Zoonotic parasites associated with domestic dogs have been well-studied in the majority of Europe. In the Balkan region, however, there is minimal knowledge of the parasites in dogs in shelters for rehoming in other European countries. This study aimed to investigate parasitic infections in dogs from two private shelters in Pristina, Kosovo. Faecal samples were collected, representing both adult dogs (72%) and puppies (28%). Coproscopic analysis revealed that 88% of dogs were infected with at least one parasite, with hookworms being the most common. Amplicon metabarcoding targeting internal tr

A myriad of diseases can be treated by efficacious and potent drugs, yet the delivery efficiency is often hindered due to absorption issues, loss during first-pass metabolism, non-specific delivery, degradation before action and failure to comply to treatment. This has motivated researchers to develop novel methods for drug delivery, including live biotherapeutic products, notably transgenic bacteria delivering foreign therapeutic molecules. Recent advancements demonstrate that controlled experimental human helminth infections are tolerated and safe and may have natural protective attributes f

Comprehensive diagnosis of gastrointestinal parasites is largely reliant on traditional stool microscopy, despite gains in molecular diagnostics. Wet-mount examinations remain a significant challenge for traditional microscopy, digital microscopy, and artifical intelligence (AI). We developed and validated a deep convolutional neural network (CNN) model that provides highly sensitive detection and presumptive classification of enteric parasites. Twenty-seven different parasites were trained on a CNN model using a wide diversity of 4,049 unique parasite-positive specimens (determined by traditi

BACKGROUND: Soil-transmitted helminths (STHs) remain a public health problem for children in low- and middle-income countries. Conventional methods for STH diagnosis require well-equipped laboratories and trained personnel, which are often scarce or inaccessible in rural and resource-limited settings. Algorithmic non-biomedical screening tools are a potential strategy for community health workers (CHWs) to ensure rapid STH detection and referral of children. This exploratory feasibility study aimed at assessing whether a CHW-led, algorithm-based screening program could identify school-aged chi

BACKGROUND: Helminth infections are widespread in resource-limited settings, and modulate host immune responses, with potential implications for viral coinfections. Intestinal helminths can alter susceptibility to respiratory viruses, but the mechanisms influencing SARS-CoV-2 infection outcomes remain poorly understood.
METHODS: Using the Syrian hamster model, we investigated the impact of prior infection with the human hookworm Ancylostoma ceylanicum on host responses to SARS-CoV-2. Tissue-specific transcriptional responses were compared among four groups: naive, hookworm-only, SARS-CoV-2-on

Pelodera (A. Schneider, 1866) is a Clade V nematode genus of the Rhabditidae family and close relative of Caenorhabditis elegans. The genus comprises 26 species poised between free-living, commensal, and parasitic lifestyles, as well as species with underappreciated roles in soil nutrient cycling. Pelodera strongyloides, the type species, is a cause of follicular larva migrans among humans and other mammals but can be confused with hookworm folliculitis. With minimal genetic resources available, the capacity to identify and diagnose infectious species and strains of Pelodera are limited. Thus,

Vaccine responses are sometimes impaired in rural, low-income settings. Helminth-associated gut barrier dysfunction and microbial translocation (MT) may be implicated. We used samples from a trial of praziquantel treatment-effects on vaccine responses in Schistosoma mansoni (Sm)-endemic Ugandan islands, measuring intestinal fatty acid-binding protein 2 (I-FABP2), lipopolysaccharide-binding protein, anti-endotoxin core antibodies (EndoCab), soluble CD14 (sCD14) in plasma, and faecal lipocalin-2, occult blood (FOB), and calprotectin (fCAL), and evaluating their associations with baseline helmint

Microbial translocation drives chronic immune activation that is linked to HIV disease progression. Invasive parasitic gut nematodes induce microbial translocation and downregulate the expression of pro-inflammatory cytokines. We evaluated the impact of albendazole anthelmintic therapy on serum markers of microbial translocation and inflammation among helminth-infected Ugandans living with HIV. Participants were randomized to an immediate or delayed 400 mg of daily albendazole and followed for 1 month. Baseline stool analysis, conducted via multiparallel real-time quantitative polymerase chain

BACKGROUND: Migration to Europe has increased in recent years, with Italy serving as a major entry point. Ensuring adequate healthcare for newly arrived migrants includes the prevention and management of infectious diseases. This study aimed to estimate the prevalence of selected infections among migrants in northern Italy.
METHODS: We conducted a retrospective cross-sectional study including newly arrived migrants screened at the Department of Infectious-Tropical Diseases and Microbiology (DITM) of the IRCCS Sacro Cuore Don Calabria Hospital, Negrar di Valpolicella (Verona, Italy) between Ja

