Ancylostoma caninum is the most significant intestinal nematode parasite of dogs. We acquired fecal surveillance data from a large population of dogs in the United States (US). A diagnostic test using real-time PCR (qPCR) for Ancylostoma spp. and allele-specific qPCR detecting the SNP F167Y in the isotype 1 of the Beta-tubulin gene, was used in 885,424 randomized canine fecal samples collected between March 2022 and December 2023. Overall, Ancylostoma spp. had a prevalence of 1.76% (15,537/885,424), with the highest in the South 3.73% (10,747/287,576), and the lowest in the West 0.45% (632/140

BACKGROUND: The antigen Na-GST-1, expressed by the hookworm Necator americanus, plays crucial biochemical roles in parasite survival. This study explores the development of mRNA vaccine candidates based on Na-GST-1, building on the success of recombinant Na-GST-1 (rNa-GST-1) protein, currently assessed as a subunit vaccine candidate, which has shown promise in preclinical and clinical studies.
METHODOLOGY/FINDINGS: By leveraging the flexible design of RNA vaccines and protein intracellular trafficking signal sequences, we developed three variants of Na-GST-1 as native (cytosolic), secretory,

Hookworms are blood-sucking intestinal parasites that can cause anaemia and protein loss in humans. Ancylostoma ceylanicum, a zoonotic hookworm species of dogs, is the second most common cause of human hookworm infections. With the increasing anthelmintic resistance risks and the uncontrolled stray dog population in Sarawak Borneo, East Malaysia, understanding the genetic structure of A. ceylanicum is crucial for tracking mutation patterns and assessing zoonotic transmission risks. This study determined the prevalence and genetic diversity of dog hookworm species using microscopy, PCR and sequ

BACKGROUND: Hookworm infections such as Ancylostoma braziliense and A. ceylanicum pose a significant threat to pets and are implicated in causing zoonotic diseases. Despite the availability of preventatives, compliance can be lacking. Increasing pet owner options and combining endo- and ectoparasite treatments might improve this compliance. In four separate studies, we investigated the efficacy of Simparica Trio® (Zoetis Inc., Parsippany, NJ, USA) containing minimum dosages of 1.2 mg/kg sarolaner, 24 µg/kg moxidectin and 5 mg/kg pyrantel against fourth- and fifth-stage larvae (L4 and L5, respe

BACKGROUND: Hookworm infection remains of considerable importance to public health. However, because critical cases caused by hookworm infection are rarely observed in China, accurate and prompt diagnoses are difficult to achieve in clinical practice. In this study, we describe how we combined morphological and molecular approaches to achieve the clinical diagnosis of hookworm infection.
CASE PRESENTATION: A 75-year-old Chinese woman who presented with dizziness, poor appetite, poor sleep, and weakness in her limbs was diagnosed with chronic atrophic gastritis and was positive for Helicobacte

BACKGROUND: Necator americanus is the predominant species causing hookworm infections in humans. Despite advancements in prevention strategies, mild cases of infection still occur, highlighting the need for improved detection technology. Recombinase Polymerase Amplification (RPA) is an isothermal molecular diagnostic known for its sensitivity, speed, portability, and widespread application in detecting various pathogens. Although several molecular assays are available for N. americanus, they have limitations in detecting mild N. americanus infections.

Ancylostoma caninum, a zoonotic hookworm species, significantly affects the global health of companion animals, humans and wildlife populations. This parasitic infection is prevalent in various environments, particularly in regions with warm climates, and affects a wide range of canids, including dogs, where it is most commonly found. A. caninum is a major concern not only due to its zoonotic potential but also because of its growing resistance to anthelmintic treatments. The development of resistance in parasitic species is primarily driven by genetic mutations that allow the parasite to surv

Hookworms infect 450 million people globally and account for the loss of 5 million disability-adjusted life years annually. Over the last decade, the Human Hookworm Vaccine (HHV) candidate N. americanus Glutathione-S-Transferase-1 (Na-GST-1) has advanced to efficacy testing. This manuscript describes the manufacture of third-stage N. americanus larvae (NaL3) as an infectious challenge agent to provide "proof-of-concept" for the efficacy of Na-GST-1 prior to more extensive and more resource-intensive vaccine field trials in hookworm endemic areas. NaL3 were produced from fecal samples of three

Soil-transmitted helminths (STHs) are parasitic nematodes that infect humans, particularly in tropical and subtropical regions, where they contribute substantially to neglected tropical diseases (NTDs). Among them, hookworms (Ancylostoma duodenale, Necator americanus and Ancylostoma ceylanicum) cause substantial morbidity, leading to anaemia, malnutrition, and developmental impairment. Despite the global impact of hookworm disease, genomic research on A. duodenale has lagged behind that of other hookworms, limiting comparative and molecular biological investigations. Here, we report the first

Reverse vaccinology is a time- and cost-effective approach to identify potential vaccinable antigens for further in vivo experimental validation. Despite its wide application to multiple organisms, the use of in silico vaccine development tools to parasitic nematodes has been limited. Herein, we have used the rodent hookworm Nippostrongylus brasiliensis as a mouse model for the human hookworm Necator americanus to identify potential vaccine candidates against the latter. Our strategy combined advanced bioinformatic evaluations with knowledge-based criteria. A cumulative rating of antigenic pro

Cases of infection with the canine hookworm Ancylostoma caninum that are resistant to all anthelmintic classes registered in the United States of America (USA) for control of this parasite in dogs were first reported in 2019. Termed Multiple Anthelmintic Drug Resistance (MADR), cases have subsequently been reported in pet dogs across the USA. Recent studies using molecular tests that detect mutations known to cause resistance to benzimidazole anthelmintics have revealed an alarming frequency of this trait in pet dogs throughout the USA, with some reports also from Canada. Ancylostoma caninum and Uncinaria stenocephala are the most prevalent canine hookworms worldwide, both having veterinary and public health importance. Considering the emergence of anthelmintic resistance in A. caninum and the inefficacy of milbemycin oxime against U. stenocephala, the accurate diagnosis is essential, especially in areas where they occur in sympatry. Here, we differentiated hookworm species in naturally infected dogs from southern Italy, using coproculture and morphometric analysis of third-stage larvae (L3) and eggs, along with molecular characterization. Dog fecal samples (n

Data are sparse on the epidemiological picture of Taenia saginata taeniasis in Kenya. Infections are underreported, and their persistence nonetheless negatively impacts the beef industry. Populations vulnerable to taeniasis in the developing world are commonly burdened with other intestinal parasites, ubiquitous in unsanitary environments. This study aimed to estimate the occurrence of human taeniasis in Narok County, Kenya, and screen for the presence of other intestinal parasitic infections. A community-based survey was conducted in five pastoral wards, and stool samples, mainly from adults,
