Maraviroc Prevents Optic Nerve Injury-Induced Retinal Ganglion Cell Apoptosis by Modulating the CCL5/CCR5/CTSS Axis.

PURPOSE: Optic nerve crush (ONC) is an animal model that induces optic nerve injury and retinal ganglion cell (RGC) death, which is partly mediated by activated microglia secreting cytokines. This study explores the role of the CCL5/CCR5 pathway in RGC damage and evaluates maraviroc, a CCR5 inhibitor, as a potential therapeutic agent. METHODS: Bioinformatic analysis of glaucoma datasets (GSE142881 and GSE26299) identified Ccr5 as a key gene linked to acute or chronic RGC injury. The ONC model was used to evaluate the neuroprotective effects of maraviroc or sh-Ccr5 using immunofluorescence, TU

From injury to recovery: Spatiotemporal dynamics of the visual pathway during spontaneous structural and functional regeneration after optic nerve transection in zebrafish.

In adult mammals, optic nerve injury leads to irreversible vision loss due to its extremely limited regenerative capacity. In contrast, adult zebrafish possess a robust capacity for spontaneous visual system regeneration, although the spatiotemporal coordination of recovery across the retina, optic nerve, and brain remains poorly understood. In the present study, the regenerative dynamics following optic nerve transection were systematically characterized in adult zebrafish over a 5 week period using hematoxylin-eosin staining, immunohistochemistry, transmission electron microscopy, single-cel

Human induced pluripotent stem cell-derived extracellular vesicles promote neuroprotection effects in rat optic nerve crush model.

Optic neuropathies significantly contribute to permanent vision loss, frequently characterized by the deterioration of retinal ganglion cells (RGCs) and axonal loss. Current therapeutic strategies remain insufficient. Induced pluripotent stem cell-derived extracellular vesicles (iPSC-EVs) have surfaced as a compelling substitute for stem cell therapies. Our study evaluates the neuroprotective effects of iPSC-EVs in a rat optic nerve crush (ONC) model. iPSC-EVs were isolated and characterized. The intravitreal (IVT) injections of iPSC-EVs or PBS were performed. RGCs survival was assessed at 7 a

Activation of Sigma 1 Receptor Preserves Visual Function After Optic Nerve Injury.

PURPOSE: Previous research has indicated that sigma 1 receptor (Sig1R) activation can help retinal ganglion cells (RGCs) survive optic nerve injury. However, in these studies, the treatments were given before the injury. We explored whether activating Sig1R as a post-injury treatment can still preserve visual function. METHODS: Wild-type or Sig1R knockout mice were subjected to optic nerve crush (ONC) injury. The contralateral eye served as sham control. Starting 6 hours after ONC, (+)-pentazocine, or (+)-PTZ, was administered daily. Visual acuity, contrast sensitivity, pattern electroretinog

Activation of the non-canonical Wnt5a signaling pathway following optic nerve injury induces time-dependent changes in pro- and anti-inflammatory gene expression.

Optic nerve (ON) injury leads to retinal ganglion cell (RGC) degeneration and axonal atrophy. Wnt ligands are embryonic growth factors that regulate cellular differentiation and survival. We recently demonstrated that canonical and non-canonical Wnt signaling induces RGC survival and axonal regrowth after optic nerve crush (ONC) injury in mouse. Here, we investigated whether the non-canonical Wnt5a ligand induces pro-regenerative inflammation after ONC. Mice were intravitreally injected with Wnt5a or saline during ONC and retina tissue was collected for QPCR and immunofluorescence. We demonstr

MicroRNA-21-5p from induced pluripotent stem cells promotes neuroprotection of retinal ganglion cells in optic nerve crush model.

INTRODUCTION: Retinal ganglion cells (RGCs) are susceptible to degenerative conditions such as glaucoma and traumatic optic neuropathies, which lead to vision loss. MicroRNA-21-5p has demonstrated potential neuroprotective effects, but its mechanisms in optic nerve injury remain underexplored. This study evaluates the neuroprotective role of microRNA-21-5p derived from induced pluripotent stem cells (iPSCs) in an optic nerve crush (ONC) model. MATERIALS AND METHODS: In vitro qPCR demonstrated that the expression of microRNA-21-5p was increased in the co-culture medium of RGCs and iPSCs. Subseq

Optimized in vivo two-photon imaging reveals the essential role of the contralateral eye in functional optic nerve regeneration in zebrafish larvae.

