Fatty acid binding protein is induced in neurons of the dorsal root ganglia after peripheral nerve injury.

Peripheral nerve trauma induces the expression of genes presumed to be involved in the process of nerve degeneration and repair. In the present study, an in vivo paradigm was employed to identify molecules which may have important roles in these processes. A cDNA library was constructed with RNA extracted from rat dorsal root ganglia (DRG) 3 days after a sciatic nerve crush. After differential hybridization to this library, several cDNAs were identified that encoded mRNAs that were upregulated in the DRG ipsilateral to the crush injury, as opposed to the contralateral or naive DRG. Approximate

Pre-emptive dynorphin and N-methyl-D-aspartate glutamate receptor antagonism alters spinal immunocytochemistry but not allodynia following complete peripheral nerve injury.

The development of chronically painful states following peripheral nerve injury may involve different mechanisms depending on the nature and extent of the nerve lesion. The altered spinal neurochemistry of two substances, the excitatory amino acid glutamate operating via the N-methyl-D-aspartate receptor and the endogenous opioid peptide dynorphin, have been implicated in behavioral sequelae that follow partial peripheral nerve injury. In addition, dynorphin has nonopioid functions which may involve the N-methyl-D-aspartate receptor. We investigated two hypotheses: that the development of mech

Prosaposin facilitates sciatic nerve regeneration in vivo.

Prosaposin, a multifunctional protein, is the precursor of saposins, which activate sphingolipid hydrolases. In addition to acting as a precursor for saposins, prosaposin has been shown to rescue hippocampal CA1 neurons from lethal ischemic damage in vivo and to promote neurite extension of neuroblastoma cells in vitro. Here we show that prosaposin, when added to a collagen-filled nerve guide after sciatic nerve transection in guinea pigs, increased dramatically the number of regenerating nerve fibers within the guide. To identify the target neurons of prosaposin during peripheral nerve regene

Localization of synapsin I in normal fibers and regenerating axonal sprouts of the rat sciatic nerve.

The localization of synapsin I, a synaptic vesicle-associated protein, was investigated immunocyto-chemically in normal nerve fibers and regenerating axonal sprouts following crush-injuries to the rat sciatic nerve. In normal myelinated axons, weak synapsin I immunoreactivity was found in the axoplasmic/smooth endoplasmic domains, but not in the cytoskeletal domains comprising neurofilaments and microtubules. In non-myelinated axons without dense cytoskeletal structures, moderate immunoreactivity was distributed diffusely throughout the axoplasm. In the crush-injured nerves, intense synapsin I

Spontaneous action potential activity in isolated dorsal root ganglion neurons from rats with a painful neuropathy.

One of the physiological changes accompanying neuropathic pain from nerve injury is the spontaneous firing of primary afferent fibers. At least some of this activity is thought to arise from the dorsal root ganglion. We have investigated whether this activity is resident in the cell bodies of dorsal root ganglion neurons and if it is retained in vitro. Dorsal root ganglion neurons from rats with a chronic constriction injury (CCI) from 4 loose ligatures of chromic gut sutures around the sciatic nerve were used. Isolated neurons were studied using the whole cell patch technique in current clamp

[Different proteins in NRCF (nerve regeneration conditioned fluid) from the proximal and distal nerve stump].

80 SD rats were randomly divided into four groups of 20 each; electrostimulation of myeloneure (Group N); electrostimulation of the denervated muscle (Group M); Group C1; and Goup C2. The left lateral sciatic nerve of rats was exercised 2mm in length, and the distal nerve stump of the goup C2 and M and proximal nerve stamp of the Group C1 and N were each sulured into the silcon chamber with the opposile end ligated so that a 10mm was left between the stump and the blind end of chamber. At 3rd, 7th, 14th, 21th and 28th day after nerve transection and tube implantation, the fluid in the silicon

Ca2+/calmodulin-dependent protein kinase type IV in dorsal root ganglion: colocalization with peptides, axonal transport and effect of axotomy.

