Induction of c-fos mRNA and protein in neurons and glia after traumatic brain injury: pharmacological characterization.

Focal brain injury in mice induced c-fos mRNA and protein in neurons throughout the damaged neocortex, including the piriform and the entorhinal cortices, as well as in nonneural brain cells (e.g., glia, pia, ependyma). The pattern of c-fos induction after injury suggested that injury led to spreading depression which then led to c-fos induction in neurons. Human neurons in the temporal cortex and hippocampus also showed c-fos protein induction after neurosurgical trauma. The c-fos mRNA and protein induction in mouse neurons was prevented by the noncompetitive NMDA antagonist ketamine but only

The effects of aortic crossclamping and resuscitation on intracranial pressure, cerebral blood flow, and cerebral water content in a model of focal brain injury and hemorrhagic shock.

Aortic crossclamping (AOXC) is performed frequently in hypotensive trauma patients who may have had a head injury. The effect of AOXC on the injured brain is unknown. We studied the effect of AOXC on mean arterial pressure (MAP), intracranial pressure (ICP), cerebral blood flow (CBF), cerebral perfusion pressure (CPP), and cerebral water content in a porcine model of focal cryogenic brain injury. Four groups of animals were studied: Group I--brain injury only; Group II--brain injury and AOXC; Group III--brain injury with hemorrhage and AOXC; and Group IV--AOXC only. Focal cryogenic grain injur

Platelet-activating factor and progressive brain damage following focal brain injury.

The effects of a platelet-activating factor (PAF) antagonist on brain edema, cortical microcirculation, blood-brain barrier (BBB) disruption, and neuronal death following focal brain injury are reported. A neodymium:yttrium-aluminum-garnet (Nd:YAG) laser was used to induce highly reproducible focal cortical lesions in anesthetized rats. Secondary brain damage in this model was characterized by progressive cortical hypoperfusion, edema, and BBB disruption in the vicinity of the hemispheroid lesion occurring acutely after injury. The histopathological evolution was followed for up to 4 days. Neu

Induction of Fos in glia-like cells after focal brain injury but not during wallerian degeneration.

Focal brain injury or perforant-path transections respectively led to an increase in the number of glial-fibrillary acidic protein (GFAP)-immunopositive astrocytes around the focal wound or in the terminal fields of the perforant path in the dentate molecular layer. This GFAP accumulation occurred 48-72 h after focal brain injury or perforant-path transection (wallerian degeneration). Focal brain injury also led to an accumulation of c-fos protein (Fos) in glial cells, ependyma and cells in the pia mater of the brain within 6 h of injury and this effect dissipated within 72 h. However, perfora

Growth factor expression after stroke.

Fibroblast growth factors are polypeptides with potent trophic effects on central nervous system cells. Both acidic and basic forms of fibroblast growth factor are found in the mammalian brain. We have examined the expression of these factors after focal brain injury or stroke. After infarction of the lateral cerebral cortex in the mature rat brain, we found a twofold to threefold increase during the first 3 weeks after stroke in levels of fibroblast growth factors in tissue surrounding infarcts. This increase persisted for at least 2 months and appeared mainly to be due to increased levels of

Presence and induction of Fos B-like immunoreactivity in neural, but not non-neural, cells in adult rat brain.

The presence and induction of Fos B, a novel growth factor-activated gene, was investigated in adult rat brain using an antiserum to Fos B and immunocytochemical methods. In normal rat brain immunoreactivity was detected in the nuclei of nerve cells scattered in the cerebral cortex, striatum, amygdala, hippocampus and dentate gyrus. This immunostaining was not present in adjacent brain sections incubated with anti-Fos B serum preadsorbed with the Fos B peptide. Furthermore, Fos B-like immunoreactivity was induced in neurons by two treatments (focal brain injury and haloperidol injection) that

Traumatic shock and head injury: effects of fluid resuscitation on the brain.

UNLABELLED: The effects of resuscitation of traumatic-hemorrhagic shock on the brain are unknown. Traumatic shock in sheep (fracture/crush injury, 2-hr hemorrhage to 40 mm Hg) was followed by resuscitation to baseline mean arterial pressure. Two groups without brain injury were resuscitated with lactated Ringer's (LR1, n = 7) or albumin (ALB1, n = 6). Focal brain injury was added in two further groups (LR2, n = 6; ALB2, n = 6). Hemodynamics, intracranial pressure (ICP), EEG, and colloid osmotic pressure (COP) were followed. Brain water (BW) and cerebral blood volume (CBV) were compared to thos

Effects of neonatal hemispherectomy on location and number of corticospinal neurons in the rat.

Corticospinal neurons in the rat normally project to the contralateral spinal cord only. After neonatal hemispherectomy, the remaining single hemisphere forms both ipsilateral and contralateral projections. The distribution and number of corticospinal neurons projecting to the contralateral and to the ipsilateral spinal cord were studied in the hemispherectomized rat. The method used was that of retrograde labelling of corticospinal neurons by unilateral spinal cord injection of horseradish peroxidase conjugated to wheat germ agglutinin (WGA-HRP). The establishment of both crossed and uncrosse

Some correlates of intra- and interhemispheric speech organization after left focal brain injury.

