Specialized proresolving mediator-loaded extracellular vesicles mitigate pulmonary inflammation.

Extracellular vesicles (EVs) have emerged as versatile carriers of therapeutic cargo, including nucleic acids, proteins, and small molecules. However, their potential to deliver bioactive lipid mediators remains largely unexplored. Here, we present a novel synthetic biology-based strategy to selectively load EVs with proresolving lipid mediators of the resolvin D- and E-series by coexpressing the resolvin biosynthetic enzymes cyclooxygenase 2, 5-lipoxygenase, and 15-lipoxygenase using a custom-designed multigene expression vector. Human embryonic kidney 293 T cells transfected with the multige

Clonal Hematopoiesis and Incident Heart Failure.

IMPORTANCE: Clonal hematopoiesis of indeterminate potential (CHIP), the age-related clonal expansion of hematopoietic cells with acquired preleukemic variants, has been associated with cardiometabolic diseases, including heart failure (HF). However, prior studies have lacked power to examine less common CHIP driver variants and have not investigated potential mediators of the CHIP-HF association. OBJECTIVE: To test whether specific CHIP subtypes are associated with incident HF and determine the extent to which CHIP-associated comorbidities mediate this association. DESIGN, SETTING, AND PARTI

Transforming growth factor-β3 attenuates septic cardiomyopathy by reversing cardiomyocyte metabolic reprogramming through Smad7 signaling.

Septic cardiomyopathy (SCM) exhibits over 70% mortality, primarily attributed to cardiomyocyte metabolic reprogramming shifting from oxidative phosphorylation (OXPHOS) to glycolysis. Although transforming growth factor-β3(TGF-β3) modulates energy metabolism in other tissues, its function in SCM pathogenesis remains unexplored. In vivo, cecal ligation puncture (CLP) rat models received myocardial injections of TGF-β3-overexpressing or interfering adenovirus. Myocardial injury through histopathology (H&E) and apoptosis (TUNEL), and mitochondrial ultrastructure via transmission electron microscop

Liraglutide Ameliorates Injury-Related Vascular Repair by Activating Autophagy Through Modulation of the PI3K-mTOR Signaling Pathway.

Chronic stress is a significant risk factor for postoperative restenosis and other cardiovascular diseases, as it can promote the intimal thickening of damaged vessels. Liraglutide is used primarily for the treatment of type 2 diabetes and obesity. Recently, the protective effects of liraglutide on the cardiovascular system have garnered widespread attention; however, its specific mechanism of action remains unknown. The aim of this study was to investigate the impact of liraglutide on chronic stress-induced thickening of the damaged vascular intima and to elucidate the underlying mechanism in

Co-regulatory crosstalk between m5C, m7G, and o8G RNA modifications via QKI/YBX1 axis in myocardial ischemia-reperfusion injury.

BACKGROUND: Ischemia-reperfusion (I/R) injury in the heart triggers oxidative stress and alters post-transcriptional gene regulation. Reactive oxygen species (ROS) generated during oxidative stress induce RNA modifications such as 8-oxo-guanosine (o8G). Other modifications including 5-methylcytosine (m5C) and 7-methylguanosine (m7G) may also contribute to cardiac dysfunction. While the roles of individual RNA modifications in I/R injury are increasingly recognized, the global dynamics and crosstalk among these modifications under oxidative stress remain largely unexplored. METHODS: We perform

Intrinsic muscle stem cell dysfunction underlies functional deficits in models of type 1 diabetes.

Muscle function and regeneration are impaired in type 1 diabetes, but whether this arises directly from muscle stem cell (MuSC) dysfunction has not been addressed. Here, we utilized three-dimensional MuSC cultures (micromuscles) to demonstrate that hyperglycemia drives deficits in muscle stem cell function, leading to impaired force production in differentiated myotubes. The functional capacity of skeletal muscle was shown to decline after repeated bouts of injury in mouse models of type 1 diabetes, and this was replicated in micromuscles derived from MuSCs isolated from diabetic mice, indicat

Impact of partial hepatectomy versus other major abdominal surgeries on postoperative myocardial injury: a propensity score-matched analysis.

BACKGROUND: Partial hepatectomy (PH) may increase the risk of myocardial injury after non-cardiac surgery (MINS), a complication associated with substantial perioperative morbidity and mortality. Direct comparisons of MINS incidence between PH and other major abdominal surgeries (MAS) remain limited. This study evaluated whether PH confers greater risk of postoperative MINS compared with other MAS. METHODS: We conducted a retrospective propensity score-matched cohort study using the INSPIRE database. Adult patients undergoing PH or other MAS between 2011 and 2020 were identified. After 1:2 pr

GLP-1 receptor agonists and next-generation metabolic hormone therapies in chronic kidney disease.