Using the indirect immunofluorescence technique, the distribution of Ca2+/calmodulin-dependent protein kinase IV (CaM kinase IV) was studied in dorsal root ganglia (DRGs) and the sciatic nerve under normal circumstances and after axotomy and nerve ligation. CaM kinase IV-like immunoreactivity (-LI) was observed mainly in small DRG neurons but also in some large ones with the immunoreactivity mainly confined to the cell nuclei and with varying levels in the cytoplasm. CaM kinase IV-LI was present in around 1/4 of all CGRP-positive neurons and in the vast majority of the somatostatin-positive ne

Application of nitric oxide synthase inhibitor, N omega-nitro-L-arginine methyl ester, on injured nerve attenuates neuropathy-induced thermal hyperalgesia in rats.

This study tested the ability of a nitric oxide synthase inhibitor, N omega-nitro-L-arginine methyl ester (L-NAME), to attenuate behavioral hyperalgesia in a rat model of neuropathic pain [Bennett, G.J. and Xie, Y.-K., Pain, 33 (1988) 87-107]. A mononeuropathy was produced by chronic constriction injury (CCI) of the sciatic nerve. Thermal hyperalgesia was assessed by a reduction of paw withdrawal latency to a noxious heat source. Following CCI, there was significant hyperalgesia in groups of rats treated with D-NAME (n = 7), an inactive isomer of L-NAME, saline (n = 7) or systemic L-NAME (n =

The recovery of 5-HT immunoreactivity in lumbosacral spinal cord and locomotor function after thoracic hemisection.

To determine the role of serotonin (5-HT) in recovery from spinal cord injury, we examined spinal cord 5-HT immunohistologically and assessed locomotor recovery after thoracic (T8) spinal cord hemisection in 68 rats. Forty eight rats had laminectomy and hemisection, while the remaining 20 rats received laminectomy only. All rats were evaluated every other day for 4 weeks, using a 0-14 point scale open field test. Hemisection markedly reduced mean hindlimbs scores from 14 to 1.5 +/- 0.32 and 5.6 +/- 0.31 (mean +/- standard error of mean) in the ipsilateral and contralateral side, respectively.

Tubulization increases axonal outgrowth of rat sciatic nerve after crush injury.

It is important to develop methods which increase nerve regeneration since restoration of function following injury to peripheral nerves often requires outgrowth of the injured axons over long distances. In this study, axonal outgrowth after bilateral crush injury to the sciatic nerve of the rat was measured. One group with large-diameter nonpermeable silicone tubes and one group with large diameter permeable silicone tubes applied around the crush site on one side had regeneration following nerve injury compared to controls on the other side. The length of regeneration of the regenerating axo

Effect of hyperbaric oxygenation on peripheral nerve regeneration in adult male rabbits.

Oxygen environments were used to study the regenerative effects of hyperbaric oxygen on crushed sciatic nerves in 30 adult male rabbits. Six different oxygen environments were used, and treatments were initiated 4 days post injury. Transmission electron microscopy and light microscopy were used to evaluate the regenerative morphology of crushed nerves. The morphology of crushed nerves after 7 wk of treatment with compressed oxygen at 202, 242, and 303 kPa resembled normal uncrushed nerves, with nerve fibers uniformly distributed throughout the section. The treatment groups receiving 202 kPa co

TNF alpha inhibits Schwann cell proliferation, connexin46 expression, and gap junctional communication.

Schwann cell responses to nerve injury are stimulated, in part, by inflammatory cytokines. This study compares changes in the phenotype of cultured Schwann cells after exposure to the cytokine tumor necrosis factor (TNF)-alpha or the mitogen neu differentiation factor (NDF)-beta. TNF alpha inhibited proliferation in a dose-dependent manner without altering Schwann cell survival. TNF alpha also reduced both gap junctional conductance and Lucifer yellow dye coupling between Schwann cells. Moreover, both Po and glial fibrillary acidic protein (GFAP) immunoreactivity were reduced. By contrast, NDF

Altered connexin expression after peripheral nerve injury.

The identification of connexin32 (Cx32) in myelinating Schwann cells and the association of Cx32 mutations with peripheral neuropathies suggest a functional role for gap junction proteins in the nerve. However, after nerve crush injury, Cx32 expression dramatically decreases in Schwann cells in the degenerating region, returning to control levels at newly formed nodes of Ranvier and Schmidt-Lantermann incisures by 30 days. The present study examined increases in expression of other connexins that occur after peripheral nerve injury. A 56/58-kDa connexin46 (Cx46) protein species was detected in

Sciatic nerve section induces mechanical hyperalgesia in skin adjacent to the deafferented region in rats: lack of correlation with autotomy behavior.

