Hookworm genes encoding intestinal excreted-secreted proteins are transcriptionally upregulated in response to the host's immune system.

Hookworms are intestinal parasitic nematodes that chronically infect ~500 million people. How hookworms successfully overcome host protective mechanisms is unclear, but it may involve hookworm proteins that digest host tissues, or counteract the host's immune system, or both. To find such proteins in the zoonotic hookworm Ancylostoma ceylanicum, we used mass spectrometry to identify 565 genes encoding excreted-secreted (ES) proteins from adults, and used RNA-seq to identify genes expressed both in young adults (12 days post-infection) and in intestinal and non-intestinal tissues dissected from

Current issues in heartworm chemotherapy.

Several issues that affect prevention and treatment of heartworm infections require more intensive research. The incidence of heartworm infection in the USA is increasing, but the factors that underlie this trend remain incompletely understood. The contributions of climate change, vector range expansion, client compliance and resistance to macrocyclic lactones (ML) are likely interrelated and require investigation. Molecular-level research has not yet identified the causative mechanisms underlying ML resistance (MLR), but surveys of genomic markers associated with the trait reveal worrying tre

Na-GST-1 adsorbed on Alhydrogel co-administered with different Toll-like receptor agonists in hookworm-naive adults using a controlled human infection model in the USA: a phase 2, double-blind, randomised controlled trial.

BACKGROUND: Necator americanus glutathione S-transferase-1 (Na-GST-1) performs a crucial enzymatic step in hookworm blood feeding and is a leading target for vaccine development. Phase 1 trials in Brazil, Gabon, and the USA of recombinant Na-GST-1 adsorbed on Alhydrogel (Na-GST-1/Al), co-administered with or without AP 10-701 (Toll-like receptor [TLR] 4 agonist) or CpG 10104 (TLR9 agonist), showed the vaccine was well tolerated, safe, and immunogenic. A controlled human hookworm infection (CHHI) model was previously developed to evaluate the effect of candidate hookworm vaccines on infection.

Advances in vaccine development through the controlled human infection models for hookworm and schistosomiasis.

Controlled human infection models can play an important role in vaccine development, particularly for neglected tropical diseases such as helminth infections. Currently, controlled infection models have been established for schistosomiasis and hookworm. This review highlights the developments in the controlled human schistosomiasis infection model (CHI-S) and the controlled human hookworm infection model (CHHI) and their contributions to vaccine development. In general, both models are considered safe and well-tolerated. Measures to decrease risk of potential adverse events were taken when dev

Community-wide deworming strategies to reduce high hookworm burden in endemic communities: Results from a cluster randomized trial in Southern India.

OBJECTIVES: Targeted deworming of high-risk groups (usually school-based programs), although effective in controlling morbidity, may not be sufficient to reduce hookworm burden in endemic communities due to the persistence of infection in the adult population. The effectiveness of community-wide mass drug administration (MDA) strategies were evaluated in a hookworm-endemic tribal community in southern India. METHODS: In this open-label, cluster-randomized community-intervention trial, 45 villages comprising of 2770 households were randomly assigned (1:1:1) to receive community-wide MDA with a

The detection of three independent benzimidazole resistance mutations in a single Ancylostoma caninum population from a Golden Retriever breeding kennel in Sao Paulo, Brazil.

Multiple anthelmintic drug resistance (MADR) is widespread in the canine hookworm Ancylostoma caninum in kenneled greyhounds in the USA and Australia with benzimidazole resistance being associated with the isotype-1 β-tubulin SNPs Q134H(CAA > CAT) and F167Y(TTC > TAC) in both cases. These two SNPs have also been found to be widespread, often at high frequency, in A. caninum populations in pet dogs across the USA but the situation in domestic dogs in other countries is currently unknown. Here, we report the identification of a single A. caninum population, from a Golden Retriever breeding kenne

Maternal anemia in helminth-infected pregnant women: Dose-response with hookworm intensity and other risk factors in a Ugandan cross-sectional study.

BACKGROUND: Anemia remains a significant global public health challenge, affecting approximately 25% of the world's population and disproportionately impacting pregnant women, with an estimated 40% prevalence worldwide. Intestinal helminth infections exacerbate this burden, with anemia prevalence reported as high as 55.6% among infected pregnant women compared to 16.4% in those uninfected. This study aimed to determine the burden of anemia and its determinants among pregnant women with helminthiasis attending antenatal care at tertiary Hospitals in western Uganda. Methods: A hospital-based cro

Immune and Iron Metabolism Responses During the Acute Phase of Experimental Hookworm Infection in the Ancylostoma ceylanicum-Hamster Model.

