Silybum marianum peptide-rich fraction alleviates acute liver injury by inhibiting TRAF3/NIK/NF-κB and is associated with changes in gut microbiota composition.

Acute liver injury (ALI) is a severe and prevalent condition that can progress to irreversible liver damage and failure. This study aimed to investigate the prophylactic hepatoprotective potential of Silybum marianum peptides (SMP) in ALI and elucidate its underlying mechanisms. Using a carbon tetrachloride (CCl4)-induced ALI mouse model, we evaluated the dose-dependent effects of SMP. Additionally, we explored the modulatory effects of SMP on the NF-κB pathway, gut microbiota composition, and metabolite profiles. The role of SMP in the TRAF3/NIK/NF-κB signaling axis was further validated in a

Glycochenodeoxycholate promotes IL-8 expression through ER-mitochondrial calcium signaling in human hepatocytes.

Accumulation of excessive bile acids in the liver contributes to the progression of liver injury; thus, elucidating the bile acid-induced responses remains a critical challenge. Although conjugated bile acids have recently been reported to induce inflammatory responses at non-cytotoxic concentrations, the precise molecular mechanisms involved are not fully understood. The present study investigated the role of intracellular calcium dynamics-focusing particularly on mitochondria-in bile acid-induced inflammatory signaling. Exposure of human hepatocytes isolated from humanized liver chimeric mic

Protective effects of Opuntia ficus-indica (L.) Mill flower extract against imidacloprid-induced hepatotoxicity: Modulation of oxidative stress, apoptosis, and Raf/MEK/ERK-related gene expression.

Imidacloprid (IMI), a widely used neonicotinoid insecticide, is associated with hepatotoxic effects. This study evaluated the protective potential of Opuntia ficus-indica (L.) Mill. flower extract against IMI-induced liver injury in rats. Animals were allocated into four groups: control, extract-treated, IMI-treated, and extract plus IMI. Hepatic damage was assessed using histopathology, biochemical oxidative stress markers, and gene expression analysis of apoptotic and Raf/MEK/ERK pathway markers. IMI exposure induced marked liver injury, oxidative stress, and apoptosis, while co-treatment wi

Inhibition of histone deacetylase 3 alleviates lithocholic acid-induced intrahepatic cholestasis by enhancing expression of bile acid and lipid homeostasis-related genes.

There is a lack of effective therapies for cholestatic liver injury. Recent studies have shown that pan-inhibitors of histone deacetylases (HDACs) play a protective role in cholestasis and fibrosis, but the dynamic changes and role of histone deacetylase 3 (HDAC3) under these conditions remain unclear. In the current study, the expression of acetylation-modifying enzymes, including HDAC3, in livers from lithocholic acid (LCA)-induced intrahepatic and bile duct ligation (BDL)-induced extrahepatic cholestatic mice, as well as in cholestatic patients, was examined. HDAC3 was aberrantly upregulate

Epinephrine-mediated sensitization of thioredoxin 1 enhances chlorpromazine protection against acute hepatotoxicity.

Acute liver injury (ALI) is characterised by rapid hepatic necrosis and compromised liver functions. Drug-induced ALI, in particular, is associated with glutathione depletion and overproduction of reactive oxygen species (ROS). These alterations highlight the contribution of the thioredoxin system (Trx/TrxR/NADPH) in preserving hepatic redox homeostasis. Previously, we demonstrated that thioredoxin-1 (Trx1) overexpression exerts protective effects against ischemic heart and peripheral vascular disease. Also, the neuroprotective role of thioredoxin reductase (TrxR1) has been reported in Parkins

Identification and validation of early vital sign trajectory subphenotypes for risk stratification in sepsis-associated liver injury: a multicenter retrospective cohort study.

BACKGROUND: Sepsis-associated liver injury (SALI) is a serious clinical syndrome with heterogeneity in outcome among affected individuals. Physiological changes that are critical to outcome prediction are not considered in the existing system of stratification based on single-point observations. This could compromise the effectiveness of individualized treatment. OBJECTIVE: To develop and validate dynamic clinical subphenotypes of SALI using early vital sign trajectories. METHODS: This retrospective cohort study included 1,159 patients from MIMIC-IV as the derivation cohort and 168 patients

Evaluation of large language models for ICI-related liver injury with an emerging microbiome focus.

BACKGROUND: Immune checkpoint inhibitor-related liver injury (ICI-LI) has heterogeneous phenotypes, a complex differential diagnosis, and evolving management. Gut microbiome research provides biological plausibility for immune regulation during ICI therapy, but evidence specific to ICI-LI remains limited and is not yet clinically actionable. We evaluated whether publicly accessible large language model (LLM) chatbots can organize this mixed evidence base and communicate its uncertainty. METHODS: We developed a disease-specific benchmark of 25 questions covering immunological mechanisms, micro

Based on the skeletal-muscle dual stem cell chemotaxis and bidirectional differentiation mechanism to reinterpret the "sinew-bone interdependence" theory.

The "sinew-bone interdependence" theory is a core paradigm in traditional Chinese medicine (TCM) orthopedics and traumatology. Rooted in the visceral manifestation doctrine of "the liver governing sinews and the kidney governing bones," this theory emphasizes the structural dependency, functional synergy, and pathological inter-transmission between sinews and bones. However, traditional interpretive pathways have largely remained at the level of descriptive deduction concerning anatomy, visceral manifestations, and biomechanics, failing to elucidate the cellular executors and molecular mechani

Effects of three doses of tea polyphenols on lipid metabolism, intestinal barrier function, and gut microbiota in overweight mice.

