Acutely denervated muscle EVs reshape neuronal mitochondrial metabolism via retrograde signaling to rescue peripheral nerve injury.

Peripheral nerve injury causes muscle atrophy due to slow axonal regeneration, highlighting an unmet therapeutic need. Although neuromuscular interactions are classically viewed as unidirectionally nerve dominated, we show that acutely denervated muscle (adMu) regulates nerve regeneration via a retrograde signaling pathway. adMu initiates a trans-tissue regulatory mechanism through extracellular vesicles (EVsadMu) that orchestrate neural energy homeostasis to accelerate regeneration. Functional profiling identifies IDH2 and CS as key metabolic enzymes within EVsadMu. Neurons treated with EVsad

Changes in Spinal Neural Circuit Plasticity in a Rat Sciatic Nerve Transection Model.

BACKGROUND AND AIM: Changes in neural plasticity crucially modulate functional recovery after central nerve injury. To describe morphological peripheral nerve injury-related changes, we comprehensively elucidated post-sciatic nerve transection (post-SNT) plasticity changes in the lumbar spinal corticospinal tract (CST), motor neurons, and cholinergic interneurons (CINs). METHODS: In a rat SNT model, we compared post-SNT changes in lumbar spinal cord plasticity at 2, 4, and 6 weeks with those in the Sham group. Using neural tracers and immunohistochemistry, we labeled and analyzed the CST axon

CCR2/CCR5 antagonism with cenicriviroc relieves neuropathic pain induced by sciatic nerve injury and delays morphine tolerance in mice.

The neuroimmune system is known to be pathologically activated after nerve injury and prolonged exposure to opioids. Whereas previous studies have predominantly focused on the involvement of individual chemokine receptors in this phenomenon, dual CCR2/CCR5 blockade with cenicriviroc represents a novel therapeutic strategy towards greater efficacy in alleviating hypersensitivity. In this study, the expression of opioid (MOR, DOR) and chemokine (CCR2, CCR5) receptors in the spinal cord was assessed by immunohistochemistry after chronic constriction injury (CCI) of the sciatic nerve. Analgesic ef

MaR1 and NGF combine to inhibit autophagy through the GSK-3β/β-catenin pathway to promote sciatic nerve repair.

BACKGROUND: Peripheral nerve injury (PNI) is a public health problem that can lead to sensory and motor deficits as well as neuropathic pain and secondary lesions. We explored the effects of the combination of MaR1 and NGF on sciatic nerve regeneration, reduction of neuropathic pain, and anti-inflammation, and further elucidated the associated molecular mechanisms. METHODS: After treatment of PC12 (adrenal pheochromocytoma cells) cells with NGF, MaR1 and H₂O₂, changes in proliferation were detected by CCK8; cell migration ability was detected by Transwell; reactive oxygen species (ROS) and ap

Integrative Proteomics Reveal Neuroimmune and Dopaminergic Alterations Across the Nociceptive Neuraxis in Neuropathic Pain.

Neuropathic pain (NP) arises from maladaptive changes in peripheral and central nociceptive circuits, yet molecular alterations spanning the entire pain neuraxis remain poorly understood. Neuroinflammation is increasingly recognized as a central mechanism in NP chronification, yet the region-specific molecular events linking immune activation to affective pain processing remain inadequately defined. In this study, we employed high-resolution LC-HRMS-based quantitative proteomics to investigate chronic constriction injury (CCI)-induced molecular alterations in the sciatic nerve (SN), spinal cor

Multidimensional Oriented Piezoelectric Conduits for Peripheral Nerve Defect Regeneration.

Peripheral nerve regeneration and functional recovery hold immense clinical significance, as they are critical for reinnervating the motor and sensory functions of organs. In this study, piezoelectric nerve guidance conduits (NGCs) with multidimensional oriented structures and zinc oxide (ZnO) nanoparticles were developed for this purpose, fabricated via a combination of digital light processing (DLP) 3D printing and directional freezing technologies. The therapeutic efficacy of the NGCs was evaluated in a rat model of sciatic nerve defect, with continuous monitoring of neuromorphological and

Nanoparticle-Mediated Immunometabolic-Epigenetic Remodeling Enhances Schwann Cell-Macrophage Interaction for Sciatic Nerve Regeneration.

Peripheral nerve regeneration continues to pose a significant clinical challenge, primarily attributable to the inherently limited regenerative capacity of axons and the intricate inflammatory microenvironment that develops following injury. While immunometabolic modulation has emerged as a promising therapeutic avenue, achieving precise and sustained intervention within the injury microenvironment remains technically challenging. Here, we introduce a biomimetic Prussian White nanoparticle (PW) that facilitates long-term local retention and drives coordinated immunometabolic-epigenetic remodel

Punicalagin Promotes Peripheral Nerve Repair by Enhancing Mitophagy via the TLR4/MAPK Signaling Pathway and Facilitating Angiogenesis.

