Clinical and Laboratory Dynamics of Bear Meat-Associated Trichinellosis in Hokkaido, Japan.

Trichinellosis is a rare infection associated with bear meat consumption found in Japan, where limited access to parasite-specific antibody testing makes definitive diagnosis challenging. In the present study, 19 patients, comprising 16 confirmed and three probable cases, treated at a tertiary care hospital in Hokkaido, Japan, during three independent outbreaks in 2018, 2019, and 2025, were retrospectively analyzed. Patients prepared brown bear meat as a roast (n = 18) or a cutlet (n = 1) and consumed it as a main dish (n = 7) or in smaller portions of a few slices (n = 12). Ten patients consu

Ameliorative effects of resveratrol versus sulforaphane on muscular trichinellosis in mice: The role of SIRT1 and SLC7A11 expression.

Trichinella spiralis (T. spiralis) infection induces oxidative stress, inflammation, and increased larval burden. Albendazole (ABZ) remains the standard treatment; however, adjunct therapies may enhance host protection. Thus, this study aimed to comparatively evaluate, for the first time, the individual therapeutic and antioxidant effects of resveratrol (RSV) and sulforaphane (SFN) relative to standard ABZ therapy against trichinellosis, along with the associated biochemical and histopathological changes in mice. Mice were divided into five main groups: (I): Non-infected, non-treated; (II): In

The Palette of Science and Emotions: Art-Based Learning With Structured Peer Role-Plays for Early Clinical Exposure in Biochemistry.

INTRODUCTION: Biochemistry is often regarded as conceptually dense yet emotionally disconnected, limiting its relevance to patient care. To bridge this gap, an art-based learning intervention was developed to connect biochemistry of diseases with empathy, perspective-taking, and ethical awareness through visual thinking strategies, structured peer role-play, and guided reflection. METHODS: This small-group activity was conducted for 150 first-year medical students over a total duration of 180 minutes. Students were divided into 30 groups and each group received a curated artwork and a biochem

Trichinellosis knowledge and preventive practices in Mapuche communities of southern Chile: Evidence for targeted One Health implementation.

Trichinellosis is a foodborne zoonosis that persists in rural areas where backyard pig farming and informal slaughtering occur. In Indigenous communities, prevention depends on interconnected human behaviors, animal management, and local environmental factors. This study aimed to describe knowledge and preventive practices regarding trichinellosis among Mapuche communities in south-central Chile and to assess whether higher knowledge is associated with safer practices, while controlling for community and demographic factors. A cross-sectional survey was conducted among 180 adults from nine Map

Discovery of novel repurposed anthelminthics against Trichinella spiralis and albendazole-resistant nematodes through metabolomics-guided virtual screening.

Trichinella spiralis is a parasitic nematode that causes trichinellosis, leading to weakness, muscle pain, facial edema, and potentially death. Albendazole (ABZ) is the current drug of choice; however, resistance has been increasingly reported, highlighting the urgent need for novel therapeutics. Developing new drugs is costly and time-consuming, making computational approaches and drug repurposing attractive alternatives. In this study, we combined metabolomics and virtual screening to identify potential anthelmintic candidates for trichinellosis and ABZ-resistant nematodes. Both adult and la

Validation of an optimized in-house enzyme-linked immunosorbent assay for enhanced detection of Trichinella spp. exposure in swine.

The International Commission on Trichinellosis (ICT) and the World Organization for Animal Health (WOAH) recommend the use of an indirect enzyme-linked immunosorbent assay (ELISA) that utilizes excretory-secretory (E-S) antigens (ESA) of Trichinella spiralis for surveillance and epidemiological studies in pigs and wild boars. Our efforts to optimize and standardize ESA production and ELISA protocols resulted in improved diagnostic performance of an in-house E-S ELISA. To validate the optimized assay, we compared its performance with that of a commercial E-S ELISA kit using sera from a represen

First Report of Trichinella spiralis in Free-Living Invasive American Mink (Neovison vison) in Lithuania.

The genus Trichinella comprises zoonotic nematodes infecting a wide range of carnivorous and omnivorous animals, including humans. Infection occurs through the consumption of raw or undercooked meat containing viable Trichinella larvae. Among the species within this genus, Trichinella spiralis is considered one of the most epidemiologically important due to its high reproductive capacity and its frequent association with infections in domestic animals and humans. In this study, muscle samples from 18 invasive American minks (Neovison vison) were examined for Trichinella larvae using the magnet

First Report of Trichinella britovi in Serbian Domestic Pigs Linked to Two Human Outbreaks.

Sporadic cases of trichinellosis have occurred almost every year in Serbia. The success of trichinellosis control in the country depends on effective communication among medical professionals who discover a case of trichinellosis, the Public Health Service, and the Veterinary Service, in line with the One Health concept. The collaborative approach, supported by a robust reporting and monitoring system, has been essential for identifying infection sources and preventing further spread. However, in March 2023, the National Reference Laboratory for Trichinellosis (NRLT) received reports of hospit

Protective effect of Lactococcus lactis NZ9000 secreting Bcl2 against Trichinella spiralis infection in mice.