BACKGROUND: Overweight induced by a high-fat diet (HFD) is closely associated with dyslipidemia, oxidative stress, intestinal barrier dysfunction, and gut microbiota imbalance. Tea polyphenols have shown metabolic benefits; however, comparative information regarding their biological effects across different intervention doses remains limited. This study evaluated the effects of different doses of green tea polyphenols (GTP) on metabolic disorders in HFD-induced overweight mice. RESULTS: Green tea polyphenols reduced body weight gain and improved lipid metabolism without affecting food intake,

Enrofloxacin Disrupts Microbiota-Gut-Brain Axes in Marine Medaka: Mechanistic Insights from Multiomics and Germ-Free Models.

The widespread use of antibiotics has resulted in their frequent occurrence in marine environments, yet their ecological risks remain poorly understood. Enrofloxacin (ENR) is one of the most frequently detected antibiotics in aquatic ecosystems. However, its toxicological mechanisms in marine fish remain unclear. Here, multiomics, Vibrio parahemolyticus infection, short chain fatty acid supplementation and germ-free fish models were integrated to investigate ENR induced neurotoxicity and the role of gut microbiota. The results showed that ENR exposure impaired intestinal function in marine med

Precise immunomodulation by mesenchymal stem cells mitigates hepatitis B virus-specific CD8⁺ T cell-mediated acute liver injury without compromising viral clearance.

Hepatitis B virus-related acute-on-chronic liver failure (HBV-ACLF) is associated with a high mortality rate. Although mesenchymal stem cell (MSC) therapy markedly improves liver function and survival in patients with HBV-ACLF, it remains unclear whether the immunomodulatory effects of MSCs suppress antiviral CD8⁺ T cell function and trigger HBV reactivation. Here, we developed a novel in vitro HBV-specific CD8⁺ T cell-mediated acute liver injury organoid model, which was co-cultured with either menstrual blood-derived MSCs (MenSCs) or umbilical cord-derived MSCs (UC-MSCs). We found that both

Life-Threatening Aberrant Omental Variceal Bleeding with Interventional Treatment: A Case Report.

Ectopic varices resulting from portal hypertension can cause fatal bleeding. Intra-abdominal hemorrhage from omental varices is rare, and no previous reports have described a direct connection between the splenic and ovarian veins. We report a case of intra-abdominal hemorrhage caused by an omental varix in a 49-year-old women with liver cirrhosis. Because she was hemodynamically unstable, surgical treatment was not feasible; therefore, an interventional procedure was planned. The varices originated from the splenic vein and drained into the ovarian vein. Through percutaneous portal venous acc

Suspected fibrosing cholestatic hepatitis in an immunocompetent child with hepatitis C: Case report.

A 12-year-old boy presented to our center with elevated aminotransferases, jaundice, right upper quadrant tenderness, and hepatomegaly. Evaluation revealed a highly elevated blood hepatitis C virus (HCV) ribonucleic acid (RNA) level, and liver biopsy showed canalicular and intrahepatic cholestasis, feathery degeneration of hepatocytes, and periportal/perisinusoidal fibrosis. Though there were atypical biopsy features, including absence of bile ductular reaction, we diagnosed suspected fibrosing cholestatic hepatitis (FCH). FCH secondary to HCV infection has been reported exclusively in immunos

A phase II study combining radiotherapy and atezolizumab/nab-paclitaxel for inoperable or recurrent programmed death-ligand 1-positive triple-negative breast cancer: IMRad-Br study protocol and safety run-in.

BACKGROUND: Triple-negative breast cancer (TNBC) is an aggressive subtype lacking estrogen receptor, progesterone receptor, and human epidermal growth factor receptor 2 expressions, with poor prognosis and limited treatment options. Studies have demonstrated the efficacy of immune checkpoint inhibitors (ICIs) combined with chemotherapy in patients with programmed death-ligand 1 (PD-L1)-positive advanced TNBC. Radiotherapy (RT) may enhance therapeutic effects when combined with ICIs. METHODS: In this ongoing single-arm, multicenter phase II trial (IMRad-Br; jRCTs021210010), we aimed to evaluat

Adult intestinal malrotation: A rare incidental finding in trauma imaging.

Intestinal malrotation is a congenital anomaly resulting from abnormal rotation and fixation of the midgut during embryonic development. It is typically diagnosed in infancy, and its detection in adults-particularly in asymptomatic individuals-is rare. In adults, clinical presentation ranges from chronic, nonspecific symptoms to acute intestinal obstruction and midgut volvulus. A 36-year-old woman presented to the emergency department following a motor vehicle collision. She reported no gastrointestinal complaints and denied any history of recurrent abdominal pain, bilious vomiting, abdominal

Distinct collagen resolution dynamics in early versus advanced liver fibrosis: a quantitative SHG/TPEF study in mice.

BACKGROUND & AIMS: Liver fibrosis regression occurs after the source of injury is removed, but its dynamics and histologic features remain poorly characterized. Second-harmonic generation/two-photon excited fluorescence (SHG/TPEF) microscopy enables quantitative, high-resolution collagen assessment. We employed SHG/TPEF to characterize spatiotemporal collagen remodeling during spontaneous regression in carbon tetrachloride (CCl4)-induced mouse models. METHODS: Early- and advanced-stage liver fibrosis were established by intraperitoneal CCl4 injection for 8 or 12 weeks. After peak injury, mice