BACKGROUND: The visual pathway, consisting of the eye, optic nerve, and brain, serves as a valuable model for studying neural regeneration. The exceptional regenerative capacity of the zebrafish visual system enables detailed investigation of neural repair mechanisms in vivo. Although the transparency of zebrafish larvae permits real-time imaging of axonal regeneration following transection, previous methodological limitations such as pigment interference and suboptimal imaging protocols have hindered high-resolution analyses of structural recovery and cellular interaction throughout the entir

Melatonin alleviates retinal damage and visual function impairment by suppressing ROS/TXNIP/NLRP3 signalling and inhibiting microglial activation in rats with optic nerve crush injury.

This study aimed to investigate the protective mechanism of melatonin on retinal structure and optic nerve function after optic nerve crush (ONC) in a rat model. SD rats aged 6-8 weeks were used to establish an ONC model. The rats were intraperitoneally administered with melatonin daily after ONC. Loss of retinal ganglion cells (RGCs)/axons was confirmed by haematoxylin & eosin staining, optical coherence tomography, and retina flat mounts. Visual evoked potentials were measured to evaluate optic nerve function. Protein expression was determined by Western blot and immunofluorescence assays, a

MXene-chitosan photo-responsive conduit for wireless optogenetic stimulation to enhance neural regeneration and functional recovery after optic nerve injury.

Optic nerve injury triggers progressive degeneration of retinal ganglion cells (RGCs) and axonal loss, driven by inhibitory microenvironmental factors such as glial scarring, myelin debris, and growth-inhibitory signaling. Physical stimuli such as photothermal and photoelectric stimulations have gained attention, yet little is known about their potential on normal cells or the optic nerve due to setbacks from over-exposure. Photothermal stimulus presents photoelectric cues and, at the same time, energy conversion for heat generation. Herein, a bio-functional platform was designed by incorporat

Oriented porous microtubules combined with CNTF-delivery directional guide axonal regeneration after traumatic optic neuropathy.

Neuroinflammation microenvironment and retinal ganglion cell (RGC) apoptosis are two critical barriers to axonal regeneration following traumatic optic neuropathy (TON). To overcome these challenges, we developed an innovative dual drug delivery strategy utilizing oriented porous nanofiber (OF) and ciliary neurotrophic factor (CNTF)-loaded delivery systems, aiming to promote axonal regeneration and restore RGC survival. Cerium oxide nanoparticles (Ce NPs) were physically mixed with poly(L-lactic acid)/polycaprolactone (PLA/PCL) solution to prepare oriented porous nanofibers (OF-Ce) via electro

Zebrafish optic nerve injury results in systemic retinal ganglion cell dedifferentiation.

Retinal ganglion cells (RGCs) are the sole projection neurons connecting the retina to the brain and therefore play a critical role in vision. Death of RGCs during glaucoma, optic neuropathies and after ocular trauma results in irreversible loss of vision as RGCs do not regenerate in the human eye. Moreover, there are no FDA approved therapies that prevent RGC death and/or promote RGC survival in the diseased or injured eye. There is a critical need to better understand the molecular underpinnings of neuroprotection to develop effective therapeutic approaches to preserve damaged RGCs. Unlike i

Qingguang'An II decoction attenuates retinal ganglion cell pyroptosis in glaucoma via inhibition of the TRPV4/NF-κB signaling pathway.

ETHNOPHARMACOLOGICAL RELEVANCE: Glaucomatous optic neuropathy (GON), characterized by the progressive degeneration of retinal ganglion cells (RGCs), is the primary pathological basis of irreversible visual impairment in glaucoma. Qingguang'An II decoction (QGAII), a traditional Chinese medicine formula with decades of clinical application, has been shown to promote optic nerve repair and preserve the visual field in patients with GON. Nevertheless, the precise mechanisms underlying the neuroprotective effects of QGAII remain incompletely understood. AIM OF THE STUDY: This study aimed to inves

Sphenoid sinus hyperpneumatization: anatomical variants, molecular blueprints, and AI-augmented roadmaps for skull base surgery.

