The immunomodulatory p43 secreted protein of Trichuris whipworm parasites is a lipid carrier that binds signalling lipids and precursors.

Trichuris whipworms cause disease and morbidity in humans and other animals. Their prolonged intestinal infections persist despite intact immune systems of their hosts and are attributed to immunomodulatory activities of their secretions. The p43 (Tm-DLP-1) protein of Trichuris muris of mice comprises 95% of the protein secreted by adult parasites, binds matrix proteoglycans, and has immune cytokine (IL-13)-neutralising activity. Using fluorescence-based methods we show that p43 binds fatty acids and retinol, including signalling lipids or precursors thereof. The orthologue of p43 from the hum

Anthelmintic efficacy of mebendazole and levamisole in Olive Baboon (Papio anubis) infected with Trichuris trichiura.

The olive baboon (Papio anubis) belongs to the family Cercopithecoidea and lives in different environmental conditions as well as they are also kept in zoological gardens as a source of public entertainment. Poor growth and health disorders are commonly reported issues in olive baboons in captivity mainly because of gastrointestinal infections and endo-parasites. The present study was conducted on 15 dewormed olive baboons kept in Lahore Zoo, Lahore, Pakistan, and diagnosed with Trichuris trichiura (roundworm). Infected olive baboons were selected and randomly divided into 3 groups, i.e., A, B

Efficacy and safety of moxidectin-albendazole combination therapy for Trichuris trichiura infections in school-aged children: a double-blind, randomised, controlled, superiority trial.

BACKGROUND: Infection with the soil-transmitted helminth Trichuris trichiura affects up to 300 million people globally, with children in rural areas in less economically developed countries being most at risk. If untreated, infection compromises physical and cognitive development and leads to long-lasting morbidity. We assessed whether moxidectin co-administered with albendazole is superior to the recommended albendazole monotherapy in treating trichuriasis in school-aged children. METHODS: This randomised, double-blind, parallel-group, superiority, phase 3 trial took place between May 14 and

Geostatistical modelling of soil-transmitted helminth prevalence in Kenya: Informing targeted interventions to accelerate elimination efforts.

OBJECTIVES: This study estimates the prevalence of soil-transmitted helminths (STH) in Kenya, identifying high- and low-burden communities to optimize intervention strategies. METHODS: Using prevalence data from 146,668 children in 3752 schools across 207 sub-counties in Kenya, collected in 2021 and 2024, alongside environmental and socio-economic data, we modelled the prevalence of hookworm, Ascaris lumbricoides and Trichuris trichiura at a 5 × 5 km resolution. We estimated the probability of exceeding predefined prevalence thresholds (2%, 10% and 20%) and the population-weighted mean preval

Transcriptional responses of mouse proximal colon and colonoids during early whipworm infection.

Trichuriasis, caused by the parasitic nematode Trichuris trichiura, affects 429-508 million people worldwide. Although the early phase of whipworm infection is crucial for establishing infection, its underlying molecular mechanisms remain unclear. In this study, we examined and compared host transcriptional responses during early infection of the mouse whipworm, T. muris, using first-stage larvae (L1) in proximal colons from in vivo models (B6 and STAT6-deficient mice) and in vitro colonoid models. Differentially expressed genes and functional enrichment analysis revealed that while the "neuro

Whipworm infection remodels the gut microbiome ecosystem and compromises intestinal homeostasis in elderly patients revealed by multi-omics analyses.

INTRODUCTION: Whipworm (Trichuris trichiura) coexists with symbiotic microbiota in the gastrointestinal ecosystem. There is a paucity of data on the association between whipworm infection and the gut microbiota composition in elderly individuals. This study was designed to investigate changes in gut microbiota and function and its metabolite profile in patients with whipworm infection. METHODS: We used 16S rRNA gene sequencing to identify microbial signatures associated with whipworm infection. Subsequently, shotgun metagenomic sequencing revealed functional changes that highlighted disruptio

Soil-Transmitted Helminths and the Intricacies of Immunoregulation: Evidence From Amazonian Ecuador for the Importance of Considering Species-Specific Effects Within the Old Friends Hypothesis.

OBJECTIVES: The old friends hypothesis (OFH) examines connections between the global increase in immunoregulatory diseases (e.g., allergy and autoimmunity) and reduced exposure to immune-priming symbionts like soil-transmitted helminths. Helminth species, however, vary in their effects on hosts and should be considered separately. We examined relationships between species-specific helminth infection and circulating biomarkers of adaptive immune antibodies (total immunoglobulin E [IgE]), systemic inflammation (C-reactive protein [CRP]), and immune regulation (interleukin-6 [IL-6]), among Indige

Prevalence & determinants of soil-transmitted helminth infection among 1-15 year-olds in a tribal-dominated district of Odisha, India.

Background & objectives Soil-transmitted helminth (STH) infections are a significant public health concern, affecting over 1.5 billion people worldwide. STH infection among children leads to stunted growth, impaired cognitive development, and reduced productivity. In India, while multi-prong efforts are being made to control STH infections including mass drug administration (MDA) programme, the problem is still prevalent. This study aimed to investigate the prevalence, type of STH and determinants associated with STH infection among children (1-15 yr) in a tribal dominated district of Odisha,

Diagnostic sensitivity of formalin-fixed faecal microscopy for the detection of soil-transmitted helminths.

