia and Juvenile Macromastia.

INTRODUCTION: Prepubertal gynecomastia (PG) and juvenile macromastia (JM) are rare breast overgrowth phenotypes arising from hormonal or genetic abnormalities. While many cases reflect physiological or endocrine variation, a subset may involve increased peripheral aromatization driven by genomic factors. The molecular basis remains unknown in many idiopathic cases. We evaluated whether copy number variations (CNVs) in CYP19A1 and other estrogen-related genes are associated with PG or JM. METHODS: We performed a cross-sectional study of 17 patients suspected of aromatase excess syndrome: 10 bo

Alterations of insulin sensitivity, clearance, and secretion, either alone or in combination, in women with PCOS: impact on metabolic profile and androgenemia.

STUDY QUESTION: What is the role of insulin resistance (IR), increased insulin secretion, and impaired insulin clearance, either alone or in combination, on metabolic features and androgen levels in women with PCOS? SUMMARY ANSWER: IR, reduced insulin clearance and increased insulin secretion independently contribute to predicting hyperandrogenemia, whereas metabolic abnormalities may differ according to the mechanisms underlying hyperinsulinemia. WHAT IS KNOWN ALREADY: Hyperinsulinemia is a common finding in women with PCOS, and it is closely associated with the metabolic and endocrine alte

Bone Material Strength Index Is Low in Patients With Cushing's Syndrome Even After Long-term Remission.

OBJECTIVE: Hypercortisolism in endogenous Cushing's syndrome (CS) results in decreased bone mineral density (BMD) and increased fracture risk. Although after remission BMD improves, the fracture rate remains elevated, suggesting that BMD may not adequately reflect fracture risk in this group. The aim was to evaluate bone material properties, another component of bone quality, using impact microindentation in patients with CS in remission. METHODS: Cross-sectional study in 60 CS patients and 60 age-, sex-, and BMD-matched controls at a tertiary referral center between 2019 and 2021. Bone mater