Hookworm infections continue to impose a substantial burden on human and animal health, but the early host responses that influence parasite establishment are not fully characterized. Experimental models that reproduce key features of hookworm biology and host-parasite interactions remain essential for advancing translational research. In this study, we examined hematological, biochemical, immunological, and parasitological parameters during the acute phase of experimental hookworm infection using the Ancylostoma ceylanicum-Mesocricetus auratus model, a small-animal system widely employed for

Travelers Returning to Denmark with Hypereosinophilia and Ancylostoma Infections Detected Using Broad-Specificity Amplicon-Based Next-Generation Sequencing.

In nonendemic countries such as Denmark, hookworm infections in travelers may be rare and difficult to detect because of low-grade shedding of parasite eggs and a lack of clinical suspicion and relevant diagnostic capacity. Two cases of hookworm infection observed in 2025 that were detected only through amplicon-based next-generation sequencing (NGS) of fecal DNA are presented in the current study. Both infections were characterized by hypereosinophilia and acquired during 2-week holidays in Asia. Case 1 was infected with Ancylostoma ceylanicum during a holiday in the Philippines, and Case 2 a

American Association of Veterinary Parasitologists Hookworm Task Force review on best practices for diagnosis and treatment of multidrug-resistant hookworms.

Ancylostoma caninum, the canine hookworm, is the most common and clinically important gastrointestinal nematode parasite infecting dogs in the US. Common clinical signs associated with acute infections include anemia, melena, hematochezia, dehydration, and poor weight gain or weight loss. In the past few years, multiple anthelmintic drug resistance to fenbendazole, moxidectin, and pyrantel pamoate has been documented in A caninum in the US, and cases of drug-resistant canine hookworms are increasingly being reported in the US and Canada. Clinical and genetic evidence strongly suggests that mul

Prevalence and intensity of soil-transmitted helminths infections and associated factors among households in Gorgora, Ethiopia.

Although 44.6 million adults live in endemic areas, epidemiological studies and interventions primarily target school-age children (SAC). Hence, data on the prevalence and intensity of Soil-Transmitted Helminths (STHs) infections and associated factors among all age groups is crucial to expand interventions. This study aimed to determine the prevalence, intensity, and associated factors of STHs infections and to compare the diagnostic performance of Kato-Katz thick smear (KK) and formol-ether concentration techniques (FECT) in Gorgora, Western Dembia district, Ethiopia. A community-based cross

Comparison of Ritchie and Kato-Katz methods for the detection of intestinal helminths in humans: a systematic review and meta-analysis.

BACKGROUND: Comparative evidence on the diagnostic performance of Formalin-Ether Concentration (FEC) versus the Kato-Katz thick smear (KK) for detecting intestinal helminths in humans remains inconsistent. This systematic review and meta-analysis compared the odds of detecting intestinal helminths using FEC versus the single-slide KK. METHODS: A systematic search was conducted across six databases (PubMed, Scopus, EMBASE, Web of Science, Nursing and Allied Health Premium, and Ovid). Eligible studies directly compared FEC with single-slide KK for stool examination. Data was extracted using a s

Imported zoonotic Ancylostoma ceylanicum and Ancylostoma braziliense infections in a cat in Romania.

Hookworms are widespread zoonotic parasites affecting humans and companion animals worldwide. We report molecular characterization of Ancylostoma ceylanicum and A. braziliense in a cat imported into Romania from Guadeloupe, an overseas administrative region of the French Republic in the Caribbean. This finding underscores the role of molecular diagnostics in detecting the transboundary introduction of exotic hookworms with significant zoonotic potential.

Hookworm infestation in the biliary tract: a case report.

BACKGROUND: Hookworms are parasitic nematodes that primarily infect humans in impoverished communities. They attach to the intestines to feed on blood, which results in anemia and other health complications. While parasitic infections of the biliary tract can occur and lead to significant issues such as cholangitis, pancreatitis, and biliary obstruction, hookworms are not documented to be associated with these biliary infestations. CASE PRESENTATION: A 53-year-old female Tibeto-Burman ethnicity following Buddhism, with a significant smoking history and no alcohol consumption presented to Trib

Endoscopic Detection and Molecular Confirmation of Ancylostoma ceylanicum with Atypical Gastrointestinal Manifestations: A Case Series.

BACKGROUND: Hookworm is a widely known soil-transmitted helminth (STH) traditionally linked to iron-deficiency anemia caused by chronic blood loss in the small intestine. The classical human pathogens are Ancylostoma duodenale and Necator americanus. However, emerging evidence indicates that Ancylostoma ceylanicum, a hookworm of cats and dogs, is a significant cause of human infection, particularly in Southeast Asia and other tropical regions. Its detection in human populations signals an evolving epidemiological landscape, challenging the previous notion that zoonotic hookworms have a negligi