BACKGROUND: Trichinellosis, a globally distributed zoonosis, threatens both human health and livestock productivity. The limited efficacy of existing anthelmintics underscores the need for innovative strategies to control Trichinella spiralis infection. RESULTS: A recombinant food-grade Lactococcus lactis strain (NZ-Bcl2) was engineered to secrete the anti-apoptotic protein Bcl2 by fusing the Bcl2 gene to the Usp45 signal peptide. Western blotting confirmed successful expression and secretion. NZ-Bcl2 strains exhibited stable growth, maintained plasmid inheritance over 24 generations without

Identification of maternal antibodies targeting a cystatin-like protein of Trichinella spiralis in offspring.

Vertical transfer of maternal antibodies can provide passive protection to offspring against specific pathogens. In this study, we detected antibodies in the sera of uninfected offspring born to chronically Trichinella spiralis-infected female mice. Immunoblotting consistently revealed a distinct band at ~38 kDa in both T. spiralis excretory-secretory products and total somatic extracts. This band was identified by MALDI-TOF/TOF mass spectrometry as a cystatin-like protein of T. spiralis (Ts-CLP). Structural modeling and domain analysis indicated a typical cystatin-like fold comprising a centr

Trichinellosis: A zoonosis that still requires vigilance.

UNLABELLED: Trichinellosis is a parasitic disease in humans caused by nematodes of the genus Trichinella, usually acquired through the consumption of raw or undercooked meat, mainly from pigs or wild animals, containing infective larvae. Although often overlooked, this disease has affected humans for centuries and continues to be a public health concern in many parts of the world, especially where food safety measures are inadequate. In this millennium, significant progress has been made in understanding the disease pathophysiology and host immune response, mainly through experimental studies

Mammalian senescent nurse cell provides Trichinella spp. muscle larvae with cell growth inhibitory capacity.

Muscle larva of the mammalian parasitic nematode Trichinella spp. resides in a single cell called a nurse cell (NC). Despite originating from muscle cells the NC is of the non-muscular type. Its only role is to sustain muscle larva throughout the host's life. Our previous studies documented cellular senescence as the main phenomenon associated with the NC development and its long-term maintenance. Cell-cycle arrest of the NC was underpinned by p57 Kip2 cyclin-dependent kinase inhibitor (CDKI) expression. Since p16 INK4a and p21 Cip1 CDKIs are recommended as the key markers of growth arrest in

Trichinellosis after Raw Bear Eyeball Consumption.

Trichinellosis, a zoonotic parasitic disease, is usually transmitted through raw or undercooked muscle tissue. We report a 25-year-old hunter from Hokkaido, Japan, who acquired trichinellosis by consuming a raw bear eyeball, a tissue previously presumed to be parasite-free. He developed fatigue and rash within 3 days, followed by fever and myalgia 9 days after consumption. Serology confirmed trichinellosis, and treatment with albendazole led to full recovery. Among five hunters who consumed meat from the same bear, only the two who ate raw eyeballs developed the disease, whereas those who ate

Transcriptomic profiling reveals Trichinella spiralis-mediated attenuation of respiratory syncytial virus-induced inflammation in mice.

Helminth-mediated immunomodulation has been extensively studied in animal models, demonstrating its potential as both a prophylactic and therapeutic option for inflammatory lung diseases. However, its role in attenuating respiratory virus-induced inflammation remains largely unexplored. In this study, we examined whether pre-existing infection with the helminth Trichinella spiralis confers protection against pulmonary pathology induced by respiratory syncytial virus (RSV) infection in mice. Mice with prior T. spiralis infection exhibited reduced pulmonary inflammation and lower viral titers in

Trichinella-Derived Extracellular Vesicles: Implications and Future Prospects.

Parasite extracellular vesicles (EVs) are concise and versatile messages for parasite-parasite and parasite-host crosstalk. These vesicles are loaded with a cargo of diverse heterogeneous molecules, some of which are of potent immunomodulatory nature, and others have specific functions. Those EVs carrying the Trichinella signature are no exception. They play pivotal roles in the establishment of the parasite inside its niche within the host, ensuring better survival for both. They can also serve as biomarkers for diagnosis, follow-up, and prognosis of Trichinella infection. Owing to their immu

Experimental infection of immunosuppressed mice demonstrates freeze tolerance of Trichinella chanchalensis.

Trichinella chanchalensis (T13) remains poorly characterized, limiting understanding of its biology, freeze-tolerance, and zoonotic potential. One major challenge is that infective first-stage larvae can only be obtained from naturally exposed wildlife in northern North America, which often harbour co-infections with other Trichinella taxa. Therefore, we used next-generation sequencing (NGS) to identify naturally infected wolverines with high proportions of T13 larvae. We recovered larvae from muscle tissue previously frozen at -20 °C for up to 15 months and orally inoculated a total of 55 CD-
