Stability of salivary microRNA measures across an NCAA Division I football season: Implications for microRNA as a biomarker of concussion.

BACKGROUND: Clinicians often face challenges in concussion care due to a heavy reliance on subjective patient input. Recently, research has sought objective biomarkers, like salivary microRNAs, to improve concussion management. However, significant limitations hinder the use of microRNAs as a diagnostic tool, including the cumulative effects of a contact sport season. A better understanding of the response to a contact sport season would help researchers and clinicians interpret expression changes at the time of injury in the context of seasonal variation. in Therefore, this study investigated

A longitudinal study of error-related neural responses in adolescents following mild traumatic brain injury (mTBI).

Error processing, a neural process critical for adaptive learning, may be disrupted by mild traumatic brain injury (mTBI), also known as concussion. Both cross-sectional and longitudinal studies of adults following mTBI indicate a variable impact on neural correlates of error processing, including the error related negativity (ERN) and post error positivity (Pe) scalp-recorded event-related potential (ERP) components. A similar study of adolescents indicated smaller ERN and Pe amplitudes in those with mTBI compared to healthy control participants. To date, no longitudinal studies measuring the

IV dexamethasone as adjuvant therapy to metoclopramide for acute posttraumatic headache in the ED: A randomized controlled trial.

OBJECTIVE: We conducted a randomized study to determine if, among emergency department (ED) patients with acute posttraumatic headache, the combination of intravenous (IV) metoclopramide plus dexamethasone would result in less headache intensity during the 48 h after ED discharge than IV metoclopramide plus placebo. BACKGROUND: Intravenous metoclopramide can improve acute posttraumatic headache among ED patients, though this benefit is not sustained beyond the ED visit. METHODS: This was a randomized, double-blind, placebo-controlled, parallel group study of IV dexamethasone for acute posttr

CCL5/RANTES signaling in inflammation dysregulation after mild traumatic brain injury.

BACKGROUND: Mild traumatic brain injury (mTBI) is the most prevalent form of brain injury. Secondary damage following mTBI contributes to neuronal degeneration by promoting neuroinflammation, amyloid accumulation, and oxidative stress (OS). Microglia exhibit dual roles after injury, contributing to both pro-inflammatory (M1) and anti-inflammation/neuroprotective (M2) responses. Targeting microglial polarization may therefore represent a therapeutic strategy for mitigating secondary damage after TBI. METHODS: A weight-drop mTBI model (30 g, 100 cm) was applied to both C57BL/6 (wild-type) and C

N-Stearidonoylethanolamine Restores CA1 Synaptic Integrity and Reduces Astrocytic Reactivity After Mild Traumatic Brain Injury.

Mild traumatic brain injury (mTBI) disrupts hippocampal network function through coordinated alterations in glial reactivity, synaptic integrity, and adult neurogenesis. Effective therapeutic approaches targeting these interconnected processes remain limited. Lipid-derived molecules capable of modulating these mTBI-induced disturbances are emerging as promising neuroprotective candidates. Here, we investigated the effects of N-stearidonylethanolamine (SDEA), an ω-3 ethanolamide, in a mouse model of mTBI. SDEA treatment attenuated astrocytic reactivity, restored Arc expression, and improved den

Injury-environment interaction: microglial priming by mTBI creates vulnerability to subsequent stress via the HMGB1-RAGE axis.

BACKGROUND: The mechanisms underlying the increased risk of depression following mild traumatic brain injury (mTBI) remain poorly understood, particularly the synergistic interaction between the initial injury and subsequent environmental stress. This study aims to elucidate the molecular cascade governing this "injury-stress" synergy. METHODS: We developed a "two-hit" mouse model combining mTBI with chronic unpredictable mild stress (CUMS) to investigate their interaction. Our approach integrated behavioral testing with molecular, immunohistochemical, and targeted genetic manipulations (AAV-

Knowledge and Attitudes of Youth Ice Hockey Players and Their Parents Toward Concussion in the United Kingdom.

Introduction and aim There is a lack of research investigating the knowledge and attitudes of youth ice hockey players and their parents toward concussion in the United Kingdom. This is a topic of increasing concern given the well-known risks associated with contact sports. Previous research in this area has primarily come from North America. The primary objective of our research was to evaluate the knowledge and attitudes regarding concussion among youth ice hockey players and their parents in the United Kingdom. The secondary objective was to examine whether increased knowledge was associate

The Role of Nutritional Supplements in the Treatment of Sport-Related Concussion.

