BACKGROUND: Blastocystis sp. is a highly prevalent anaerobic intestinal protozoan increasingly associated with gastrointestinal disorders. Its survival within the gut microenvironment depends on metabolic pathways such as glycolysis, where glyceraldehyde-3-phosphate dehydrogenase (GAPDH) plays a central role, alongside interactions with nitric oxide (NO) derived from host defenses and surrounding microbiota. The emergence of metronidazole resistance necessitates exploration of alternative therapeutic strategies.
OBJECTIVE: To evaluate the in vitro effects of metronidazole, metformin, and Alli

BACKGROUND: The earliest interactions between Fasciola hepatica newly excysted juveniles (FhNEJ) and the intestinal epithelium remain poorly understood, partly due to the limited availability of physiologically relevant in vitro models. Here, we establish a human duodenum organoid-derived epithelial monolayer (ODM) co-culture system to investigate the early host-parasite interface, providing a controlled experimental platform for analysing interactions between FhNEJ and the intestinal epithelium.
METHODS: ODMs representing the human duodenum were co-cultured with either live FhNEJ or a tegume

Blastocystis is an anaerobic stramenopile that colonizes the large intestine of humans and many animals worldwide. Small-subunit rRNA-based typing reveals deep genetic diversity: at least 44 genetic variants, namely, subtypes (STs), are recognized, with ST1-ST4 accounting for most human carriage. Its medical relevance remains contested. Blastocystis carriage is frequently asymptomatic, and while associations with non-specific gastrointestinal symptoms (e.g. diarrhoea, abdominal pain and bloating), irritable bowel syndrome and extraintestinal manifestations such as urticaria have been proposed.

Giardia duodenalis is a flagellated commensal protozoan that can infect both humans and animals. Infection can result in clinical signs such as diarrhea or, more often, the infection can be subclinical. Within the G. duodenalis species, there are several major genetic assemblages, A - H. Certain assemblages are species specific (C and D in dogs, F in cats) while assemblages A and B can infect humans as well as other mammals. This retrospective study used data collected from a commercial reference laboratory (Antech Diagnostics, Mars Petcare Science & Diagnostics). Real-time PCR data using feca

Many parasites are transmitted through trophic interactions, yet the specific pathways connecting intermediate and definitive hosts often remain poorly understood in marine food webs. In the Southwestern Atlantic, the Argentine anchovy (Engraulis anchoita) has been considered a potential vector of larval Anisakis to higher trophic levels, but low infection levels in San Matías Gulf suggest alternative transmission routes in this gulf. We investigated the role of pearlsides (Maurolicus spp.), abundant mesopelagic fishes in the gulf, in the transmission of anisakid nematodes. Examination of 229

Giardiasis, caused by Giardia intestinalis, is a prevalent intestinal protozoan infection worldwide. Clinical manifestations are variable, and many infections remain asymptomatic. Disease pathogenesis is multifactorial, involving brush border enzyme deficiency, apoptosis, dysbiosis and increased intestinal permeability. Advances in genomics, genetic tools, and model systems have substantially improved our understanding of the parasite's virulence mechanisms. Giardia exhibits considerable genetic diversity, with distinct assemblages associated with differences in parasite biology. Attachment is

Through systematic structural optimization and modification of anti-Cryptosporidium parvum compound NSC158011 (N-1-naphthalenyl-2-(phenylthiol) ethane thioamide), we constructed a virtual compound library comprising 1,440 derivatives, and 29 derivatives were selected via molecular docking for chemical synthesis. Among them, eight derivatives exhibited significant inhibitory effects on LDH compared with NSC158011. In vitro examination found that two derivatives could markedly reduce the number of C. parvum in cells while exhibiting reduced toxicity. In vivo studies in model animals (IFN-γ knock

The genus Chelonoidis tortoises are terrestrial reptiles susceptible to gastrointestinal endoparasites, mainly due to improper management. The present study aims to analyze the frequency of gastrointestinal parasites detected in samples from Chelonoidis carbonarius and Chelonoidis denticulatus, and to identify and characterize taxonomically the main parasite types. Seven samples of fresh feces were collected from enclosures at the Jungle Warfare Training Center (CIGS) Zoo in Manaus, Amazonas, and evaluated. A high frequency of intestinal endoparasites was observed, with nematode eggs from the