The sphenoid sinus is a complex part of the skull base that has a high degree of anatomical variation, the most interesting of which occurs with hyperpneumatization, in which pneumatized air cells extend beyond their normal limits into the clivus, pterygoid processes, and sphenoidal wings. These hard to note hyperpneumatized imaging variants are disregarded in routine imaging but have potential to grossly alter important neurovascular landmarks, which is a challenge for the precision and safety of transsphenoidal surgical approaches. In this review, we provide an exten- sive, state-of-the-art

Polydatin's neuroprotective mechanism in optic nerve injury: targeting mitochondrial function and glial cell activation.

Optic nerve injury (ONI) frequently causes irreversible visual impairment, making it a significant clinical challenge. This study evaluated the neuroprotective effects of polydatin (PD), focusing on its ability to preserve mitochondrial function and inhibit glial cell activation. We utilized an in vitro retina-ON explant culture model and a mouse ON crush (ONC) model. PD was administered to assess its impact on mitochondrial protection, apoptosis of retinal ganglion cells (RGCs), glial cell activation, and glial inflammatory responses. Western blot and immunofluorescence analysis were employed

Successful axonal regeneration is associated with intraneuronal metabolic reprogramming.

Unlike mammals, zebrafish can regrow axons after injury and restore circuit function in the central nervous system (CNS). Mitochondria have been identified as key players in this process, but how different metabolic pathways work together to sustain regeneration remains unclear. Using the RNA sequencing of adult zebrafish retinal ganglion cells after optic nerve crush injury, we found that oxidative phosphorylation is downregulated during axonal regrowth. Simultaneously, the thioredoxin antioxidant system was upregulated, likely to limit oxidative damage. Additionally, we observed an integrate

Spatiotemporal Immune Dynamics in Experimental Retinal Ganglion Cell Injury Models.

BACKGROUND: The damage and regeneration of retinal ganglion cells (RGCs) have been extensively studied. Among them, immune cells in different parts of the visual pathway play an important role in injury, regeneration and repair, but a comprehensive analysis of their spatial and temporal distribution is lacking. PURPOSE: This review emphasizes the unique characteristics of immune cells within the visual input pathway, focusing on their spatiotemporal dynamics in the retina, optic nerve head (ONH), and optic nerve during glaucoma and traumatic optic nerve injury. METHODS: A comprehensive searc

Alterations of Bioactive Lipid Profiles in the Retina Following Traumatic Optic Neuropathy in Mice.

Traumatic optic neuropathy (TON) causes vision loss through compression and contusion, yet there is no consensus on the most effective treatment. Polyunsaturated fatty acid (PUFA)-derived bioactive lipids metabolized by lipoxygenase (LOX), cytochrome P450 (CYP), and cyclooxygenase (COX) enzymes are known mediators of inflammation and neurodegeneration. However, their role in TON-related retinal pathology remains unclear. Controlled orbital impact (COI) was used to induce unilateral TON in mice with controlled velocity (2-3 m/s), with the fellow eye serving as an internal control. Retina tissue

Piperine alleviates retinal ischemic injury by mediating the APE1 signaling.

Ischemic retinal damage is the most common cause of severe vision impairment and blindness. Anti-vascular endothelial growth factor (anti-VEGF) agents have transformed the treatment of retinal ischemic disorders and have become the cornerstone therapy for these conditions. Nonetheless, the risk for systemic and ocular adverse effects necessitates careful consideration. Meanwhile, the therapeutic potential of natural compounds for ischemic retinal injury is increasingly attracting attention. In this study, piperine (PIP), a natural compound derived from pepper, was found to reduce apoptosis by

Alpinetin Nanoparticles Alleviate Optic Nerve Injury Induced by Acute Glaucoma via LRP1-PPARγ Mediated Regulation of Microglial Lipid Metabolism.

Glaucoma is a leading cause of irreversible blindness, characterized by progressive retinal ganglion cells (RGCs) loss. Increasing evidence links microglial activation and lipid metabolism dysregulation to neurodegeneration. However, the role of microglial lipid metabolic reprogramming in disease pathogenesis remains unclear. This study finds that microglia in an acute ocular hypertension (AOH) model exhibit abnormal lipid droplet accumulation, downregulation of low-density lipoprotein receptor-related protein 1 (LRP1), and a shift toward a pro-inflammatory M1 phenotype. Importantly, serum sam
