Characterization of optic nerve regeneration following Nd:YAG laser-induced injury in Xenopus tropicalis.

A stable, precise, and replicable amphibian model of optic atrophy would advance the study of the mechanisms underlying post-injury optic nerve (ON) regeneration. A recent study developed a transgenic Xenopus laevis model that induced ON transection via laser microdissection, enabling fluorescence microscopy-based monitoring of axon dynamics. However, no studies have used Xenopus tropicalis to investigate optic atrophy. In the current study, we developed an ON injury model in Xenopus tropicalis using an Nd:YAG laser that exhibited several histopathological features resembling traumatic optic n

Overexpression of cholesterol 24-hydroxylase CYP46A1 attenuates retinal dysfunction and ganglion cell loss via regulating the Nrf2 pathway in optic nerve crush injury.

Cholesterol 24-hydroxylase CYP46A1 (CYP46A1) has been confirmed to be correlated with the processes of multiple neurological disorders, but its role in neurodegenerative optic diseases remains unclear. This article aimed to evaluate the neuroprotective effects of CYP46A1 on mouse retinal ganglion cells (RGCs) and retinal function. Mice were subjected to optic nerve crush (ONC) injury after intravitreal injection of rAAVs. RGCs' survival was quantified by immunofluorescence staining of retinal flat mounts. Retinal electrophysiological function and visual acuity were quantitatively assessed usin

The Profile of Retinal Ganglion Cell Death and Cellular Senescence in Mice with Aging.

Older age is a risk factor for glaucoma, in which progressive retinal ganglion cell (RGC) loss leads to visual field defects and irreversible visual impairment and even blindness. We recently identified the involvement of cellular senescence in RGC cell death post-optic nerve injury. Here we further aimed to delineate the profile of RGC survival in mice with aging, a physiological process with increasing cellular senescence. The numbers of senescent cells in the ganglion cell layer (GCL) significantly and progressively increased starting at 8 months of age. Yet, significant reduction of gangli

The alternative complement pathway drives neuroinflammation and neurodegeneration in mouse models of glaucoma and optic nerve injury.

Glaucoma is a leading cause of irreversible blindness worldwide, characterized by progressive retinal ganglion cell (RGC) loss and optic nerve degeneration. Although elevated intraocular pressure (IOP) is a major risk factor, disease progression can occur despite normal IOP, highlighting the need for neuroprotective strategies beyond IOP reduction. Neuroinflammation has been implicated in glaucomatous neurodegeneration through complement system activation via the classical and lectin pathways. However, the role of the alternative pathway (AP), which functions as an amplification loop for centr

Regional Position and Axonal Environment Shape Astrocyte Morphology in the Mouse Optic Projection.

Optic neuropathies encompass a diverse group of disorders that result in damage to the optic nerve and lead to vision loss. Despite the initial site of damage often being injury specific, we have an incomplete understanding of how differential regional susceptibility arises. Recent studies have implicated astrocytes in this phenomenon in mice, showing regional transcriptional heterogeneity in optic nerve astrocytes in response to glaucoma and optic nerve crush. Because astrocytes provide structural support and protection from tissue deformation, we examined the length of the optic nerve for as

ATF3 prevents retinal ganglion cell apoptosis and mitigates microglia-mediated neuroinflammation in retinal ischemia-reperfusion injury.

INTRODUCTION: Neuroinflammation is a key pathological response involved in secondary optic nerve injury following retinal ischemia-reperfusion injury. The expression of activating transcription factor 3 (ATF3), a highly conserved protein, is rapidly induced post-injury and is crucial for regulating immunity and inflammation. The potential neuroprotective mechanisms, function, and therapeutic potential of ATF3 following retinal ischemia-reperfusion remain largely unexplored. In this study, we examined the expression and distribution of ATF3 and achieved the overexpression of ATF3 in mouse retin

A Ruptured Tri-Lobulated ICA-PCom Aneurysm Presenting with Preserved Neurological Function: Case Report and Clinical-Anatomical Analysis.

Background and Clinical Significance: Although rupture of aneurysms at the internal carotid-posterior communicating artery (ICA-PCom) junction accounts for a small percentage of all ruptured intracranial aneurysms, they are clinically relevant due to their proximity to perforator-rich cisterns, the optic-carotid-oculomotor pathways and flow-diverting zones, as well as their high likelihood for causing early neurological instability. Additionally, ruptured ICA-PCom aneurysms that have multiple lobulations are associated with increased variability in wall shear stress, local inflammatory remodel

Tumor Necrosis Factor Receptor Superfamily Member 12A Enhances Retinal Ganglion Cell Survival and Promotes Axon Regeneration.