Given the grave risk posed by intestinal cryptosporidiosis to vulnerable populations, the current research focused on a new therapeutic avenue. Olibanum (OL) extract and probiotic Bacillus clausii (B. clausii) and their combination were evaluated against Cryptosporidium in vivo in immunosuppressed mice via parasitological, ultrastructural, biochemical, and histopathological analysis. Liquid chromatography-mass spectrometry (LC-MS) analysis of OL revealed the predominance of incensole isomers in its chemical composition. Regarding oocyst count in stool, the highest percentage of oocyst reductio

Mastomys natalensis is a widespread rodent species in sub-Saharan Africa and is increasingly recognised as an important component of food systems in some communities. However, rodents may harbour gastrointestinal helminths with potential implications for animal health, food safety, and zoonotic disease transmission. Information on gastrointestinal helminth infections in M. natalensis and the response of these parasites to anthelmintic treatment is limited. This study investigated the prevalence and diversity of gastrointestinal helminths in M. natalensis and evaluated changes in helminth egg b

BACKGROUND: Infestations with fleas, ticks and intestinal nematodes are commonly identified in domestic dogs and have significant veterinary and public health importances. The study described in this manuscript was conducted to verify the natural parasitism of domestic dogs in two Brazilian regions, and to confirm the efficacy of the isoxazoline afoxolaner, formulated alone (NexGard®) or in combination with the macrocyclic lactone milbemycin oxime (NexGard Spectra®), for the control of fleas and ticks, and to confirm the efficacy of the afoxolaner/milbemycin oxime (MO) combination for the cont

Intestinal parasitic infections (IPIs) remain a critical global public health concern, particularly for epidemiological surveillance in resource-limited tropical regions. Standard laboratory diagnostics heavily rely on the manual microscopic examination of stool samples, a process that is notoriously time-consuming, labor-intensive, and highly dependent on the scarce availability of experienced parasitologists. To alleviate medical workloads and enhance community-wide screening efficiency, reliable, automated diagnostic tools are urgently needed. From an epidemiological control perspective, su

Giardia, a flagellated protozoan, is widely distributed in wild rodents. To characterize the presence of Giardia in wild rodents in southern China, this study collected 344 fecal samples from wild rodents in Guangxi, Hunan, and Yunnan provinces. The β-giardin gene (bg) was amplified to identify the presence of Giardia sp. The results showed that 48 samples (13.95%, 48/344) tested positive for Giardia, including three positives for G. duodenalis (assemblage G n = 1, assemblage F n = 2) and 45 positives for G. microti. Among these, Microtus fortis had the highest prevalence at 30.22% (42/139), f

This systematic review and meta-analysis aimed to evaluate the global epidemiology, subtype (ST) distribution, and zoonotic potential of Blastocystis sp. in birds. A systematic review of the literature up to July 26, 2024, identified 47 articles/98 datasets meeting the inclusion criteria, covering data from 19 countries and over 41 bird species. Total estimates and 95% confidence intervals (CIs) were calculated using a random-effects model. The weighted prevalence of Blastocystis sp. in birds was 26.4% (95% CI: 22.5-30.6%), varying by location and species. The highest pooled prevalence was in

Taenia saginata, commonly known as the beef tapeworm, is a prevalent zoonotic cestode with an estimated global prevalence of 60-70 million carriers. This parasite is frequently located in the gastrointestinal tract, and previous reports have described complications such as acute appendicitis, Meckel's diverticulitis, pancreatitis, cholecystitis, liver abscess, intestinal obstruction, and perforation. Intestinal perforation due to parasitic diseases is a rare complication. Surgical complications associated with T. saginata are uncommon, with few reported cases of bowel perforation. Here, we pre

A 36-year-old man presented with sudden onset of severe abdominal pain. A CT scan was performed first, which showed marked thickening of the gastric wall. Gastric anisakiasis was suspected because he had eaten sashimi (a raw fish dish), including salmon and tuna, the night before. An upper gastrointestinal endoscopy revealed three Anisakis larvae invading the gastric wall. All larvae were removed by the biopsy forceps. His symptoms were resolved after the removal. Anisakiasis is a gastrointestinal parasitic infection of humans caused by ingestion of raw or undercooked seafood containing Anisak
