Peiminine-Induced Selective Autophagy of AIM2 Inflammasomes Rescues Cerebral Ischemic Injury.

BACKGROUND: Inflammasome hyperactivation drives neuroinflammation following cerebral ischemia, yet pharmacological strategies inducing their selective clearance remain largely unexplored. Selective autophagy holds particular promise as a therapeutic strategy by eliminating detrimental substrates while avoiding the side effects of excessive autophagy. However, few compounds are currently available to achieve this selective regulation. METHODS: Transient middle cerebral artery occlusion was used to screen the active compounds from Fritillaria, combined with TTC staining and neurological scores.

Sustained alogliptin elicits superior control of glycemia and related complications in diabetic rats: Effects on vital organs functions, biochemistry, and structure.

Despite its efficacy in diabetes management, concerns about potential cardiovascular injury associated with alogliptin have limited its widespread use. This study investigated whether sustained, once-weekly low-dose alogliptin provides greater efficacy with fewer adverse effects than daily administration. Type 2 diabetes was induced in rats using a high-fructose, high-fat, high-salt diet for three weeks, followed by streptozotocin (STZ) injection. Diabetic rats were then allocated into three groups: diabetic, S Alogliptin (weekly administered sustained alogliptin preparation), and F Alogliptin

Integrative insights into fatty acid metabolism in diabetic cardiomyopathy: From molecular mechanisms to therapeutic strategies.

Diabetic cardiomyopathy (DCM) is a diabetes-specific cardiac dysfunction independent of other cardiovascular diseases. Recent studies highlight dysregulated fatty acid (FA) metabolism as a central driver of its pathogenesis. In the diabetic heart, excessive FA uptake and oxidation imbalance with glucose utilization led to lipid accumulation, mitochondrial overload, and oxidative stress. These changes trigger inflammatory responses, impair mitochondrial structural integrity and quality control, and disrupt cellular energy homeostasis. Over time, this maladaptation promotes cardiomyocyte hypertr

Risk factors for electrographic seizures in infants after cardiac surgery and their association with outcomes: a retrospective cohort study.

Neonates and infants undergoing cardiac surgery are prone to cerebral injuries. Neuromonitoring with continuous conventional electroencephalography (cEEG) and/or amplitude integrated electroencephalography (aEEG) is able to detect subclinical abnormal cerebral patterns, especially seizures, which have been found to correlate with medium-long term neurocognitive deficits. The endpoints of this study are (i) to define a risk model for electrographic subclinical epileptic seizures (ES) and (ii) to investigate the relationship between ES and neurologic and non-neurologic outcomes. Retrospective an

Exploring Sex Differences in Stroke Outcomes: A Comprehensive Analysis From the SPAN 1 Trial.

BACKGROUND: Stroke is a sexually dimorphic disease, with different risk factors, incidence, outcomes, and treatment responses in men and women. While sex differences have been documented in preclinical studies, these findings often come from single-site studies with small sample sizes and require validation across diverse research settings. METHODS: We used data from the SPAN (Stroke Preclinical Assessment Network), a randomized, placebo-controlled, blinded, multilaboratory trial, to determine if sex differences in neurological outcomes are present in preclinical stroke models. We analyzed da

A >8-year two-generation study on the long-term effects of genetically modified maize (Zea mays) containing cry1Ab/cry2Aj and EPSPS genes on cardiac function and arrhythmia risk of cynomolgus macaques.

The long-term safety of genetically-modified (GM) crops, particularly their effects on cardiovascular health, remains a subject of scientific debate. This study systematically evaluated the effects of GM maize (Zea mays) feed on cardiovascular function in cynomolgus macaques (Macaca fascicularis) through two-generation feeding over 8 years. A comprehensive assessment of effects of the GM diet on myocardial injury, cardiac function, and arrhythmia risk was conducted using cardiac-specific biomarker assays, echocardiography, surface electrocardiography (ECG), and implantable cardiac monitoring.

