Trichuriasis in Human Patients from Côte d'Ivoire Caused by Novel Trichuris incognita Species with Low Sensitivity to Albendazole/Ivermectin Combination Treatment.

Albendazole/ivermectin combination therapy is a promising alternative to benzimidazole monotherapy alone for Trichuris trichiura control. We used fecal DNA metabarcoding to genetically characterize Trichuris spp. populations in patient samples from Côte d'Ivoire showing lower (egg reduction rate <70%) albendazole/ivermectin sensitivity than those from Laos and Tanzania (egg reduction rates >98%). Internal transcribed spacer (ITS) 1 and ITS2 metabarcoding revealed the entire detected Côte d'Ivoire Trichuris population was phylogenetically distinct from T. trichiura found in Laos and Tanzania an

Soil-transmitted helminthiasis among adolescents in Anaocha Local Government Area, Anambra State, Nigeria: Insights and recommendations for effective control.

Over the past decade, Anambra State, Nigeria, has implemented mass administration of medicines (MAMs) to combat soil-transmitted helminthiasis (STH), a significant public health challenge in low-income regions. Nevertheless, these efforts have predominantly focused on pre-school and school-aged children, leaving a notable gap in understanding STH infection rates and the efficacy of these campaigns among secondary school adolescents, who have been excluded from this initiative. Our study aimed to address this critical knowledge gap by assessing soil-transmitted helminthiasis (STH) prevalence an

Trichuris infection in captive non-human primates in zoological gardens in Spain.

Currently, there is limited available information on the epidemiology of parasitic infections in captive non-human primates (NHPs) and their zoonotic potential. However, numerous cases of helminth infections in NHPs have been documented in several zoos around the world, with one of the most prevalent being those of the genus Trichuris. The main objective of this study is to investigate the occurrence of infection by Trichuris spp. in primates from zoological gardens in Spain and to ascertain, at the species level, the specific Trichuris species harbored by these hosts by using mitochondrial an

A combined treatment regimen for Trichuris rhinopiptheroxella infection in Rhinopithecus roxellana in southern China.

The Sichuan snub-nosed monkey (Rhinopithecus roxellana) is an endangered species in China and is susceptible to infection with Trichuris species. However, the worms are difficult to remove completely. A practical treatment regimen for trichuriasis was conducted over a seven-month period on 15 R. roxellana in a wildlife zoo in southern China. Initially, a combination of fecal examination, morphological observation, molecular identification of ITS1 and mitogenome, and infective pattern analysis revealed that R. roxellana was susceptible to Trichuris rhinopiptheroxella. Three rounds of treatment

Hatching of whipworm eggs induced by bacterial contact is serine-protease dependent.

Whipworms (Trichuris spp) are ubiquitous parasites of humans and domestic and wild mammals that cause chronic disease, considerably impacting human and animal health. Egg hatching is a critical phase in the whipworm life cycle that marks the initiation of infection, with newly hatched larvae rapidly migrating to and invading host intestinal epithelial cells. Hatching is triggered by the host microbiota; however, the physical and chemical interactions between bacteria and whipworm eggs, as well as the bacterial and larval responses that result in the disintegration of the polar plug and larval

Chronic diarrhea unveiled by colonoscopy.

A 62-year-old woman from Bogotá, Colombia, had chronic diarrhea for 4 years. The diarrhea was lienteric, occurring day and night, with mucus but no blood. She had no significant weight loss, abdominal pain, or other systemic symptoms, and no notable medical history. Initial stool tests showed leukocytes and blood but no parasites. Routine lab tests were normal. Due to the chronic symptoms and nocturnal diarrhea, a colonoscopy was performed. It revealed numerous mobile, white worms in the right colon, identified as Trichuris trichiura, causing friable mucosa. She was diagnosed with trichuriasis

Experimental trichuriasis: Changes in the immune response and bacterial translocation during acute phase development illustrated with 3D model animation.

Trichuriasis, a well-known type of soil-transmitted helminthiasis, is a neglected gastrointestinal nematode disease predominantly affecting children in tropical regions and is caused by Trichuris trichiura. The potential zoonotic transmission of this disease is indicated by its presence in nonhuman primates. Chronic infection leads to mucosal damage, bacterial translocation, and intense inflammatory infiltration; however, the progression of these processes remains poorly understood. This study tracks the acute phase of experimental trichuriasis, providing detailed insights into nematode tissue

Efficacy and safety of ivermectin-albendazole combination versus ivermectin or albendazole monotherapy in soil-transmitted helminthiasis: A systematic review and meta-analysis.

BACKGROUND: Soil-transmitted helminthiasis remains a daunting challenge to global health, exerting its greatest toll on resource-limited regions of the world. A dual drug approach using the co-administration of ivermectin and albendazole has shown promising results in comparison to the traditional monotherapy strategy. In light of this, a systematic review and meta-analysis of randomized controlled trials was conducted. METHODS: Several electronic databases including PubMed, Cochrane Central, Google Scholar, and Embase were explored to search for relevant studies from inception to September 2

A Honduran Prevalence Study on Soil-Transmitted Helminths Highlights Serological Antibodies to Tm-WAP49 as a Diagnostic Marker for Exposure to Human Trichuriasis.

Soil-transmitted helminth (STH) infections rank among the most prevalent communicable diseases of humans, yet detection of these parasites is mostly restricted to identifying active infection through fecal examinations. Currently, there are no commercial diagnostic tools to identify a prior whipworm or hookworm exposure, and the few serological assays for roundworm infection have not been well validated for crossreactivity or infections in humans. Such diagnostic restrictions limit the range of scientific and clinical questions that surround STH exposures and their implicated relationship to c

An optimised faecal microRNA sequencing pipeline reveals fibrosis in Trichuris muris infection.

