Betaine Mitigates Paclitaxel-Induced Hepatotoxicity in Rats by Regulating Oxidative Stress, Inflammation, Apoptosis, and Autophagy.

Paclitaxel (PTL) is widely used in chemotherapy; however, its efficacy is compromised by the risk of liver toxicity. This is the first study to investigate the protective effect of betaine (BTN) against PTL-induced liver injury. Rats were randomly divided into five experimental groups (n = 7 per group): control (saline), BTN (100 mg/kg/day), PTL (2 mg/kg/day), PTL + BTN (2 and 50 mg/kg/day), and PTL + BTN (2 and 100 mg/kg/day). After the rats received 2 mg/kg body weight PTL intraperitoneally for the first five consecutive days, BTN was administered orally for 10 days. Our results indicate tha

Innate immune cell LXR-β deficiency exacerbates hepatic injury and fibrosis in murine models of primary sclerosing cholangitis.

BACKGROUND AND AIMS: Primary sclerosing cholangitis (PSC) is an autoimmune liver disease characterized by complex pathogenesis and limited available therapeutic options. The mechanisms underlying the development and progression of PSCs remain unclear. Liver X receptor beta (LXR-β) is recognized to modulate lipid metabolism and immune response, but its specific involvement in the PSC has not been elucidated. Here, we explored the role and mechanism of LXR-β in PSC induced by 3, 5-diethoxycarbonyl-1, 4-dihydro-2, 4, 6-collidine (DDC). METHODS: CRISPR-Cas9 technology was applied to generate Abcb

Pathogenesis of metabolic dysfunction-associated steatotic liver disease and donor liver damage.

Metabolic dysfunction-associated steatotic liver disease (MASLD) affects more than a quarter of adults worldwide. MASLD is associated with severe medical burdens and leads to further reduction of the donor pool for liver transplantation. However, there is a lack of systematic evaluation of the pathogenesis of MASLD development and MASLD graft damage. As a multisystem disorder, the pathogenesis of MASLD is closely related to genetics, metabolic and endocrine disorders, imbalanced intestinal flora, abnormal hepatocyte homeostasis, and hepatic inflammation. Mutations or single nucleotide polymorp

Metabolic stress-induced dysregulation of the Hippo signaling pathway inhibits the occurrence of hepatocarcinoma.

Liver fibrosis is the natural stress response of the liver to injury and a critical intermediate stage in the progression of most liver diseases. Here, we first demonstrated via a retrospective clinical study that the incidence of advanced hepatocellular carcinoma (HCC) was significantly higher in patients infected with hepatitis B virus (HBV) than in those with hepatolenticular degeneration (HLD). Further analyses involving micro RNA (miRNA) and proteomics were conducted to investigate the distinct mechanisms underlying liver fibrosis induced by HBV and HLD. Results showed significant changes

Cytoglobin suppresses oxidative damage and compensatory proliferation via inhibiting AKT/ERK1/2/CyclinD1 axis in hepatocellular carcinoma.

The chronic liver injury caused by diverse etiologies provokes abnormal compensatory proliferation and contributes to hepatocarcinogenesis. Oxidative stress (OS) is an independent etiology which induces cell injury and triggers oncogenes activation and tumor suppressor genes dysfunction. Cytoglobin (CYGB) is a member of the human hexacoordinate hemoglobin (HHB) family and functions as a dynamic antioxidant enzyme. We previously reported that CYGB decreases the intracellular ROS/RNS level and inhibits HCC proliferation and stemness under normal oxidative level. In this study, we observed that C

The mutational landscape and actionable targets of gallbladder cancer: an ancestry-informed and comparative analysis of a Chilean population.

INTRODUCTION: Gallbladder cancer (GBC) is a highly aggressive malignancy with one of the highest incidence rates reported in Chile. Despite its clinical impact, molecular characterization of GBC in Latin American populations remains limited, and the absence of effective targeted therapies underscores the urgent need for new therapeutic strategies. METHODS: We collected 118 tumor samples, of which 56 passed sequencing quality control using the Oncomine™ Comprehensive Assay v1. Somatic variants were identified with ANNOVAR and Cancer Genome Interpreter, and ancestry was inferred using ADMIXTURE

Hydroxytyrosol Improves Metabolic Dysfunction-Associated Fatty Liver Disease Dependent on the Modulation of Gut Microbiota.