There is ongoing interest in the use of nutritional supplements for the prevention and treatment of sports-related concussions. While many athletes and clinicians incorporate vitamins and supplements into concussion recovery plans, human-based studies specific to sports concussion remain limited. This article reviews the most current evidence regarding supplements with emerging data in concussion management, including docosahexaenoic acid and fish oil, branched-chain amino acids, melatonin, magnesium, and riboflavin. Other supplements such as probiotics, vitamins C, D, and E, and taurine are d

Evaluation of Neck Strength and Concussion Risk: A Systematic Review.

Concussions are a major concern in sports, particularly contact sports. Neck strength may be a protective factor in limiting concussion risk by reducing head acceleration. This meta-analysis aimed to determine whether an increase in neck strength reduced concussion risk in athletes. Articles were selected based on determined inclusion and exclusion criteria. Five articles met the criteria and were included in our analysis. Standardized mean differences and 95% confidence intervals were calculated for each and visualized using a forest plot. Across five planes of motion (flexion, extension, rig

Customized Face Guards as Protective Devices for Ocular Trauma in Futsal Players: A Case Report and Clinical Efficacy Evaluation.

We present a case involving a futsal player with brain concussions and commotio retinae to protect the affected area and support a safe, prompt return to play custom face guard instead of a commercial face guard and sports goggles. For a better fit, we fabricated a custom face guard, for which a facial impression was taken using dental alginate material and plaster in July 2019. The following month, the custom face guard was set on the player's face so that its outline provided minimal coverage of the nose area compared with a conventional face guard. The field of view during play and the guar

Bidirectional Relationship Between Mental Health and Sports Injury in Adolescents: A Systematic Review and Meta-analysis.

BACKGROUND: Sports injuries are linked to negative impacts on mental health and well-being, including depression, anxiety, stress and lower quality of life. Conversely, poor mental health and well-being have been found to increase the risk of sports injury, injury severity and time taken to recover. Although existing research indicates these associations in athletes broadly, the nature and directionality of these relationships among adolescents are not well characterised. A related limitation is that much of the existing evidence, in both children and adult athletes, is cross-sectional in desi

C-DIR: Double Inversion Recovery with Controlled Artifact Suppression in Brain MRI.

BACKGROUND AND PURPOSE: Double inversion recovery (DIR) is an MRI technique in which 2 types of tissue are suppressed, usually CSF and WM. The suppression is achieved with 2 inversion pulses before the acquisition of the imaging data. In the presence of strong inhomogeneities in the static magnetic field B0 and/or the radiofrequency (RF) field, inversion may be inadequate, resulting in bright signal in tissues that should have been suppressed. The purpose of this work was to develop a DIR scan with inversion pulses that are robust against inhomogeneities in the B0 and RF field. MATERIALS AND

A Novel NLRP3 Inhibitor AMS-17 Rescues Deficits in Long-Term Potentiation Following Mild Traumatic Brain Injury in Adult C57Bl/6 Mice.

Traumatic brain injury (TBI) is a leading cause of long-term disability, with limited effective treatment options. A key factor of TBI pathophysiology is neuroinflammation, which can involve the activation of the nucleotide-binding domain leucine-rich repeat protein 3 (NLRP3) inflammasome. Aberrant inflammation following injury has the ability to reduce the capacity to induce long-term changes in synaptic plasticity, a leading mechanism for the development of learning and memory deficits following injury. This study investigated the potential of a novel NLRP3 inhibitor, AMS-17, to mitigate syn

miR-26b-5p responds to aerobic exercise intervention for concussion recovery in adolescent athletes: a pilot trial.

BACKGROUND: Guidelines for sport-related concussion (SRC) management recommend aerobic exercise treatment. Salivary microRNAs (miRNAs) associate with exercise and concussion; therefore, we investigated their expression in adolescents with SRC during prescribed exercise. METHODS: Before and after intervention, athletes with SRC (n = 32, 15.78 ± 1.5 y/o, 66% male, 5.26 ± 3.56 days from injury) and controls (n = 28, 15.96 ± 1.7 y/o, 68% male) provided saliva samples and completed clinical exams. High (5 days/week, 30 min) or low (3 days/week, 20 min) volumes of aerobic exercise were assigned. RT