We have studied the development of cutaneous hypersensitivity in the innervation area of the saphenous nerve and autotomy behavior in rats after unilateral sciatic nerve section. Hypersensitivity was assessed by stimulating the saphenous area with mechanical (von Frey hairs and analgesimeter), cold (immersion) or heat (immersion or radiant heat) stimuli 10 days after sciatic nerve section. We did not observe any hypersensitivity to thermal stimulation or weak mechanical stimulation produced by von Frey hairs. A significant decrease in withdrawal threshold to intense mechanical pressure produce

Ascending myelinated axons of primary sensory neurons of the sciatic nerve in the posterior column of the rat spinal cord.

The present study was undertaken to obtain morphologic data about the posterior column of the spinal cord to characterize ascending myelinated axons of primary sensory neurons of the sciatic nerve. By applying doxorubicin to the right sciatic nerve in eight male Wistar rats, selective degeneration of centrally directed axons of these neurons in the posterior column was produced. Epon-embedded transverse sections of the posterior column at spinal cord segments C1, C3, C8, T6, L3 and L5 showed a circumscribed area (R) that contained a cluster of degenerated myelinated fibers. To characterize are

Axonal viability and the persistence of thermal hyperalgesia after partial freeze lesions of nerve.

Partial freeze injuries of rat sciatic nerve have been used to investigate the relationship between the amount of tissue injured and the magnitude and duration of the resulting thermal hyperalgesia. Complete freeze injury of peripheral nerve produces temporary anesthesia to peripheral stimuli and is a useful neurolytic technique. The present study examined the hypothesis that incomplete nerve lesions, in which some fibers survive while others undergo Wallerian (axonal) degeneration, lead to development of thermal hyperalgesia. In this study, performed on rats, one sciatic nerve was frozen with

Sciatic neuropathies--a retrospective review of electrodiagnostic features in 29 patients.

Sciatic neuropathy is an uncommonly diagnosed focal mononeuropathy. We reviewed the aetiology and electrodiagnostic features of 29 patients studied at the Neurodiagnostic Laboratory, Tan Tock Seng Hospital from January 1989 to April 1995. External nerve compression was the most common cause (38%) followed by trauma (21%). Other rare causes in this series included intragluteal injections, hip surgery and diabetic mononeuropathy while 24% had uncertain aetiology. Electrodiagnostic studies showed preferential involvement of the peroneal division in 51%. Axonal loss was found in 97%. We conclude t

Differential sympathetic neural control of oxygenation in resting and exercising human skeletal muscle.

Metabolic products of skeletal muscle contraction activate metaboreceptor muscle afferents that reflexively increase sympathetic nerve activity (SNA) targeted to both resting and exercising skeletal muscle. To determine effects of the increased sympathetic vasoconstrictor drive on muscle oxygenation, we measured changes in tissue oxygen stores and mitochondrial cytochrome a,a3 redox state in rhythmically contracting human forearm muscles with near infrared spectroscopy while simultaneously measuring muscle SNA with microelectrodes. The major new finding is that the ability of reflex-sympatheti

Ninjurin, a novel adhesion molecule, is induced by nerve injury and promotes axonal growth.

Peripheral nerve injury results in axonal degeneration and in phenotypic changes of the surrounding Schwann cells, whose presence is critical for nerve regeneration. Using differential screening strategies, we identified a novel protein, termed ninjurin (for nerve injury-induced protein), that is up-regulated after axotomy in neurons and in Schwann cells surrounding the distal nerve segment. Ninjurin is located on the cell surface, is capable of mediating homophilic adhesion, and promotes neurite extension of dorsal root ganglion neurons in vitro. Ninjurin is also expressed in a number of othe

Schwann cells produce tumor necrosis factor alpha: expression in injured and non-injured nerves.

We investigated the production of tumor necrosis factor alpha, a pleotropic cytokine, in the rat sciatic nerve using immunohistochemistry for the protein and in situ hybridization for specific messenger RNA sequences. Results demonstrated that in the uninjured sciatic nerve, few endothelial cells, fibroblasts, and Schwann cells were immunoreactive for tumor necrosis factor. In situ hybridization did not yield visible results in this control tissue. Following a compression injury, the number of immunoreactive cells in the nerve fascicle was increased. In situ hybridization showed positive stain