Regeneration of retinal ganglion cells (RGCs) in adult mammals is highly limited following injury. To uncover transcriptional pathways that enhance this process, we analyzed single-cell transcriptomic profiles of RGCs in mice after optic nerve crush (ONC) and identified potential targets for neuroprotection and axonal regeneration. scRNA-seq revealed that Tumor necrosis factor receptor superfamily member 12A (Tnfrsf12a, also known as Fn14) is enriched in biological processes linked to axonal regeneration in αRGCs/ipRGCs. To validate its neuroprotective and regenerative effects, we engineered a

NLRX1 limits inflammatory neurodegeneration in the anterior visual pathway.

Chronic innate immune activation in the central nervous system (CNS) significantly contributes to neurodegeneration in progressive multiple sclerosis (MS). Using multiple experimental autoimmune encephalomyelitis (EAE) models, we discovered that NLRX1 protects neurons in the anterior visual pathway from inflammatory neurodegeneration. We quantified retinal ganglion cell (RGC) density and optic nerve axonal degeneration, gliosis, and T-cell infiltration in Nlrx1-/- and wild-type (WT) EAE mice and found increased RGC loss and axonal injury in Nlrx1-/- mice compared to WT mice in both active immu

Coordinated stimulation of axon regenerative and neurodegenerative transcriptional programs by ATF4 following optic nerve injury.

Stress signaling is important for determining the fates of neurons following axonal insults. Previously we showed that the stress-responsive kinase PERK contributes to injury-induced neurodegeneration (Larhammar et al., 2017). Here we show that PERK acts primarily through Activating Transcription Factor-4 (ATF4) to stimulate not only pro-apoptotic but also pro-regenerative responses following optic nerve damage. Using conditional knockout mice, we find an extensive PERK/ATF4-dependent transcriptional response that includes canonical ATF4 target genes and modest contributions by C/EBP Homologou

The Involvement of Unfolded Protein Response in the Mechanism of Nitrogen Mustard-Induced Ocular Toxicity.

Nitrogen mustard (NM) is a known surrogate of sulfur mustard, a chemical-warfare agent that causes a wide range of ocular symptoms, from a permanent reduction in visual acuity to blindness upon exposure. Although it has been proposed that the two blistering agents have a similar mechanism of toxicity, the mode of NM-induced cell death in ocular tissue has not been fully explored. Therefore, we hypothesized that direct ocular exposure to NM in mice leads to retinal tissue injury through chronic activation of the unfolded protein response (UPR) PERK arm in corneal cells and VEGF secretion, event

Brachial Plexus Root Avulsion Injury-Induced Endothelin-Converting Enzyme-Like 1 Overexpression Is Associated with Injured Motor Neurons Survival.

Brachial plexus root avulsion (BPRA) injury arises from challenging delivery during childbirth, sports-related incidents, or car accidents, leading to extensive loss of motor neurons (MNs) and subsequent paralysis, including both motor and sensory impairment. Surgical nerve re-implantation cannot effectively restore motor function, and the survival of injured MNs is vital for axon regeneration and re-innervating the target muscles. Therefore, identifying novel molecular targets to improve injured MNs survival is of great significance in the treatment of BPRA injuries. Endothelin-converting enz

Targeting eIF2α in TBI-induced traumatic optic neuropathy: Effects of Salubrinal and the Integrated Stress Response Inhibitor.

Traumatic brain injury (TBI) can induce traumatic axonal injury in the optic nerve, which is referred to as traumatic optic neuropathy (TON). TON occurs in up to 5% of TBI cases and leads to irreversible visual deficits. TON-induced phosphorylation of eIF2α, a downstream ER stress activator in the PERK pathway presents a potential point for therapeutic intervention. For eIF2α phosphorylation can lead to apoptosis or adaptation to stress. We hypothesized that dephosphorylation, rather than phosphorylation, of eIF2α would lead to reduced apoptosis and improved visual performance and retinal cell

Characteristics of ocular injuries associated with mortality in patients admitted with major trauma.

BACKGROUND: Few ocular trauma studies have addressed mortality outcomes. We sought to determine characteristics of mortality-related ocular trauma admissions and compared them with non-fatal injuries. METHODS: A retrospective study was conducted using de-identified data of patients admitted with major trauma from the National Trauma Data Bank (2008-2014). Patients with ocular injury were identified using ICD- 9CM codes. Demographics, intention and mechanism, types of ocular and head injuries, and injury severity were documented. Mortality was determined using post-admission disposition. Stati

Quantitative Proteomics Reveal Region-Specific Alterations in Neuroserpin-Deficient Mouse Brain and Retina: Insights into Serpini1 Function.

Neural regeneration and neuroprotection represent strategies for future management of neurodegenerative disorders such as Alzheimer's disease (AD) or glaucoma. However, the complex molecular mechanisms that are involved in neuroprotection are not clearly understood. A promising candidate that maintains neuroprotective signaling networks is neuroserpin (Serpini1), a serine protease inhibitor expressed in neurons which selectively inhibits extracellular tissue-type plasminogen activator (tPA)/plasmin and plays a neuroprotective role during ischemic brain injury. Abnormal function of this protein
