Modulating primary cilia and cilia-dependent Shh signaling increases retinal ganglion cell survival by repressing astrocyte reactivity after optic nerve injury.

The primary cilia and cilia-dependent signaling are key regulators of neurodegenerative diseases. However, their roles in glaucoma and optic nerve injury remain largely unexplored. Here, we reported that astrocyte primary cilia elongated after optic nerve injury, and genetic ablation or shortening of these cilia significantly increased retinal ganglion cell (RGC) survival. Furthermore, single-cell transcriptomic analysis identified astrocytes as the predominant cell type transducing Sonic hedgehog (Shh) signaling. Activation of canonical Shh signaling, which is transduced through primary ciliu

IL-1-mediated vitreous inflammation as an early indicator of retinal ganglion cell loss following acute optic nerve injury.

BACKGROUND: Retinal ganglion cell (RGC) degeneration in optic neuropathies is often preceded by neuroinflammatory changes, yet the earliest in vivo indicators of this process remain poorly defined. Vitreous hyperreflective foci (VHRFs) emerging within 24 h following optic nerve crush (ONC) might represent a promising early in vivo indicator of RGC loss. METHODS: VHRFs were longitudinally tracked by visible-light optical coherence tomography (vis-OCT) imaging post-ONC. Whole-eye sectioning, immunohistochemistry, and confocal imaging revealed the identity and migration of the VHRFs. RNAscope in

A novel cartilage-based reference point for the recurrent laryngeal nerve: implication for preoperative risk stratification in posterior subcapsular thyroid tumors.

BACKGROUND: Protecting the recurrent laryngeal nerve (RLN) is central in thyroidectomy, yet little is known about preoperative localization of the RLN and its role in assessing injury risk. We evaluated whether a novel cartilage-based landmark, the inferior cricoid-tracheal (ICT) point, located at the intersection of the lateral tracheal border and the inferior margin of the cricoid cartilage, approximates the laryngeal entry point (LEP) of the RLN and enables ultrasound-based measurement of tumor-nerve distance. METHODS: The study consisted of two independent cohorts. In the first cohort, th

Neuritin1 Cis-Regulatory Elements Enable Gene Expression Preferentially in Retinal Ganglion Cells.

PURPOSE: Retinal ganglion cells (RGCs) are essential for visual signal transmission, yet they are vulnerable to injury and degeneration. Gene modulation in RGCs using adeno-associated virus (AAV) offers a promising avenue for neuroprotection and regeneration, but promoters lack sufficient RGC specificity, limiting the precision needed for preclinical studies. This study aims to identify novel promoter-enhancer combinations (PECs) to achieve gene expression preferentially in RGCs. METHODS: We evaluated existing transcriptomic data to identify neuritin 1 (Nrn1) as a gene with highly restricted

HCN2 channels: a potential therapeutic target for orofacial neuropathic pain after trigeminal nerve injury.

Hyperpolarization-activated cyclic nucleotide-gated (HCN) channels have recently emerged as promising targets for the treatment of neuropathic pain. This study investigated the potential involvement of HCN2 channels in the development of trigeminal neuropathic pain following peripheral nerve injury. Infraorbital nerve chronic constriction injury (ION-CCI) model was adopted to rats, and head withdrawal thresholds (HWT) to mechanical stimulation were assessed pre- and postoperatively, as well as after pharmacological intervention. In the trigeminal ganglion (TG), intracellular cyclic adenosine m

Overexpression or Activation of Potassium Channel TASK-3 Protects Retinal Ganglion Cells and Restores Visual Function in Optic Nerve Crush.

PURPOSE: This study explored in a mouse model whether activation or upregulation of the two-pore domain potassium channel tandem pore domain acid-sensitive potassium channel 3 (TASK-3) in retinal ganglion cells (RGCs) could protect RGCs and reverse the vision loss arising through optic nerve injury. METHODS: TASK-3 activity was assessed using patch-clamp electrophysiology. The optic nerve of each mouse was crushed, and the selective TASK-3 agonist CHET3 was applied to the surface of the eye once daily for 1 week or TASK-3 was overexpressed specifically in RGCs through infection with recombina

Experimental upregulation of Lancl1 promotes axon regeneration after optic nerve injury in vivo.

Lanthionine synthetase C-like protein (Lancl) gene family members 1-3 play roles in responses to oxidative stress and cellular metabolic processes. Lancl1 in particular functions as a neuroprotective factor in the central nervous system (CNS), but it is unknown whether experimentally upregulating Lancl1 in neurons could also repair injured circuits by regenerating damaged axons. We recently identified Lancl1 as a gene upregulated in response to an axon-regenerating treatment, which prompted us to ask here if Lancl1 could directly repair injured axons. First, we characterized expression of Lanc

Melatonin in Peripheral Nerve Regeneration: Molecular Mechanisms, Schwann Cell Dynamics, and Translational Therapeutic Potential.

Peripheral nerve injury (PNI) presents a significant clinical challenge often resulting in long-term functional disability. Following injury, successful regeneration heavily depends on the cellular plasticity of Schwann cells (SCs) to undergo dedifferentiation, proliferate, and guide axonal growth. Recently, the pineal hormone melatonin has emerged as a promising therapeutic candidate due to its potent antioxidant properties and ability to modulate SC biological behavior. However, its exact molecular mechanisms and direct impact on SC proliferation remain fragmented across literature. Followin

Connector-Assisted Repair of Transected Inferior Alveolar and Lingual Nerves Using Photochemical Tissue Bonding.

BACKGROUND: Inferior alveolar nerve (IAN) and lingual nerve (LN) injury are recognized complications of oral and maxillofacial trauma or surgical procedures, often requiring subsequent microsurgical repair. This study explores the potential of photochemical tissue bonding (PTB), a light-activated technique that forms molecular crosslinks between collagen molecules, to seal a commercial nerve connector in place for IAN and LN nerve repair. PURPOSE: The purpose of this study was to measure and compare the repair strength of conventional suture and PTB repair in a nerve transection model. STUDY

Age and Risk of Inferior Alveolar Nerve Injury After Removal of Lower Third Molars Close to the Mandibular Nerve.

BACKGROUND: Extraction of lower third molars (3 Ms) carries a risk of inferior alveolar nerve (IAN) injury. Anatomical proximity is a well-known risk factor, although the effect of patient age remains insufficiently explored. PURPOSE: The study purpose was to measure the association between age and IAN injury among patients whose 3 Ms are close to the mandibular canal. STUDY DESIGN, SETTING, SAMPLE: This case-control study included patients at increased risk for IAN injury due to close proximity of the lower 3 Ms to the mandibular canal, as assessed by preoperative cone-beam computed tomogra