Soil-transmitted helminth (STH) infections are common. Indeed, more than 1 billion people are affected, mainly in the developing world where poverty prevails and hygiene behavior, water supply, and sanitation are often deficient. Ascaris lumbricoides, Trichuris trichiura, and the two hookworm species, Ancylostoma duodenale and Necator americanus, are the most prevalent STHs. The estimated global burden due to hookworm disease, ascariasis, and trichuriasis is 22.1, 10.5, and 6.4 million disability-adjusted life years (DALYs), respectively. Furthermore, an estimated 30-100 million people are inf

BACKGROUND: Mass drug administration (MDA) program of albendazole to at-risk populations as preventive chemotherapy is the core public health intervention to control soil-transmitted helminths (STHs). Achieving this goal relies on drug effectiveness in reducing the parasite reservoirs in the community and preventing reinfection. We assessed the efficacy of albendazole against STH parasite infection and reinfection status after cure.
METHODS: A total of 984 schoolchildren infected with at least one type of STH parasite (hookworm, Ascaris lumbricoides, Trichuris trichiura) in southern Ethiopia

Nationwide sampling by Venkatesan and colleagues (2023) described the resistance status of the canine hookworm, Ancylostoma caninum, to benzimidazoles across the USA via β-tubulin isotype-1 amplicon metabarcoding. In this study, we aimed to use the existing public amplicon metabarcoding data and mine it for the presence of β-tubulin isotype-1 sequences that belong to hookworm species other than A. caninum. Through bioinformatics analysis we assigned species to A. caninum, Ancylostoma braziliense, Ancylostoma tubaeforme and Uncinaria stenocephala. All non-A. caninum sequences contained only the

Although vaccines have reduced COVID-19 disease burden, their efficacy in helminth infection endemic areas is not well characterized. We evaluated the impact of infection by Heligmosomoides polygyrus bakeri (Hpb), a murine intestinal hookworm, on the efficacy of an mRNA vaccine targeting the Wuhan-1 spike protein of SARS-CoV-2. Although immunization generated similar B cell responses in Hpb-infected and uninfected mice, polyfunctional CD4+ and CD8+ T cell responses were markedly reduced in Hpb-infected mice. Hpb-infected and mRNA vaccinated mice were protected against the ancestral SARS-CoV-2

The amounts of parasite DNA in soil samples from different playgrounds and other public areas can help identify areas of possible microbe transmission and give indications of the possible occurrence of parasite infection in nearby communities. We collected 207 soil samples from parks in Paiute indigenous tribal areas in southwestern Utah and from the higher income city of St. George, Utah, and tested them for 11 parasites that can cause human disease. Molecular tests revealed an elevated odds ratio (OR) of 3.072 (range, 1.114-8.065) for detecting the helminth Trichuris trichiura and an elevate

Over the years, there has been progress in understanding the molecular aspects of iron metabolism and erythropoiesis. However, despite research conducted both in laboratories and living organisms, there are still unanswered questions due to the complex nature of these fields. In this study we investigated the effects of hookworm infection on iron metabolism and how the hosts response to anemia is affected using hamsters infected with Ancylostoma ceylanicum as a model. Our data revealed interesting relationships between infection-induced anemia, erythropoiesis, iron metabolism, and immune modul

My scientific life in translational medicine runs in two parallel, yet often converging paths. The first, is four-decade-long commitment to develop new vaccines for parasitic and neglected tropical diseases, as well as pandemic threats. This includes a vaccine for human hookworm infection that I began as an MD-PhD student in New York City in the 1980s, and a new low-cost COVID vaccine that reached almost 100 million people in low- and middle-income countries. Alongside this life in scientific research, is one in public engagement for vaccine and neglected disease diplomacy to ensure that peopl

OBJECTIVES: Kenya has implemented a national school-based deworming program, which has led to substantial decline in the prevalence of soil-transmitted helminths (STHs), although some pockets of infections remain. To effectively design an STH control program that leads to significant reductions of Trichuris trichiura, there is a need to understand the drivers of persistent infection despite ongoing treatment programs.
METHODS: This study was conducted between July and September 2019 at the south coast of Kenya, using a two-stage sampling design. First, a school-based cross-sectional survey wa

Parasites are generally associated with lower income countries in tropical and subtropical areas. Still, they are also prevalent in low-income communities in the southern United States. Studies characterizing the epidemiology of parasites in the United States are limited, resulting in little comprehensive understanding of the problem. This study investigated the environmental contamination of parasites in the southern United States by determining each parasite's contamination rate and burden in five low-income communities. A total of 499 soil samples of approximately 50 g were collected from p