Peripheral nerve injury (PNI) often leads to functional impairment, and current therapeutic options are limited. Punicalagin (PUN), a polyphenolic compound derived from Punica granatum , exhibits antioxidant and anti-inflammatory properties. This study investigated the therapeutic potential of PUN for PNI and its underlying mechanisms, focusing on mitochondrial autophagy (mitophagy), TLR4/MAPK signaling, and angiogenesis. In vitro, Schwann cells were treated with PUN under oxidative stress induced by tert-butyl hydroperoxide to evaluate cell viability, mitochondrial function, mitophagy, reacti

Bioinspired Multilayered Conductive Nerve Guidance Conduit Built on Hydrogels and Knitted Silk Fiber Sleeves for Peripheral Nerve Regeneration.

Repairing long-gap peripheral nerve injuries remains challenging. The main obstacles lie in the donor site morbidity and size restriction for autologous nerve transplantation as well as the lack of biomimetic structures for conventional artificial nerve conduits. Inspired by the multilayer structures of nerve conduit, we here present a composite nerve guidance conduit to mimic the hierarchical structures and bioelectrical properties of the native nerves. The conduit integrates an outer silk fibroin (SF) knitted sleeve (KS) to provide mechanical support and structural mimicry of epineurium, an

Study on the Analgesic Mechanism of Aloperine in Mice with Neuropathic Pain Based on the p38MAPK/FKN/CX3CR1 Pathway.

A lesion or disease of the somatosensory nervous system causes neuropathic pain (NP). Previous experiments have demonstrated that aloperine (ALO) exhibits antiallodynic effects on neuropathic pain in mice; however, the mechanism underlying its analgesic action remains unclear. This study investigated the analgesic mechanism of ALO on neuropathic pain in mice using a chronic constriction injury (CCI) model. Experiments were conducted using the ALO and p38MAPK inhibitor SB 203580. In vivo experiments demonstrated that ALO played an analgesic role by inhibiting p38MAPK phosphorylation, and it imp

f-SPION-mediated magnetic stimulation induces reparative Schwann cell reprogramming via cytoskeletal dynamics - gated activation of Piezo1.

The remarkable intrinsic regenerative capacity of peripheral nerves following injury is largely attributed to the phenotypic plasticity of Schwann cells (SCs) and their ability to transition into a repair-supportive state (rSCs). Transcriptional reprogramming of SCs into this reparative phenotype is pivotal for facilitating successful nerve regeneration. While traditionally considered a biochemically regulated process, recent advances in mechanobiology have underscored the crucial role of mechanical cues in modulating SC behavior and gene expression. In this study, we sought to develop a magne

Fatty acid binding protein 4 (FABP4): a key player in neuroinflammation and neuropathic pain.

Neuropathic pain (NP) is caused by lesions or diseases of the somatosensory system. Emerging evidence implicates adipokines in NP pathogenesis, yet the role of fatty acid-binding protein 4 (FABP4) remains unclear. Using a mouse model of sciatic nerve crush injury, we found that wild-type (WT) mice developed robust NP behaviors, concomitant with significant FABP4 upregulation and extensive macrophage infiltration in the injured nerve. In contrast, FABP4-knockout (FABP4-KO) mice exhibited markedly attenuated pain hypersensitivity. Single-cell RNA sequencing and subsequent histological analyses r

α-(Phenylselanyl) acetophenone reverses comorbid depressive-like behavior and mechanical allodynia induced by partial sciatic nerve ligation in mice.

This research investigated the potential therapeutic role of α-(phenylselanyl) acetophenone (PSAP) in the comorbidity of chronic pain and depression triggered by partial sciatic nerve ligation (PSNL). Male Swiss mice underwent PSNL surgery, and after a four-week period, they received either PSAP (1-50 mg/kg, administered intragastrically) or imipramine (IMI) (50 mg/kg) 30 min prior to behavioral assessments. Both PSAP and IMI effectively alleviated PSNL-induced hypersensitivity to pain and depressive-like symptoms, as demonstrated in forced swim and allodynia tests. Additionally, PSAP countera

Sinomenine ameliorates experimental autoimmune neuritis by suppressing pro-inflammatory immune components and restoring immune balance.

