Diagnostic performance of a dual western blot for the simultaneous detection of anti-HTLV-1 and anti-HIV-1 antibodies.

INTRODUCTION: There is no consistent definition of burnout. It is neither a defined diagnosis in ICD-10 nor in DSM-IV. Yet it is diagnosed by office-based doctors and clinicians. Mainly due to reimbursement reasons, diagnoses like depression are used instead of burnout diagnoses. Therefore burnout has a very high individual, social and economic impact. OBJECTIVES: How is burnout diagnosed? Which criteria are relevant? How valid and reliable are the used tools? What kind of disorders in case of burnout are relevant for a differential diagnosis? What is the economic effect of a differential diagn

Structural basis for Mg²⁺-glycine synergy in the tandem glycine riboswitch revealed by cryo-EM.

Part I of this article discussed the potential functional importance of genetic mutations and alleles of the human cytochrome P450 2D6 (CYP2D6) gene. The impact of CYP2D6 polymorphisms on the clearance of and response to a series of cardiovascular drugs was addressed. Since CYP2D6 plays a major role in the metabolism of a large number of other drugs, Part II of the article highlights the impact of CYP2D6 polymorphisms on the response to other groups of clinically used drugs. Although clinical studies have observed a gene-dose effect for some tricyclic antidepressants, it is difficult to establ

Genetic identification of zoonotic parasite Anisakis pegreffii (Nematoda: Anisakidae) parasitizing the shortfin squid Illex argentinus under commercial exploitation in the Southwestern Atlantic Ocean.

Despite the shortfin squid, Illex argentinus, is one of the most important commercial species for the Argentine fisheries, being the third frozen product exported to Europe, the occurrence and distribution of zoonotic anisakid nematodes is scarcely reported. A total of 712 I. argentinus distributed in 17 samples, corresponding to its three main commercial stocks, caught along its distribution range in Argentine waters were examined for anisakid parasites. In total, 360 nematodes were detected in the viscera, however no infestations in the mantle were observed. According to their morphology, al

Exploring the exoproteome of the parasitic nematode Anisakis simplex (s. s.) and its impact on the human host - an in vitro cross-talk proteomic approach.

INTRODUCTION: Anisakis simplex sensu stricto (s. s.) is one of the most widespread parasitic nematodes of marine organisms, with humans as accidental hosts. While many studies have explored nematode biology and host interactions, the role of extracellular vesicles (EVs) as signaling molecules in parasitic nematodes is less understood. MATERIALS AND METHODS: Therefore, the proteins present in the EVs of A. simplex (s. s.) (Anis-EVs) were identified. In addition, a cross-talk proteomic approach was used to identify differentially regulated proteins (DRPs) in the proteome of the human intestinal

Evaluation of the resistance of Anisakis spp. larvae to products regularly used in the industry and households.

The rise in popularity of raw or undercooked fish dishes like sushi, sashimi, and ceviche has led to an increase in human cases of anisakiasis. This study aimed to evaluate the resistance of Anisakis larvae to substances like bleach, salt, brine, and marinades. Batches of 10 larvae were exposed to various concentrations of commercial bleach (pure, 50 %, 25 %, 12.5 %, 6.25 %) and salt (dry, 360 g/L, 90 g/L, 45 g/L, 20 g/L). Larvae were also tested with two vinaigrettes (one with mustard and one without) and four marinades made with vinegar and salt. Sodium hypochlorite was highly effective, kil

IgE-Crosslinking-Induced Luciferase Expression Test as a Sensitive Indicator of Anisakis Allergy.

Background:Anisakis allergy has been increasing, and the diagnosis of it is based on specific serum IgE detection. Recently, the IgE-crosslinking-induced luciferase expression (EXiLE) test has been proposed as convenient tool for detecting functionally specific IgE antibodies. Here, we investigated if the EXiLE test is a useful tool in the diagnosis of Anisakis allergy. Methods: HuRa-40 cells were sensitized using six serum types from three patients with Anisakis allergy at the time of the initial test and after 6-12 months. Thereafter, various concentrations of Anisakis worm protein (AWP) wer

Anisakidae and Anisakidosis: A Public Health Perspective.

Fish and seafood are increasingly recognised as safe and nutritiously valuable foods of animal origin, being a source of about 17% of animal protein globally. Novel culinary trends encourage the consumption of raw or thermally lightly processed fishery products. At the same time, consumers prefer wild, fresh and whole fish over farmed or processed fish. However, the consumption of raw or undercooked fish and other marine organisms poses a risk of contracting parasitic infections, potentially representing a public health risk. Among the most common seafoodborne parasites are members of the Anis

First Insights from On-Board Fish Gutting into the Zoonotic Nematode Burden of Pouting (Trisopterus luscus) at the Point of Sale to the Consumer.

