Structural conservation of ligand binding reveals a bile acid-like signaling pathway in nematodes.

Bile acid-like molecules named dafachronic acids (DAs) control the dauer formation program in Caenorhabditis elegans through the nuclear receptor DAF-12. This mechanism is conserved in parasitic nematodes to regulate their dauer-like infective larval stage, and as such, the DAF-12 ligand binding domain has been identified as an important therapeutic target in human parasitic hookworm species that infect more than 600 million people worldwide. Here, we report two x-ray crystal structures of the hookworm Ancylostoma ceylanicum DAF-12 ligand binding domain in complex with DA and cholestenoic acid

In vitro and in vivo efficacy of tribendimidine and its metabolites alone and in combination against the hookworms Heligmosomoides bakeri and Ancylostoma ceylanicum.

Worldwide, 3 billion people are at risk of hookworm infection, particularly in resource-poor countries. While control of soil-transmitted helminthiases relies mostly on chemotherapy, only few drugs are available and concern about potential emergence of drug resistance is rising. In the present study, tribendimidine, a derivative of amidantel, and its metabolites deacylated amidantel (dADT) and acetylated deacylated amidantel (AdADT) were tested in vitro and in vivo against Heligmosomoides bakeri and Ancylostoma ceylanicum, two hookworm rodent models, alone or in combination with standard drugs

The mucosal response to secondary infection with Ancylostoma ceylanicum in hamsters immunized by abbreviated primary infection.

We assessed the mucosal response of previously infected hamsters to low-dose challenge with the hookworm, Ancylostoma ceylanicum. Hamsters were assigned to five treatment groups (Groups 1-5, respectively): naïve, controls; uninterrupted primary infection from day 0; infected, but treated with anthelmintic on day 35 p.i.; challenge control group given only the second infection on day 63; infected initially, cleared of worms and then challenged. Animals were culled on days 73 and 94 (10 and 31 days after challenge), but additional animals were culled from Group 5 on days 80 and 87. The results s

Hookworms of dogs and cats as agents of cutaneous larva migrans.

Dogs and cats are hosts to hookworms that may cause zoonotic disease, most notably, cutaneous larva migrans. Ancylostoma braziliense is most often implicated in dermatological lesions, and Ancylostoma caninum has been associated with eosinophilic enteritis and suggested as a possible cause of diffuse unilateral subacute neuroretinitis in humans. Other manifestations include eosinophilic pneumonitis, localized myositis, folliculitis, erythema multiforme, or ophthalmological manifestations. Ancylostoma eggs are morphologically indistinguishable, which complicates epidemiological studies. Surveys

[Prevalence and factors associated with geohelminth infections in children living in municipalities with low HDI in North and Northeast Brazil].

Stool surveys were conducted to estimate the prevalence and identify risk factors for geohelminth infections among children in ten Brazilian municipalities with low human development indices (HDI). Socioeconomic and environmental data were obtained from the children's parents or guardians, and stool samples were examined. The proportion of geohelminth infections according to target variables was calculated. Risk factors were evaluated using multilevel logistic regression. Of the 2,523 children, 36.5% were infected with one or more geohelminths (Ascaris lumbricoides, 25.1%; hookworm, 15.3%; Tri

Infantile hookworm disease.

Hookworm infection is common but has rarely been reported in neonates or infants. Two cases of hookworm infestation in early infancy are described. The infants presented with malena, severe pallor, lethargy and failure to gain weight. Initial stool examination was non-contributory and diagnosis was made by upper gastrointestinal endoscopy.

Experimental infection with Ancylostoma ceylanicum in dogs and efficacy of a spot on combination containing imidacloprid 10% and moxidectin 2.5% (Advocate/Advantage Multi, Bayer Animal Health).

Ancylostoma ceylanicum is a common hookworm of dogs, cats and humans in Asia. More recently, this hookworm was found to infect dogs in Australia. The objective of this study was to determine the efficacy of a spot on combination product containing imidacloprid 10% and moxidectin 2.5% (Advocate/Advantage Multi, Bayer Animal Health) against A. ceylanicum in experimentally infected dogs. Twelve dogs were each subcutaneously injected with 300 infective third-stage larvae of A. ceylanicum. Pups were stratified by egg count and randomly allocated equally into control and treatment groups. The pups i

CD4 T cells mediate mucosal and systemic immune responses to experimental hookworm infection.