Sinomenine (SIN), a bioactive alkaloid derived from Sinomenium acutum, exhibits well-documented anti-inflammatory and analgesic properties with a favorable safety profile. Here, we pharmacologically characterized the preventive and therapeutic efficacy of SIN in a rat model of experimental autoimmune neuritis (EAN), an established model of immune-mediated peripheral neuropathy. Although SIN has been evaluated in several neuroinflammatory disorders, its mode of action in EAN has not been systematically addressed. Disease progression, body-weight changes, and neurological scores were monitored f

Missouri Osteochondral Preservation System for Nerve Preservation of Nerve Isografts Improves Functional Recovery After Peripheral Nerve Reconstruction With Polyethylene Glycol-Mediated Fusion.

PURPOSE: Functional outcomes after peripheral nerve reconstruction remain suboptimal, particularly for large-gap defects. This study evaluated the combined use of the Missouri Osteochondral Preservation System for nerve (MOPS-N) preservation for nerve isografts and polyethylene glycol-mediated fusion (PEGf) in a rat sciatic nerve gap model. METHODS: Adult male Lewis rats underwent an 8-mm sciatic nerve gap reconstruction. Animals were randomized to the following 6 groups: (1) MOPS-N isografts stored 28 days + PEGf, (2) MOPS-N isografts stored 56 days + PEGf, (3) Normosol-preserved isografts s

Refined model of common peroneal nerve injury and regeneration: A neurophysiological, morphometric, and functional analysis in the adult rat.

The common peroneal nerve lesion induces a specific peripheral nerve injury, characterized by a specific motor deficit with little animal burden. Although, it has been described in the 1980s, there is a lack of consistent outcome measurements in the literature. This study presents a refined common peroneal nerve injury model in Sprague Dawley rats with crush or transection injury. It introduces a morphometric muscle analysis, a refined videometric gait analysis and isometric force measurement, as well as testing the validity of outcome measures. The tibialis anterior muscle exhibited the large

Fidelity of Sensory Reinnervation in Peroneal and Sural Nerves to Lumbar Dorsal Root Ganglia 4 and 5 After Segmental Injury to the Sciatic Nerve.

Recovery after axotomy of a peripheral nerve is dependent on regrowth of axons from the point of injury to distal sensorimotor tissues and can be complicated by nerve branching. Little is known about regeneration of sensory axons that encounter branch points distal to injuries. The experiments reported here focused on this question and sought to assess the fidelity of sensory axon regeneration, where fidelity is defined as an axon that originally innervated a distal branch of the sciatic nerve regenerated into that same distal branch after injury, with serial retrograde labeling. Rats with seg

G6PD facilitates axon regeneration via clathrin-mediated endocytosis.

Metabolic reprogramming is a hallmark of neuronal repair, yet the roles of glucose metabolism-related enzymes remain poorly understood. To investigate their functions, we employed a sciatic nerve injury model, taking advantage the intrinsic regenerative capacity of peripheral neurons. After sciatic nerve crush injury, dorsal root ganglia exhibited sustained upregulation of several enzymes in the pentose phosphate pathway. Notably, glucose-6-phosphate dehydrogenase (G6PD), the rate-limiting enzyme of the pentose phosphate pathway, was markedly increased at both RNA and protein levels. Silencing

Ultrasound-guided nanocomposite hydrogel injection to enhance nerve repair via temporal delivery of a colony-stimulating factor 1 receptor inhibitor.

Persistent or dysregulated neuroinflammation impairs regeneration following peripheral nerve injuries (PNI). Current strategies for modulating inflammation in PNI lack spatial and temporal precision, thereby limiting their regenerative efficacy. In this study, we developed a nanocomposite hydrogel (DNH/PLGA@BLZ) by combining decellularized porcine sciatic nerve hydrogel (DNH) with poly(lactic-co-glycolic acid) (PLGA) nanoparticles loaded with BLZ945, a colony-stimulating factor 1 receptor (CSF-1R) antagonist currently in clinical trials. DNH/PLGA@BLZ demonstrated excellent biocompatibility and

LARP7 enhances the potential of dental pulp stem cells to promote peripheral nerve repair.

BACKGROUND: Peripheral nerve injuries (PNIs) present a persistent clinical challenge due to the intrinsically limited regenerative capacity of peripheral nerves. While dental pulp stem cells (DPSCs) exhibit significant neuroregenerative potential, their therapeutic efficacy is constrained by hostile microenvironments and inherent functional heterogeneity. Genetic modification may offer a promising strategy to enhance their therapeutic capabilities. METHODS: DPSCs were induced toward neural lineage differentiation, and key gene candidates were identified through qRT-PCR. Lentiviral-mediated ge