A survey was conducted to assess the impact of on-board gutting in the pouting fishery, Trisopterus luscus (L.), from the Bay of Biscay (area FAO 27.VIII) on the parasite burden of macroscopic ascaridoid nematodes, including anisakids (causing anisakidosis) and raphidascaridids (causing consumer rejection) in these fish. The fish were caught in the Bay of Biscay and collected from the fish market in Granada (southern Spain). Fish larger than 25 cm were gutted on board after capture. A detailed examination of the fish revealed the presence of nematode larvae, which were identified morphological

Rapid and Conventional Freezing Conditions of Fish for the Prevention of Human Anisakiasis.

In recent years, rapid freezers have been used to freeze and preserve seafood, with advances in freezing technology. However, limited studies have examined the effect of rapid freezing on the viability of Anisakis larvae in fish muscle. In this study, freezing experiments were conducted on Anisakis larvae alone (bare group) and on larvae embedded in mackerel fish (embedded group) using an air-blast freezer (rapid freezing) as the most popular rapid-freezing method, passing through the zone of maximum ice crystal formation within 30 min, and a natural convection freezer (conventional freezing)

A Rare Subepithelial Lesion of the Proximal Colon Evaluated Using Miniprobe Endoscopic Ultrasonography: A Case Report.

Preoperative diagnosis of submucosal lesions in the colon can be challenging. Total colonoscopy comprising miniprobe endoscopic ultrasonography (mEUS) revealed a 5-mm submucosal lesion in the transverse colon as a hypoechoic area primarily within the submucosa in our patient. However, a definitive diagnosis could not be made after evaluating the biopsy results. Based on the mEUS findings, adhesion to the muscularis was localized; therefore, endoscopic submucosal dissection was performed. A postoperative pathology examination revealed a parasite-like structure at the center of the necrotic tiss

Hybridization and introgression of the mitochondrial genome between the two species Anisakis pegreffii and A. simplex (s.s.) using a wide genotyping approach: evolutionary and ecological implications.

Anisakis pegreffii and A. simplex (s.s.) are the two zoonotic anisakids infecting cetaceans as well as pelagic/demersal fish and squids. In European waters, A. pegreffii prevails in the Mediterranean Sea, while A. simplex (s.s.) in the NE Atlantic Ocean. Abiotic conditions likely play a significant role in shaping their geographical distribution. The Iberian Atlantic and Alboran Sea waters are sympatric areas of the two species. A total of 429 adults and L3 stage from both sympatric and allopatric areas were studied by a wide nuclear genotyping approach (including newly and previously found di

Effects of wood creosote on anisakiasis: A Japanese traditional medicine as a potential tool against Anisakis larva infection.

Anisakiasis has become more widely known because of the increasing popularity of raw fish consumption worldwide; it is the most common form of food poisoning in Japan. This foodborne disease has no specific treatment. Current treatments involve detecting and removing parasites that penetrate the gastric mucosa using endoscopy in medical settings. The aim of this study was to investigate the effect of wood creosote (WC) on anisakid larval movement and viability in vitro and in vivo. Using two WC concentrations, 0.67 and 6.7 mg/mL, we performed in vitro and in vivo experiments to demonstrate the

An Assessment of the Currently Available Molecular Assay for the Diagnosis of Anisakis Sensitization.

The diagnosis of allergic reactions to Anisakis remains challenging due to the lack of specific allergens available for routine clinical use. However, the latest version of the multiplex macroarray ALEX-2 now allows the detection of specific IgE against Ani s 1, the major species-specific allergen, as well as Ani s 3 (tropomyosin), a highly cross-reactive molecule with homologs in other allergenic sources. This study aimed to evaluate the potential role of ALEX-2 in diagnosing Anisakis sensitization by comparing it with a previously validated diagnostic algorithm. Serum samples from patients w

Comparative study of Taqman-based qPCR assay for the detection of Anisakis simplex and Pseudoterranova decipiens.

Anisakidosis is a foodborne parasitic infection caused by the consumption of raw or uncooked seafood that contains third stage larvae from the Anisakidae family. This infection has been observed across the globe, with a particularly high prevalence in South Korea and Japan. Consequently, there is a necessity to compare and analyze the optimal detection methods with a view to preventing Anisakis outbreaks. In this study, a species-specific Taqman-based qPCR method was developed for the detection of the internal transcribed spacer region and mtDNA genes of Anisakis simplex and Pseudoterranova de
