Aqueous extract of Artemisia capillaris Thunb. herbal tea alleviates liver injury in MASLD mice and lipotoxicity in HepG2 cells by inhibiting apoptosis and promoting autophagy.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a chronic metabolic disorder characterized by diffuse hepatic steatosis. Excessive lipid accumulation induced by a high-fat diet can trigger hepatocyte apoptosis and impair autophagic flux, thereby aggravating liver injury. Artemisia capillaris Thunb. herbal tea contains multiple bioactive constituents with hepatoprotective, choleretic, and lipid-regulatory activities. Therefore, this study aimed to evaluate the protective effects of Artemisia capillaris Thunb. herbal tea aqueous extract (ACTE) against MASLD-associated hepatic

Stepwise Design of a High Signal-to-Background Ratio Dual-Emission Probe for Imaging Mitochondrial HClO and Microenvironmental Changes in Diabetic Liver and Kidney Injury.

Diabetic hepatorenal injury is linked to oxidative stress, mitochondrial dysfunction, and ferroptosis. Elevated hypochlorous acid (HClO) levels, together with alterations in mitochondrial polarity and viscosity, are key indicators of diabetic hepatorenal injury. Early, in situ, real-time monitoring of these signals is essential for disease assessment and therapeutic intervention. However, most existing fluorescent probes target only single analytes or organs, restricting multi-signal and dual-organ monitoring, while their imaging signal-to-background ratios (SBRs) remain suboptimal. Changes in the gut microbiome during fasting-induced molting and its effects on gut-liver function.

Fasting-induced molting (FIM) rejuvenates the laying cycle in hens; however, the fasting process may result in intestinal dysbiosis, which compromises host immunity and disrupts the intestine-liver function. Consequently, investigating the changes in gut microbiota during FIM and its impacts on gut-liver metabolic homeostasis is crucial for enhancing the efficiency of FIM in laying hens. Here, a total of 90 laying hens, aged 60 weeks, were selected for the FIM. Samples were collected at four time points: the day before fasting, the 15th day of fasting, the 5th day of refeeding, and the 47th da

Refinement of Structural Alerts for Hepatic Steatosis.

With the growing focus upon application of new approach methodologies (NAMs) within regulatory risk assessment of chemical substances, there is a clear need for human-relevant in silico models to predict toxicity associated with chemical exposure. This work describes the development of an in silico profiler for hepatic steatosis, a condition that results from the excess accumulation of fat within the liver. Toxicological data for chemicals were compiled from the literature, and expert judgments were made regarding their potential to induce steatosis. This list was used to test the 214 rules de

Ginsenoside Rb3 Combined with 20(S)-Protopanaxadiol Ameliorates Hypercholesterolemia via the Regulation of Lipid Metabolism and Gut Microbiota.

Hypercholesterolemia is a major contributor to cardiovascular disease and imposes a serious global disease burden. The stems and leaves of Panax notoginseng, as edible parts of the plant, are rich in ginsenoside Rb3. However, the lipid-lowering effects and mechanism of ginsenoside Rb3 remain unclear, which limits the utilization of Panax notoginseng stems and leaves. Our results showed that ginsenoside Rb3, particularly with PPD, inhibited cholesterol absorption, enhanced LDLR-mediated LDL uptake, reduced body weight, alleviated dyslipidemia, and attenuated liver injury and lipid accumulation.

Hyperoside Derived from Actinidia arguta Bee Pollen Alleviates Zearalenone-Induced Hepatotoxicity and Oxidative Stress in Piglets via the PI3K/AKT Signaling Pathway.

Zearalenone (ZEA) is a prevalent mycotoxin inducing hepatotoxic effects. Hyperoside (HYP), a flavonoid derived from Actinidia arguta bee pollen, exhibits hepatoprotective properties against ZEA; however, the molecular mechanisms underlying this protection remain incompletely characterized. This study integrated in vivo piglet models, in vitro porcine hepatocytes, network pharmacology, and molecular docking to characterize the molecular targets of HYP. The results demonstrated that HYP significantly attenuated ZEA-induced hepatic lesions and oxidative stress. Computational analyses pinpointed t

In Situ Visualization of Tumor and Drug-Induced Liver Injury with AIEgens, Polarity-Responsive, Membrane-Targeting Fluorescent Probes.