Hookworm infection is associated with anaemia and malnutrition in many resource-limited countries. Ancylostoma hookworms have previously been shown to modulate host cellular immune responses through multiple mechanisms, including reduced mitogen-mediated lymphocyte proliferation, impaired antigen presentation/processing, and relative reductions in CD4(+) T cells in the spleen and mesenteric lymph nodes. Syrian hamsters were depleted of CD4(+) for up to 9 days following intraperitoneal injection (200 microg) of a murine anti-mouse CD4 monoclonal IgG (clone GK1.5). CD4(+) T-cell-depleted hamster

In vitro predatory activity of the fungi Duddingtonia flagrans, Monacrosporium thaumasium, Monacrosporium sinense and Arthrobotrys robusta on Ancylostoma ceylanicum third-stage larvae.

The potential role of companion animals as reservoirs for zoonotic diseases has been recognised as a significant public health problem worldwide. Ancylostoma ceylanicum is the only ancylostomatidae species known for infecting human beings. This article aimed to compare the predatory capacity of predatory fungi isolates Duddingtonia flagrans (AC001), Monacrosporium thaumasium (NF34), Monacrosporium sinense (SF53) and Arthrobotrys robusta (I31) on A. ceylanicum infectious larvae (L(3)) in a 2% water-agar plate. There was no predatory capacity variation among the fungi tested (P>0.05) over the 7-

Activation of Nippostrongylus brasiliensis infective larvae is regulated by a pathway distinct from the hookworm Ancylostoma caninum.

Developmentally arrested infective larvae of strongylid nematodes are activated to resume growth by host-derived cues encountered during invasion of the mammalian host. Exposure of Nippostrongylus brasiliensis infective larvae to elevated temperature (37°C) is sufficient to activate signalling pathways which result in resumption of feeding and protein secretion. This occurs independently of exposure to serum or glutathione, in contrast to the hookworm Ancylostoma caninum, and is not initiated by chemical exsheathment. No qualitative differences in protein secretion were induced by host serum a

The biological control of Ancylostoma spp. dog infective larvae by Duddingtonia flagrans in a soil microcosm.

Experiments to evaluate the potential ability of the nematode-trapping fungus Duddingtonia flagrans (Isolate CG768) to prey on the Ancylostoma spp. dog infective larvae (L(3)) in pasteurized soil were performed through several laboratory assays. A microcosm approach was used with increasing fungal concentrations in an inoculum of a chlamydospore water suspension. The highest fungal concentrations provide a more consistent larval reduction than the lowest concentrations, but no difference was observed from 10,000 to 25,000 chlamydospores per grain of soil. When using D. flagrans in a water susp

Characterisation of hookworm heat shock factor binding protein (HSB-1) during heat shock and larval activation.

When hookworm infective L3s infect their mammalian host, they undergo a temperature shift from that of the ambient environment to that of their endothermic host. Additionally, L3s living in the environment can be exposed to temperature extremes associated with weather fluctuations. The heat shock response (HSR) is a conserved response to heat shock and other stress that involves the expression of protective heat shock proteins (HSPs). The HSR is controlled by heat shock factor-1 (HSF-1), a conserved transcription factor that binds to a heat shock element in the promoter of HSPs, causing their

Helminthic infections in school children in Puducherry, South India.

BACKGROUND/PURPOSE: Intestinal parasites are widely prevalent in developing countries, probably due to poor sanitation and inadequate personal hygiene. It is estimated that as much as 60% of the world's population is infected with intestinal parasites, which may play a significant role in morbidity due to intestinal infections. This study was designed to estimate the prevalence of helminthic infections among school children in Puducherry, South India. METHODS: The prevalence of helminthic infections was studied in 1,172 children in public schools in Puducherry (Pondicherry), South India, betw