Activation of developmental transcription factors using RNA technology promotes heart repair.

Ischemic injury and adverse post-infarction myocardial remodeling are major causes of heart failure. We previously reported that microRNA (miR)-200c inhibition in murine embryos increased cardiogenic transcription factors (TFs) Tbx5, Gata4, and Mef2c to activate an immature cardiomyocyte cell state, suggesting miR-200 inhibition as a therapy for cardiac repair. We performed permanent ligation of the left anterior descending artery (a severe myocardial infarct, MI) on PMIS-miR-200c (inhibition of miR-200c; PMIS-C), PMIS-A (inhibition of miR-200a) and wildtype adult mice. Harnessing glucocorticoid receptor antagonism to enhance the efficacy of cardiac regenerative growth factors and cytokines.

Myocardial injuries lead to cardiomyocyte loss and heart failure. Endogenous glucocorticoids, via the glucocorticoid receptor (GR), limit cardiomyocyte regeneration. Here we show that glucocorticoids suppress mammalian (murine) cardiomyocyte proliferative response to regenerative growth factors and cytokines. GR activation in neonatal cardiomyocytes upregulated MAPK-ERK inhibitors ERRFI1 and DUSP1. Using neuregulin 1 as a model, we demonstrated that glucocorticoids inhibit growth-factor-induced ERK activation, nuclear translocation and transcriptional output. Errfi1 and Dusp1 knockdown restore

Pediatric Myocarditis: Challenges in Diagnosis and Treatment.

Pediatric myocarditis is a clinically diverse and often under-recognized inflammatory condition of the myocardium, with presentations that range from mild symptoms to acute heart failure. Advances in cardiac imaging, biomarker discovery, and molecular genetics have improved diagnostic precision and individualized care. Genetic studies indicate that 8-10% of pediatric patients with myocarditis carry pathogenic variants in cardiomyopathy-related genes, supporting the "double-hit" hypothesis where genetic predisposition interacts with viral or autoimmune triggers. This paradigm shift has undersco

South African Flag Sign in Left Anterior Descending Artery Occlusion Revealing Multivessel Coronary Artery Disease.

Early recognition of subtle or atypical electrocardiographic patterns of acute coronary occlusion remains a major challenge in contemporary practice. The "South African flag sign" (SAFS) is characterised by concomitant ST-segment elevation in leads I-aVL and in the non-contiguous lead V2, together with reciprocal ST-segment depression in lead III and often in the remaining inferior leads, resulting in a distinctive spatial distribution of injury currents in the 4x3 electrocardiogram format, reminiscent of the South African national flag. It represents a marker of mid-anterior/anterolateral tra

Periportal hepatocytes secret SAA1 to recruit MoKCs and interactions through CADM1 signaling to promote MASLD.

BACKGROUND: In non-alcoholic steatohepatitis, there is a compensatory increase in monocyte-derived Kupffer cells (MoKCs) due to the loss of resident Kupffer cells. However, the mechanisms behind MoKCs recruitment and their impact on steatohepatitis development remain unclear. We aimed to elucidate the contribution of continuously recruited MoKCs and the interaction between MoKCs and other cells in macrophage-mediated metabolic dysfunction associated steatotic liver disease (MASLD) to disease exacerbation. METHODS: We utilized snRNA-seq analysis parallel to characterize the transcriptomes of h

Cognitive Processing of Novel Auditory Stimuli in Children with Acute Disorders of Consciousness.

BACKGROUND/OBJECTIVE: Prognostication of critically ill children appearing acutely comatose remains challenging. Event-related potentials (ERPs) using electroencephalography (EEG) offer a noninvasive method to detect covert consciousness. These include the mismatched negativity (MMN) and P300b responses. We investigated ERPs using the auditory local-global oddball paradigm to assess inattentive or attentive cortical processing in critically ill, clinically unresponsive children. METHODS: We prospectively enrolled pediatric intensive care unit (PICU) patients undergoing continuous EEG with Gla