Dynamic changes in the polarity of the cellular microenvironment play a pivotal role in drug-induced liver injury (DILI) and tumors, serving as a crucial biomarker for evaluating cell membrane integrity and functional status. Precise monitoring of polarity changes is essential for elucidating the underlying mechanisms of injury. However, the real-time and specific visualization of local polarity remains a considerable challenge. Herein, polarity-sensitive fluorescent probes Probemem-1 and Probemem-2 were successfully developed. The structures adopted a D-π-A configuration, where a triphenylami

Hepatic-Renal Cascade Clearable Aminopeptidase N Probe for Rapid Detection of Early-Stage Liver Disease.

Early liver disease is a prolonged process with vague symptoms and evolving pathological causes, leading to a delayed diagnosis and poor therapeutic outcomes. The development of rapid and effective approaches for liver disease detection in both preclinical research and clinical diagnosis is in high demand yet remains challenging. Herein, we report an activatable fluorogenic probe (CyP-A) that is cleared via a hepatic-renal cascade for monitoring of hepatocyte injury and tissue fibrosis. After systemic administration, CyP-A preferentially accumulates in the liver, where it undergoes APN-mediate

Magnetic field exposure and liver disease: from current research to future directions.

As both an environmental factor and a therapeutic modality, magnetic field (MF) exposure exerts a significant regulatory effect on liver physiology and pathology. This review systematically synthesizes the current body of evidence regarding the modulatory roles of MFs in liver diseases (LDs) and elaborates on the underlying potential mechanisms. Initially, the biological basis of the interaction between MFs and hepatic tissue is delineated. Subsequently, preclinical evidence is consolidated, which demonstrates that MFs with specific parameter settings exhibit protective or therapeutic potentia

Hepatic Orosomucoid 2 aggravates demyelination and drives microglial DAM-like activation.

INTRODUCTION: Demyelination is a central pathological feature of white matter injury (WMI). Emerging evidence suggests that hepatokines act as systemic regulators of brain pathology, raising the possibility that liver-derived signals contribute to WMI. Here, we investigated whether hepatokine Orosomucoid 2 (ORM2) aggravates demyelination through liver-brain communication by modulating disease-associated microglia (DAM)-like activation in WMI. METHODS: ORM2 expression was assessed in the cuprizone (CPZ) demyelination model using RNA sequencing, quantitative PCR, western blotting, and immunoflu

c-Jun N-Terminal Kinase-2 Mediates Alcohol and Corticosterone-Induced Tissue Injury at the Gut-Liver Axis.

Chronic stress and the stress hormone corticosterone are exacerbating factors in alcohol-induced gut and liver injury; however, the underlying mechanisms are poorly defined. This study investigates the potential role of c-Jun N-terminal kinase 2 (JNK2) in alcohol- and corticosterone-mediated gut and liver injury. The effects of chronic ethanol feeding and corticosterone administration on gut and liver injury were evaluated in JNK2 knockout (JNK2-KO) and wild-type mice. We measured gut permeability to FITC-inulin in vivo; epithelial tight junction (TJ) integrity by immunofluorescence confocal m

Saroglitazar protects against cadmium-induced hepatorenal injury in rats by suppressing NF-κB and upregulating Nrf2.

INTRODUCTION: Cadmium (Cd) toxicity is one of the major public health problems because of its long half-life (10-30 years). Exposure to Cd can cause toxicity that affects several organs, including the liver and the kidney. Saroglitazar is highlighted in this study as a potential preventative medication against hepatic-renal damage induced by Cd in rats. METHODS: Cd (3 mg/kg) was injected for five successive days to induce hepatic and renal damage. Saroglitazar (2 and 4 mg/kg/day) was given orally for five successive days concurrently with Cd. RESULTS: Saroglitazar effectively improved hepati

CO-mediated restoration of mitochondrial redox homeostasis contributes to alleviating multiple programmed cell deaths in hepatic I/R injury.

Hepatic ischemia-reperfusion injury (HIRI) remains a life-threatening complication in liver surgery and transplantation with no clinically approved therapy. Here we develop a hyaluronic acid-modified, ROS-responsive nanoformulation (CPs@HA) that enables controllable delivery of carbon monoxide (CO) specifically to injured liver tissue. We show that mitochondrial dysfunction is closely associated with three distinct cell death pathways (ferroptosis, cuproptosis, and disulfidptosis) that occur in parallel during HIRI. CPs@HA restores mitochondrial homeostasis through activating the AMPK-SIRT1-PG
