Inferior functional sensory regeneration after suture of sciatic neurotomy in newborns compared with mature rats.

It is generally believed that nerve injuries in children regenerate better than those which occur in adults. However, there are no functional experimental studies that support this belief. This study evaluates the functional regeneration of polymodal C-fibres after nerve regeneration in newborn and mature rats 3 months after unilateral sciatic nerve neurotomy and suture. The distribution of polymodal C-fibres was tested by measuring the Evans blue-stained area in the skin after antidromic nerve stimulation. In the newborn group of regenerated animals showed that functional C-fibres were presen

Effect of CO2 milliwatt laser on peripheral nerves: Part I. A dose-response study.

In order to explore further the role of laser for microneural repair, the effect of CO2 laser irradiation on intact rat sciatic nerves was investigated. In total 40 rat sciatic nerves were exposed to 12 different combinations of laser power (50, 100, and 150 mW) and pulse duration (0.1 to 3 s) normally used for CO2 laser-assisted nerve repair. The results were evaluated 24 hr after surgery with functional toe-spreading test and light microscopy. Irradiations of 50 and 100 mW for up to 1 s exposure time per pulse resulted in almost no deficit in motor function, while 100 mW power with prolonged

Regeneration of putative sensory and sympathetic cutaneous nerve endings in the rat foot after sciatic nerve injury.

The present study examines the occurrence of calcitonin gene-related peptide-, substance P- and tyrosine hydroxylase-like immunoreactive profiles in glabrous and hairy foot skin from normal and nerve-injured rats. After neurotomy/suture, glabrous skin samples contain few calcitonin gene-related peptide-, substance P- and tyrosine hydroxylase-like immunoreactive profies. The number of calcitonin gene-related peptide- and substance P-like immunoreacive profiles in the epidermis is significantly subnormal. Hairy skin from these rats does also contain few calcitonin gene-related peptide-, substanc

Expression of type I and III collagen and laminin beta1 after rat sciatic nerve crush injury.

Extracellular matrix changes are thought to be essential to the regeneration of peripheral nerves. The production of this matrix is believed to be regulated by interactions between axons and their supporting cells. In this study matrix production and cell proliferation were studied during rat sciatic nerve regeneration after a crush injury, and compared to that after rat sciatic nerve transection. Expression of proalpha1(I) and proalpha1(III) collagen and laminin beta1 mRNAs was followed in isolated endoneuria by Northern and in situ hybridization both proximally and distally to the site of ei

Experimental surgery in spinal cord lesions by connecting upper motoneurons directly to peripheral targets.

This research was aimed at assessing the possibility to connect central motoneurons with skeletal muscles through PNS segments bypassing a lesion of the spinal cord. The investigation was performed in 20 non-human primates (Macaca fascicularis). The surgical paradigm consisted of anastomosing the lateral bundle of the spinal cord directly with the sciatic nerve of the right hindlimb, using the peroneal nerve as a graft. The animals were followed-up clinically for 18 months; at the end of this observation period, they underwent electrophysiological examinations before being killed. Specimens we

Effects of intraoperative irradiation and intraoperative hyperthermia on canine sciatic nerve: neurologic and electrophysiologic study.

PURPOSE: Late radiation injury to peripheral nerve may be the limiting factor in the clinical application of intraoperative radiation therapy (IORT). The combination of IORT with intraoperative hyperthermia (IOHT) raises specific concerns regarding the effects on certain normal tissues such as peripheral nerve, which might be included in the treatment field. The objective of this study was to compare the effect of IORT alone to the effect of IORT combined with IOHT on peripheral nerve in normal beagle dogs. METHODS AND MATERIALS: Young adult beagle dogs were randomized into five groups of thr

Intact female rats are more susceptible to the development of tactile allodynia than ovariectomized female rats following partial sciatic nerve ligation (PSNL).

As previously reported, this laboratory has determined that female rats are more prone to develop tactile allodynia, an indicator of neuropathic pain, than male rats using the partial sciatic nerve ligation (PSNL) model. In order to further characterize this gender difference, the role of ovarian hormones in predisposing female rats to the development of tactile allodynia was investigated. In a double blind randomized trial, 12 intact and 12 ovariectomized (ovx) female rats underwent PSNL of the right rear leg. Sham 1 operated (exposure of the nerve without ligation; 2 intact, 2 ovx) and sham

Influence of inflammation or disconnection from peripheral target tissue on the capsaicin sensitivity of rat dorsal root ganglion sensory neurones.

Dorsal root ganglion (DRG) sensory neurones from adult rats are known to lose their capsaicin sensitivity in vitro if they are cultured without nerve growth factor (NGF). Here we show similar results following peripheral nerve transection in vivo, which deprives DRG sensory neurones of target-derived NGF. By measuring capsaicin-stimulated 45Ca uptake into DRG neurones which had been briefly cultured, capsaicin sensitivity was shown to decrease in neurones whose axons had been previously severed in vivo. Conversely, during experimental inflammation of the rat paw, there is an increase in the su

Axons regulate the distribution of Schwann cell microtubules.

It is well established that axons regulate Schwann cell phenotype. The purpose of the present study was to determine whether axons influence the arrangement of Schwann cell microtubules (MTs). Using double-labeling immunocytochemistry and confocal microscopy, we show that MTs in undifferentiated Schwann cells are nucleated from and attached to a single MT organizing center (MTOC) that is associated with the centrosome. Physical contact with appropriate axons initiates a myelin-forming phenotype that disperses MT minus ends and induces multiple MT-nucleating sites in Schwann cell perinuclear cy

Characterization of insulin-like growth factor-I and its receptor and binding proteins in transected nerves and cultured Schwann cells.