Chronic kidney disease (CKD) remains a major public health problem, with type 2 diabetes and obesity representing key risk factors worldwide. The complex pathophysiology and the metabolic risk factors shared between type 2 diabetes, obesity, CKD and cardiovascular disease have led to the concept of a cardiovascular-kidney-metabolic (CKM) syndrome. The treatment landscape for CKM changed dramatically when agents from several medication classes, originally developed as glucose-lowering therapies, were recognized to reduce the risk of multiple components of CKM syndrome. Incretin-based therapies,

Anthracycline cardiotoxicity: role of metabolic vulnerability induced by cardiac pressure overload.

BACKGROUND AND AIMS: Hypertension and valvular heart disease, both associated with left ventricular (LV) pressure overload, increase the risk of anthracycline cardiotoxicity. While epidemiologically established, the underlying mechanisms remain unclear, precluding identification of therapeutic targets. METHODS: Two-month-old Yucatan pigs (males and females) underwent aortic banding to induce LV pressure overload or no operation. After 4 months, animals received a low-risk cumulative dose of doxorubicin (5 weekly 1 mg/kg intravenous injections) or vehicle, generating four groups: (i) healthy c

A multidimensional analysis of fall risk among older adults in India: evidence from the longitudinal ageing study in India (LASI).

BACKGROUND: Falls are a major public health concern among older adults in India. While earlier studies using LASI data have identified some risk factors, they did not account for key geriatric syndromes, physical performance, and functional status. This study aims to comprehensively examine the determinants of falls among older adults using nationally representative LASI Wave 1 data. METHODS: We analysed data from 26,780 community-dwelling individuals aged ≥ 60 years from LASI Wave 1. Falls were self-reported. Covariates included socio-demographics, comorbidities, ADL/IADL, cognition, depress

Ergothioneine attenuates whole-abdominal irradiation-induced multi-organ injury via the gut-heart-brain axis by modulating calcium voltage-gated channel subunit alpha1 C (Cacna1c) expression.

Despite the precise targeting of radiation therapy, collateral damage to adjacent healthy tissues remains an inevitable consequence. Currently, no effective clinical intervention exists to prevent or alleviate these adverse effects. To address this issue, our study established a whole-abdominal irradiation (WAI) model using C57BL/6J mice to investigate the systemic effects of ionizing radiation (IR) on the gut and other organs. Results confirmed that IR not only causes significant intestinal damage but also induces cardiac injury and cognitive dysfunction through remote effects. Ergothioneine

Heat stroke and the liver: mechanisms of injury and therapeutic strategies.

Global warming is leading to a continuous increase in the incidence of heat-related illnesses, among which heat stroke is the most severe type, characterized by a core body temperature > 40 °C and multiple organ dysfunction syndrome (MODS). The liver, being a thermosensitive organ, acts both as a target of injury and an "amplifier" of inflammation. Its acute injury and failure are significant causes of death in heat stress. This review summarizes the core mechanisms of heat stress-induced liver injury, including direct damage to hepatocytes by hyperthermia, the dual regulatory role of heat sho

Prognostic Value of Clinical and Instrumental Characteristics in Determining the Outcome of Patients With Acute Decompensated Heart Failure.

Aim        Evaluation of clinical, demographic, laboratory, and instrumental characteristics of patients with acute decompensated heart failure (ADHF) depending on the outcome over the hospitalization period.Material and methods    The prevalence of chronic heart failure (CHF) and ADHF remains extremely high. Hospitalization for ADHF is the most important predictor of death and readmission in the long term, and each subsequent hospitalization significantly increases the risk of death. Research of in-hospital mortality in this group of patients is limited in the Russian literature; however, num

Procoagulant platelets: linking coagulation and thromboinflammation in cardiovascular disease.

Platelets are the key cell types in haemostasis after vascular injury and crucially contribute to thrombus formation. In addition to their aggregation and clot contraction functions after stimulation by soluble agonists or extracellular matrix proteins, platelets can adopt a highly activated state known as procoagulant activation. Procoagulant platelets influence the pathophysiology underlying various cardiovascular diseases, including myocardial infarction, stroke and deep vein thrombosis. Findings described in the past decade position procoagulant platelets at the dynamic intersection betwee

Disease tolerance and infection pathogenesis age-related tradeoffs in mice.

Disease tolerance is a defence strategy essential for survival of infections, limiting physiological damage without killing the pathogen1,2. The disease course and pathology an infection may cause can change over the lifespan of a host due to the structural and functional physiological changes that accumulate with age. Because successful disease tolerance responses require the host to engage mechanisms that are compatible with the disease course and pathology caused by an infection, we predicted that this defence strategy would change with age. Animals infected with a 50% lethal dose  (LD50) o

The cumulative physiological effects of consecutive days of simulated occupational heat stress: study protocol for a randomised controlled trial with healthy adults undertaken in Australia.

BACKGROUND: Fuelled by rising global temperatures, occupational heat stress has become a critical threat to the health and safety of workers. This threat may be particularly great for workers who are required to perform prolonged work in the heat over multiple consecutive days; however, the cumulative physiological impact of these work conditions remains unclear. As such, the primary objective of this study is to determine the effects of three consecutive simulated physically demanding workdays in a hot and humid environment on core temperature (primary outcome), with secondary objectives exam