The global threat of metabolic dysfunction-associated fatty liver disease (MAFLD) is significant, but effective measures are still lacking. To explore the potential impact of hydroxytyrosol (HT), a plant polyphenol, in the metabolic outcomes of MAFLD and the mediating role of the gut microbiota, we performed an 8-week randomized placebo-controlled clinical trial in MAFLD patients and collected fecal bacteria for metagenomics analysis and targeted metabolomics. In this population-based trial, we have revealed that HT mitigates liver injury and steatosis in patients with MAFLD, as well as system

A microfluidic liver-like model for antiepileptic drugs hepatotoxicity evaluation.

Drug-Induced Liver Injury (DILI) is the leading cause of late-stage drug failure and withdrawal of medications from the market. This underscores the urgent need for newin vitroliver models that better recapitulate physiologically relevant conditions and provide controlled physical and biochemical parameters for drug toxicity testing. While advanced, complex liver-on-a-chip systems have been in use for some years, this study presents a simplified microfluidic model for technically comparing static and dynamic culture conditions using a hepatocyte-like cell line for drug toxicity testing. Roles of the Keap1/Nrf2 pathway and mitophagy in liver diseases.

Nuclear factor erythroid 2-related factor 2 (Nrf2) is an intracellular transcription factor that helps protect against oxidative stress in different types of cells under pathological conditions. Mitochondria are vital organelles that function in diverse metabolic processes in the body, including redox reactions, lipid metabolism, and cell death. Mitophagy, a specific form of autophagy for damaged mitochondria, plays a critical role in the pathophysiology of liver diseases. In this review, we explain in detail the roles of the Nrf2 signaling pathway and mitophagy, and the relationship between t

The Protective Effect of Geraniol Against Hepatic Ischemia-Reperfusion Injury by Attenuating Oxidative Stress, Inflammatory Response, and Apoptosis in Rat Model.

BACKGROUND: Hepatic ischemia-reperfusion injury (HIRI) is one of the main causes of hepatic fibrosis that occurs during liver surgery. This study aimed to investigate the protective effect of geraniol (GNL) against HIRI in a rat model. METHODS: Wistar rats were randomly divided into seven groups and subjected to 45 min of hepatic ischemia, followed by either 60 min or 6 h of reperfusion. Immediately before reperfusion, graded doses of geraniol (50 and 100 mg/kg) were administered intraperitoneally. Serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were measur

The Cannabinoid System as a Potential Novel Target for Alcohol-Associated Liver Disease: A Propensity-Matched Cohort Study.

BACKGROUND: Alcohol-associated liver disease (ALD) is a leading cause of liver-related morbidity and mortality, yet effective therapeutic options remain limited. Preclinical data suggest that modulation of the hepatic endocannabinoid system, particularly via cannabidiol (CBD), may reduce alcohol-induced liver injury. Due to CBD's limited clinical use, we sought to evaluate the association between cannabis use and ALD risk among patients with alcohol use disorder (AUD). METHODS: Using the TriNetX US Collaborative Network, we identified adult patients with AUD between 2010 and 2022. Three cohor

Molecular phenotypes in early gallbladder and liver disease in the CFTR-/- sheep highlight aspects of cystic fibrosis-relevant hepatobiliary disease.

The initiating cellular events in cystic fibrosis (CF) hepatobiliary disease are not well characterized, in part due to the lack of accessibility of primary tissues. However, enhanced longevity due to highly effective modulator therapies has generated renewed interest in the key aspects of liver and gallbladder disease, and how these might be treated in people with cystic fibrosis (pwCF). To extend the CF hepatobiliary knowledge base, we performed a transcriptomic analysis of liver and gallbladder development in the wild-type (WT) and CFTR-/- sheep. Bulk RNA was extracted from each tissue at s