BACKGROUND: Faecal microscopy is the mainstay of soil-transmitted helminth diagnosis and commonly completed on formalin-fixed samples when resources are insufficient to analyse fresh samples. This study assessed the diagnostic sensitivity of microscopic techniques using formalin-fixed samples. METHODS: Formalin-fixed faecal samples from 574 individuals were tested by the formalin-ethyl acetate concentration technique (FECT), Malachite smear, McMaster and McMaster2 methods. Agreement between tests was assessed by Kappa. Bayesian latent class models and a composite reference standard estimated

Individual-level analysis of the efficacy of albendazole and albendazole-ivermectin fixed-dose coformulation (FDC) against Trichuris trichiura and hookworms.

BACKGROUND: Anthelmintic drugs are vital for controlling soil-transmitted helminths (STH), but treatment failures remain common. The egg reduction rate (ERR) is a key metric for assessing anthelmintic efficacy. This study calculated individual ERRs (ERRi) to evaluate a new anthelmintic drug and identify factors associated with treatment failure. METHODOLOGY: Data were collected from 369 children infected with T. trichiura and 226 infected with hookworms, who participated in a Phase II/III clinical trial. Participants received a single dose of ivermectin/albendazole 9/400 mg or 18/400 mg fixed

Geostatistical analysis to guide treatment decisions for soil-transmitted helminthiasis control in Uganda.

BACKGROUND: Soil-transmitted helminth (STH) infections remain a public health problem in Uganda despite biannual national deworming campaigns implemented since the early 2000s. Recent surveys have indicated a heterogeneous STH infection prevalence, suggesting that the current blanket deworming strategy may no longer be cost-effective. This study identified infection predictors, estimated the geographic distribution of STH infection prevalence by species, and calculated deworming needs for school-age children (SAC). METHODOLOGY: Bayesian geostatistical models were applied to STH survey data (2

Efficacy and safety of ascending doses of orodispersible ivermectin co-administered with albendazole for Trichuris trichiura infections in preschool-aged children in Tanzania: a single-blind, randomised, controlled, dose-ranging, phase 2 trial.

BACKGROUND: Children in resource-limited rural areas are at a high risk of infection with Trichuris trichiura, a soil-transmitted helminth affecting up to 300 million people globally. Heavy infections can result in long-term morbidity, impairing physical and cognitive development. There is insufficient evidence on co-administration of ivermectin-albendazole in children younger than 6 years infected with T trichiura. We aimed to identify the optimal dose of ivermectin given as a new orodispersible tablet formulation (co-administered with 400 mg albendazole) in children aged 2-5 years with trich

Anthelmintic and Antibiotic Therapy Resolves Intestinal Inflammatory Infiltration in Experimental Trichuriasis.

Despite significant advances in human health, soil-transmitted helminths (STH) continue to pose a major public health challenge, particularly in impoverished regions. Albendazole has been used to treat STH for over 40 years and remains widely utilized in mass drug administration programs. However, it is estimated that over 1.5 billion people are still infected globally, with Brazil reporting a prevalence of 5.41% for human trichuriasis. The nematode Trichuris muris is widely used in murine models to study trichuriasis due to its impact on the epithelial mucosa, including tissue damage, dysbios

Ivermectin-albendazole combination versus ivermectin or albendazole alone in soil transmitted helminthiasis: An updated systematic review and meta-analysis.

BACKGROUND: Soil-transmitted helminth (STH) infections caused by Trichuris trichiura, Ascaris lumbricoides, and hookworms continue to exhibit a significant prevalence in low and middle-income countries. Monotherapy with albendazole or ivermectin demonstrates limited effectiveness against these, prompting the exploration of combinations of benzimidazoles and macrocyclic lactones as potential alternatives. METHODS: We conducted an updated systematic review and meta-analysis of randomized controlled trials (RCTs) published from March 2023 to May 2025, comparing ivermectin-albendazole (IVM+ALB) w

Anthelmintics Derived from the Kinase Inhibitor SGI-1776 for the Treatment of Gastrointestinal Worm Infections.

Human gastrointestinal nematodes (GINs) infect at least one billion people, mostly children, with five billion people at risk of infection. Reduced and low efficacy of currently used anthelmintics, e.g., benzimidazoles against Trichuris trichiura whipworms, urges new anthelmintics to control these parasites. We previously discovered two human pan-PIM kinase inhibitors, CX-6258 and SGI-1776, with potent antihelmintic activity against GINs. Here, structure-activity relationship (SAR) studies were conducted to identify SGI-1776 analogs with improved cross-clade adulticidal activity (Ancylostoma c

Molecular diagnosis of Trichuris trichiura: Prevalence and associated risk factors in children under five living in a malaria-endemic area in Papua, Indonesia.

BACKGROUND: Trichuris trichiura, a soil-transmitted helminth (STH), infection is highly prevalent in children and, if not treated, can cause adverse health consequences. Microscopy using the Kato-Katz method is the mainstay of STH diagnosis in most settings but has low sensitivity. This study aims to quantify prevalence and examine risk factors of trichuriasis among children in a malaria-endemic area. METHODS: The study recruited 181 children aged <5 years old from a household survey conducted in 16 villages in Timika, Papua, Indonesia, from April to July 2013. Clinical and laboratory data we