The insulin-like growth factors (IGFs) are trophic factors whose growth-promoting actions are mediated via the IGF-I receptor and modulated by six IGF binding proteins (IGFBPs). In this study, we observed increased transcripts of both IGF-I and IGF-I receptor after rat sciatic nerve transection. Schwann cells (SCs) were the main source of IGF-I and IGFBP-5 immunoreactivity until 7 days after nerve transection, when invading macrophages in the distal nerve stumps were strongly IGF-I positive. In vitro, IGF-I promoted SC mitogenesis. Northern analysis revealed that SCs expressed IGF-I receptor a

Multiexponential T2 relaxation in degenerating peripheral nerve.

The multiexponential T2 relaxation spectrum of peripheral nerve undergoing Wallerian degeneration has been measured both in vivo and in vitro. Degeneration of the sciatic nerve of the amphibian Xenopus laevis was induced by crush injury, and T2 relaxation spectra of the nerve were measured at several times up to 35 days following injury. Histologic evidence verified that the nerve underwent Wallerian degeneration. Relaxation spectra were observed to undergo measurable changes as degeneration progressed, the most evident being a reduction from three well-resolved T2 components to one and a decl

Basic behavior of migratory Schwann cells in peripheral nerve regeneration.

In axonal regeneration after a peripheral nerve injury, Schwann cells migrate from the two nerve ends and at last form a continuous tissue cable across the gap which guides the axons toward the bands of Bungner. However, the behavior of migratory Schwann cells and their possible role are obscure. Using a film model in which the proximal stump of a transected nerve in mice was sandwiched between two thin plastic films, we analyzed neural regeneration in the early phase up to the 6th day after axotomy. Regenerating neurites emerged from the nodes of Ranvier adjacent to the axotomized nerve stump

Full-lenth cloning, expression and cellular localization of rat plasmolipin mRNA, a proteolipid of PNS and CNS.

We have isolated a 1.476 bp cDNA (NTII11) representing a transcript that is differntially expressed during sciatic nerve development and regeneration in the rat. Nucleotide sequence comparison indicates partial identity with a recently isolated plasmolipin cDNA. However, our clone extends the published sequence by 234 bp at the 5' end and predicts a protein that contains an additional 25 amino acids at th N-terminus. The open reading frame of th NTII11 transcript encodes a 19.4 kDa protein with four putative transmembrane domains. Northern blot analyses revealed a tissue-specific expression wa

Recent advances in tissue synthesis in vivo by use of collagen-glycosaminoglycan copolymers.

Biologically active analogues of the extracellular matrix (ECM) are synthesized by grafting glycosaminoglycan (GAG) chains onto type I collagen, and by controlling the physicochemical properties of the resulting graft copolymer. Collagen-GAG ECM analogues have previously been shown to induce regeneration of the dermis in humans and the guinea pig, and of the rat sciatic nerve. Current studies have emphasized elucidation of the molecular mechanism through which tissue-specific ECM analogues induce regeneration. The contribution of the GAGs to the biological activity of the skin regeneration tem

Dynamic changes of mechanically activated channels and K+ channels at injury site of peripheral nerve in rat.

Ectopic ion channels developed locally at the injury site after nerve damage by light ligation around common sciatic nerve of the rats. Different channel types have different processes of formation, accumulation and degeneration. During the first three days after injury, mechanically activated channels that are modulated by CA++ channel activities first appeared. As the nerve fibers begin to be excited by TEA, a blocker of K+ channels, suggesting that the accumulation of K+ channels, the responsibility of mechanically activated channels was declining. Onset of K+ channels was from the 3rd post

Regional anesthesia in knee arthroscopy: a new technique involving femoral and sciatic nerve blocks in knee arthroscopy.

We describe a new anesthetic technique of femoral and sciatic nerve blocks in knee arthroscopy. The sciatic nerve block is administered through a posterior approach, and the block of the femoral and lateral femoral cutaneous nerves in a single anterior approach. This type of regional anesthesia is safe and effective, providing excellent intraoperative and postoperative analgesia as well as minimizing postoperative complications.

The axonal transport of beta III-tubulin is altered in both branches of sensory axons after injury of the rat sciatic nerve.

We have analyzed the axonal transport of beta III-tubulin in the central (dorsal root) and peripheral (sciatic nerve) branches of sensory axons after injury of the sciatic nerve. Our finding that the relative amount of beta III-tubulin transported in slow component b (SCb) is increased in both axonal branches does not support the generally accepted hypothesis that the transport of cytoskeletal proteins is altered in the peripheral, but not the central branch after injury of the sciatic nerve.

Peripheral nerve elongation by laser Doppler flowmetry-monitored expansion: an experimental basis for future application in the management of peripheral nerve defects.

Nerve grafting often fails to achieve optimal functional results and is always associated with donor-site morbidity. Peripheral nerve elongation by the use of a tissue expander may provide a useful adjunct in the management of segmental nerve loss. In the present study, rabbit sciatic nerve (n = 40) was elongated by expansion while nerve blood flow was monitored by laser Doppler flowmetry. Elongation was possible up to 40 percent with preservation of clinical nerve function. Nerve conduction velocity of the expanded nerves decreased in a linear relation to elongation. The reduction in nerve co

The effect of indirect injury to peripheral nerves on wound healing after firearm wounds.

This paper reports the pathologic and functional changes of the sciatic nerve indirectly injured by firearms and its effect on wound healing. Twelve dogs were used. Their hind legs were shot with 1.03-gram steel spheres, avoiding direct injury to the sciatic nerve. The experimental animals were divided into two groups according to the impact velocity, that is, a 1,500 m/sec high velocity projectile (HVP) group and a 400 m/sec, low velocity projectile (LVP) group as a control. After wounding, the same procedure was followed in all dogs. At intervals, the tissues of the wound track were excised