The intestine is a site of diverse functions including digestion, nutrient absorption, immune surveillance, and microbial symbiosis. Intestinal microRNAs (miRNAs) are detectable in faeces and regulate barrier integrity, host-microbe interactions and the immune response, potentially offering valuable non-invasive tools to study intestinal health. However, current experimental methods are suboptimal and heterogeneity in study design limits the utility of faecal miRNA data. Here, we develop an optimised protocol for faecal miRNA detection and report a reproducible murine faecal miRNA profile in h

Performance of real-time polymerase chain reaction and Kato-Katz for diagnosing soil-transmitted helminth infections and evaluating treatment efficacy of emodepside in randomized controlled trials.

BACKGROUND: The World Health Organization recommends the use of the microscopy-based Kato-Katz thick smear for diagnosing soil-transmitted helminth (STH) infections. Despite its simplicity and cost-effectiveness, the Kato-Katz method faces challenges, including reader subjectivity and reduced sensitivity. Real-time polymerase chain reaction (qPCR) technology offers standardized readouts and higher sensitivity, making it suitable for STH diagnosis and monitoring the treatment efficacy of emodepside within the framework of randomized controlled trials. METHODOLOGY/PRINCIPAL FINDINGS: We evaluat

The proteome of human adult whipworm Trichuris trichiura: A source of potential immunomodulatory molecules.

Soil-transmitted helminths (STHs), including Trichuris trichiura, pose a major global health challenge, particularly in children, causing significant morbidity. However, T. trichiura's ability to modulate host immune responses offers a unique opportunity to discover biomolecules with therapeutic potential for inflammatory, allergic, and autoimmune disorders. This study conducted a proteomic analysis of adult male and female T. trichiura using liquid chromatography-tandem mass spectrometry (LC-MS/MS), identifying 810 parasite proteins. Of these, 177 were exclusive to females, 277 to males, and

Structural insights into Trichuris muris eggs through 3D modeling, Cryo-SEM, and TEM of samples prepared with HPF-FS.

Trichuriasis - a disease caused by Trichuris trichiura - affects underserved communities. Infection occurs by the ingestion of embryonated eggs, a resilient structure against environmental fluctuations - an essential feature for the survival and transmission of trichurids. This study aims to enhance our comprehension of the trichurid eggs by providing a characterization of the ultrastructure of eggshell and first-stage (L1) larvae of T. muris, a key experimental model for trichuriasis. We employed the following microscopy techniques: light, fluorescence, confocal, Cryo-SEM, and Transmission el

Gut microbiota regulates intestinal goblet cell response and mucin production by influencing the TLR2-SPDEF axis in an enteric parasitic infection.

Alterations in goblet cell biology constitute one of the most effective host responses against enteric parasites. In the gastrointestinal (GI) tract, millions of bacteria influence these goblet cell responses by binding to pattern recognition receptors such as toll-like receptors (TLRs). Studies suggest that the gut microbiota also interacts bidirectionally with enteric parasites, including Trichuris muris. Here, we study the roles of T. muris-altered microbiota and the TLR2-SPDEF axis in parasitic host defense. In acute T. muris infection, we observed altered gut microbiota composition, which

Notch-activated basophils support intestinal CD4+ T cell fate and function during Trichuris muris infection.

Helminth infections affect billions of people worldwide and cause substantial morbidity. Intestinal helminth infection provokes Type 2 inflammation orchestrated by CD4+ T helper type 2 (Th2) cells. Th2 cells cooperate with group 2 innate lymphoid cells (ILC2s) to produce interleukin (IL)-4 and IL-13 that prompt an epithelial "weep and sweep" response to drive parasite clearance. Tissue-specific cues optimize CD4+ T cell responses, but the mechanisms regulating intestinal Th2 responses remain unclear. Previously, we identified that the Notch signaling pathway in basophils, rare granulocytes, dr

Nematode diversity in Mastomys rodents (Rodentia: Muridae) across a wildlife-human/domestic animal interface and molecular characterization of Trichuris species from M. natalensis.

Nematodes comprise many species with diverse life histories and zoonotic potential. Globally, over 10,500 nematode species were reported in mammals, particularly rodents. In South and southern Africa, data on nematode diversity in native rodent populations remains limited. The current study aimed to address this gap by reporting nematode diversity in two Mastomys species (Mastomys coucha and M. natalensis) and conducting molecular analysis of an unidentified Trichuris species recovered from M. natalensis. A total of 68 M. natalensis and 27 M. coucha rodents were trapped across a wildlife-human

AI-supported versus manual microscopy of Kato-Katz smears for diagnosis of soil-transmitted helminth infections in a primary healthcare setting.

Soil-transmitted helminths primarily comprise Ascaris lumbricoides, Trichuris trichiura, and hookworms, infecting more than 600 million people globally, particularly in underserved communities. Manual microscopy of Kato-Katz thick smears is a widely used diagnostic method in monitoring and control programs, but is time-consuming, requires on-site experts and has low sensitivity, especially for light intensity infections. In this study, portable whole-slide scanners and deep learning-based artificial intelligence (AI) were deployed in a primary healthcare setting in Kenya. Stool samples (n = 96

Functional annotation and identification of novel drug targets from uncharacterized proteome of Trichuris trichiura.

INTRODUCTION: Trichuris trichiura, a soil-transmitted helminth, resides in the large intestine of humans, causing an asymptomatic disease known as trichuriasis. This global health concern is particularly prevalent in low- or middle-income countries, representing a significant burden on public health as one of the most neglected tropical diseases. The diminishing effects of currently available anthelmintic drugs, attributed to escalating drug resistance, warrants an urgent need for alternative and more potent vaccines or drugs. A substantial portion of the proteins in the T. trichiura genome ar
