Evaluation of different amplification targets for the detection of Necator americanus by PCR in stool samples.

Hookworm infection, caused by Necator americanus and Ancylostoma duodenale, is prevalent in areas with poor sanitation. Coprology is the gold standard for diagnosis, but it has low sensitivity in low parasite loads. The objective of this study was to evaluate four Polymerase Chain Reaction (PCR) targets for molecular diagnosis. Stool samples were collected from 180 residents of the rural community Tierra Blanca, Bolívar State, Venezuela, in which coproparasitological tests were performed. Four DNA extraction methods were standardized, and four PCR techniques were studied: NaITS1F/NaITS1R (ITS1

Comparative Effect of Water-Diluted Stool on the Growth and Development of Rhabditis sp., Strongyloides stercoralis, and Hookworm.

BACKGROUND: Raw vegetables in southern Thailand frequently carry Rhabditis sp. Epidemiological shifts (2008-2023) show an increase in Rhabditis sp. colonization alongside declining hookworm and Strongyloides stercoralis infections, suggesting a potential association with climate variations. To evaluate moisture effects under controlled conditions, we compared nematode egg and first-stage larval development using an agar plate culture (APC) control and a 1:20 water-diluted stool suspension (SS) simulating wet environments. METHODS: Stool samples positive for Rhabditis sp. from 18 humans (12 co

Hookworm and Strongyloides stercoralis infections in school-age children and cohabiting dogs in Cambodia following 5 years of bi-annual mass drug administration: a cross-sectional observational study.

BACKGROUND: Despite sustained control efforts, soil-transmitted helminths remain a major public health burden in endemic regions, with companion animals, particularly dogs, acting as potential reservoirs further complicating elimination efforts. In this study we aimed to assess infection levels in school-age children following 5 years of bi-annual school-based deworming and to investigate the contribution of dogs to ongoing zoonotic transmission in rural Cambodia. METHODS: A cross-sectional observational study was conducted across 14 villages and five primary schools in Ta Ches commune, Kampo

Wrist Ganglion Cyst Localization in Relation to Cyst Size and Associated Injuries: An MRI-Based Retrospective Study.

BACKGROUND: Three siblings with goitre and latent to mild hypothyroidism were suspected of having thyroid peroxidase (TPO) abnormality. Direct sequencing of their genomic DNAs showed two novel mutations of the TPO gene, one of which was G1687T (Gly533Cys; exon 9) and the other 1808-13del (Asp574/Leu575del; exon 10). The two mutations were compound heterozygous, as the former was found in their father's DNA as heterozygous, and the latter was found in DNA from their mother, also as heterozygous. As Gly533 and Asp574/Leu575 were well-conserved amino acids in the peroxidase superfamily, Gly533Cys

Microbiome responses to anthelmintic treatment depend on pre-treatment helminth infection status in young Ethiopian children.

BACKGROUND: Mass deworming programs using anthelmintic drugs are a common public health strategy for controlling soil-transmitted helminth (STH) infections in high-risk, low-income countries. However, whether routine deworming induces long-term changes in gut microbiome composition remains unclear. We used longitudinal data to examine how anthelmintics alter the gut microbiome profiles of Ethiopian children. METHODS: Fecal samples were collected from school-aged children enrolled in a mass deworming program in Jimma, Ethiopia, before and after receiving a 400 mg dose of albendazole. Baseline

Placental Adaptations Under Severe Anemia: Neonatal Outcomes at the Crossroads.

Anemia is a major global health problem, particularly in developing countries such as India, due to the greater prevalence of dietary iron deficiency, hemoglobinopathies, macronutrient deficiencies, and infections such as malaria, human immunodeficiency virus (HIV), and hookworm infestation. Anemia-induced hypoxia triggers a range of vascular and structural changes in the placenta, potentially leading to complications for both the mother and the fetus. Studies have shown that placentae from anemic mothers often exhibit hypertrophy, characterized by increased placental weight relative to fetal

Exploratory study characterizing gastrointestinal physiological changes during controlled human hookworm infection.