Determinants of inter- and intrahemispheric organization following left focal injury were examined with data deriving from four different studies that used three different assessment methods (amobarbital, temporal lobectomy, dichotic listening). With regard to interhemispheric reorganization, the results revealed that the earlier the lesion onset, the higher the probability of a biomodal hemispheric reorganization (speech and hand). A unimodal reorganization (speech only) was tied to a later occurring lesion, but one before age six. Furthermore, interhemispheric speech reorganization was assoc

Increased basic fibroblast growth factor (bFGF) immunoreactivity at the site of focal brain wounds.

Basic fibroblast growth factor (bFGF) is a polypeptide found within the CNS with potent effects on the survival and proliferation of CNS glia and endothelial cells, and on the survival and outgrowth of CNS neurons. Immunohistochemical methods were used to examine relative changes in the levels and distribution of bFGF following focal brain injury. Two monospecific antisera to bFGF were used to immunostain intact mature rat brain, and brain in which a focal mechanical lesion had been made in the dorsolateral cerebral cortex one week previously. In the intact brain, staining was localized primar

Mania after brain injury. A controlled study of causative factors.

Eleven patients who developed manic syndromes after brain injury (secondary mania) were studied. Six patients had depressive episodes before mania and five had a definite or possible family history of affective disorder. Eight had lesions involving limbic areas, and nine had right hemisphere involvement. In addition to focal brain injury, mean values for bifrontal and third ventricle/brain ratios of manic patients were significantly increased when compared with non-manic patients who had lesions matched for cause, location, volume, and time since injury. Results indicate that the confluence of

Paroxysmal kinesigenic choreoathetosis in hyperthyroidism.

Paroxysmal kinesigenic choreoathetosis is an unusual movement disorder often triggered by attempts to use the limbs, and has sometimes been associated with diffuse or focal brain injury. We report its occurrence in hyperthyroidism, with which choreoathetosis has rarely been described in the past without known cause. Choreoathetosis has also occurred with other metabolic and toxic disorders, and the mechanism is uncertain. The development of involuntary movements activated by limb motion during hyperthyroidism suggests an influence by thyroid hormone or metabolic state on motor neurone excitabi

Head and neck injuries.

A survey of cranial, intracranial and neck injuries caused by blunt trauma to the head is presented. The different types of fractures to the skull (impression fractures, linear fractures and ring fractures) are discussed as well as the injury mechanisms. Brain injuries are discussed under the heading of focal brain injury (i.e. brain laceration, cortical contusion, hemorrhages of the meninges and traumatic intracerebral hemorrhages). The different types of neck injuries, including atlanto-occipital dislocation, are listed and the stability of the different fractures are discussed. Facial injur

Mechanisms of lateralized hyperactivity following focal brain injury in the rat.

Recent work in this lab which explores the differential behavioral and neurochemical changes following left versus right cerebral damage is reviewed and neural mechanisms which may account for this asymmetry are proposed. Damage to the right frontoparietal cortex in rats, caused by either ischemia or suction, produces hyperactivity for as long as a month after surgery. These lesions also produce decreases in norepinephrine (NE) levels in both ipsilateral and contralateral cortex and in the locus coeruleus. Similar lesions in the left cortex, however, do not produce these behavioral or biochemi

Hypertension after brain injury: case report.

Hypertension after brain injury requires comprehensive evaluation and management. Focal brain injury to centers of blood pressure regulation, high levels of circulating catecholamines from generalized trauma or intracranial lesions, increased intracranial pressure, pheochromocytomas unmasked after trauma, and occult spinal cord injury with hyperreflexia represent possible causes of hypertension after brain injury. This case of a brain-injured patient who had episodes of hypertension and diaphoresis with catecholamine elevations in plasma and urine, and evidence of hypothalamic-pituitary dysfun

[Electromyographic studies of plantar-mental reflex].

The authors investigated the electromyographic response to mechanical stimulation of the palmar skin (palmo-mental reflex) and plantar skin (planto-mental reflex) in the mentalis muscle. The palmo-mental reflex was present in EMG in all 22 healthy subjects an in 51 cases of 51 cases of limited cerebral lesions. The planto-mental reflex was absent in healthy subjects, but it was found in 14 out of 31 cases of focal brain injury and in 2 out of 20 cases with diffuse lesions. The planto-mental reflex is a pathological reflex appearing mainly in severe focal cerebral lesions. No description of thi

An injury-induced diffuse slow potential from brain.

Three different slow potential (SP) changes resulting from focal brain injury are described. The first is an immediate, high amplitude (in excess of 25 mV)negative shift at the site of injury. The second is a biphasic negative-positive SP wave which spreads throughout the cortex ipsilateral to injury and is similar to spreading depression (SD). The third SP change, called here the injury-induced diffuse slow potential (IDSP)is a prolonged (lasting approx. 2h) negative shift occurring simultaneously in many brain areas, also in those far removed from the injured focus. The SD can be separated f