Hookworm infects over 400 million people globally and causes gastrointestinal morbidity, yet its physiological effects remain poorly defined. Controlled human hookworm infection is also being explored as a therapy for gut diseases. We performed an exploratory study to evaluate the impact of experimental Necator americanus infection on gastrointestinal transit, motility, and luminal pH in 10 healthy adults (mean age 41 yr, 60% females) infected with 30 larvae via skin application. Assessments using the SmartPill Wireless Motility Capsule were performed at baseline, week 6 (acute infection), and

Ancylostomatidae in wild canids and felids from Romania: new host associations and haplotype diversity.

BACKGROUND: Hookworms (Ancylostomatidae) significantly impact on the health of both domestic animals and humans worldwide, with some species capable of causing zoonotic diseases. While hookworm infections in pets are frequently reported in Europe primarily through coproscopic studies, there are limited data regarding their presence in wild carnivores. To address this, this study aimed to assess the diversity, prevalence, and distribution of hookworms in wild canids and felids from Romania through both morphological and molecular analyses. METHODS: From November 2011 to February 2025, 319 carc

Comparative Survival of Hookworm Eggs and Strongyloides stercoralis First-Stage Larvae in Sludge From Septic Tanks.

BACKGROUND: This study compares the survival of hookworm eggs and Strongyloides stercoralis first-stage larvae (L1) in septic tank sludge. Laboratory evidence suggests these helminths differ in survivability, but real-world data are lacking. METHODS: We enrolled 24 households, each with at least one member positive for S. stercoralis and at least one member positive for hookworm. Each member used their own household latrine for defecation. We collected stool samples from 112 individuals, followed by sludge sampling from 24 septic tanks 2 weeks later. Stool samples were examined using agar pla

An artificial intelligence-powered digital pathology platform to support large-scale deworming programs against soil-transmitted helminthiasis and intestinal schistosomiasis in resource-limited settings.

BACKGROUND: The World Health Organization (WHO) has emphasised the need for innovative diagnostic tools to support the control and elimination of neglected tropical diseases (NTDs). Microscopy-based diagnostics, the current standard, rely on trained technicians for labour-intensive processes, posing logistical challenges in the low-resource settings where NTDs are most prevalent. This study describes the technical details of an artificial intelligence-powered digital pathology (AI-DP) platform designed to support large-scale deworming programs for two NTDs, alongside its analytical performance

Na-GST-1 adsorbed on Alhydrogel co-administered with different Toll-like receptor agonists in hookworm-naive adults using a controlled human infection model in the USA: a phase 2, double-blind, randomised controlled trial.

BACKGROUND: Necator americanus glutathione S-transferase-1 (Na-GST-1) performs a crucial enzymatic step in hookworm blood feeding and is a leading target for vaccine development. Phase 1 trials in Brazil, Gabon, and the USA of recombinant Na-GST-1 adsorbed on Alhydrogel (Na-GST-1/Al), co-administered with or without AP 10-701 (Toll-like receptor [TLR] 4 agonist) or CpG 10104 (TLR9 agonist), showed the vaccine was well tolerated, safe, and immunogenic. A controlled human hookworm infection (CHHI) model was previously developed to evaluate the effect of candidate hookworm vaccines on infection.

The detection of three independent benzimidazole resistance mutations in a single Ancylostoma caninum population from a Golden Retriever breeding kennel in Sao Paulo, Brazil.

Multiple anthelmintic drug resistance (MADR) is widespread in the canine hookworm Ancylostoma caninum in kenneled greyhounds in the USA and Australia with benzimidazole resistance being associated with the isotype-1 β-tubulin SNPs Q134H(CAA > CAT) and F167Y(TTC > TAC) in both cases. These two SNPs have also been found to be widespread, often at high frequency, in A. caninum populations in pet dogs across the USA but the situation in domestic dogs in other countries is currently unknown. Here, we report the identification of a single A. caninum population, from a Golden Retriever breeding kenne

Endoscopic Detection and Molecular Confirmation of Ancylostoma ceylanicum with Atypical Gastrointestinal Manifestations: A Case Series.

BACKGROUND: Hookworm is a widely known soil-transmitted helminth (STH) traditionally linked to iron-deficiency anemia caused by chronic blood loss in the small intestine. The classical human pathogens are Ancylostoma duodenale and Necator americanus. However, emerging evidence indicates that Ancylostoma ceylanicum, a hookworm of cats and dogs, is a significant cause of human infection, particularly in Southeast Asia and other tropical regions. Its detection in human populations signals an evolving epidemiological landscape, challenging the previous notion that zoonotic hookworms have a negligi
