Primary Amoebic Meningoencephalitis caused by Complement C2 Deficiency.

BACKGROUND: Primary amoebic meningoencephalitis (PAM) is a rapidly progressive and often fatal central nervous system infection caused by Naegleria fowleri. Despite widespread environmental exposure to this free-living amoeba, clinical disease is rare, suggesting that it requires not only exposure to the amoeba but also a host vulnerability. Yet, the immune mechanisms controlling protection vs. susceptibility to N. fowleri remain poorly understood. METHODS: We conducted comprehensive clinical, immunological, and genetic investigations in one of the few survivors of PAM. We performed high-dime

Endoparasites in carnivores in Swiss zoological institutions between 2009 and 2024: evaluation of risk factors and deworming strategies.

Parasitic infections in zoo carnivores pose risks to animal health and zoonotic transmission. Over a 15-year period, endoparasites were detected in faecal samples from Swiss zoo carnivores, with clinical and management data used to identify factors linked to parasite occurrence. The samples, originating from 50 species kept in 30 different institutions, were collected in 2009 (n = 140), 2018 (n = 149) and 2024 (n = 116), and analysed by Baermann-Wetzel technique, combined sedimentation/flotation, Giardia coproantigen detection and Ziehl-Neelsen staining of faecal smears. Fifteen samples were f

Epidemiology of gastrointestinal parasitism and field applicability of FAMACHA© in indigenous goats of Southern India.

Gastrointestinal parasitism severely impacts small ruminant production in tropical regions. This study investigated prevalence, risk factors, infection intensity, and co-infection patterns in 1013 goats across five districts in Southern India, while evaluating the field applicability of FAMACHA© scoring as a practical clinical indicator. Overall parasite prevalence was 80.4%, dominated by strongyle nematodes (84.9% of infected animals) and Eimeria spp. (57.3%), with 71.5% mixed infections. Multivariable logistic regression identified breed, sex, season, age, and deworming history as independen

Prevalence and Genetic Diversity of Blastocystis sp. in Humans, Dogs and Cats in Gabon: A One Health Perspective.

INTRODUCTION: Blastocystis sp. is a cosmopolitan protist that colonises the gastrointestinal tract of mammals, including humans, dogs and cats. Although its pathogenicity remains debated, Blastocystis sp. has been linked to gastrointestinal symptoms and alterations in the gut microbiota. In Gabon, most epidemiological data on Blastocystis sp. rely on microscopy, which may underestimate true prevalence. Moreover, molecular studies remain scarce, and none have investigated the parasite's presence in domestic animals. This study aimed to estimate the prevalence and subtype (ST) distribution of Bl

Parasitic surveillance in wolves of central Italy: a focus on the Abruzzo region.

BACKGROUND: Monitoring parasitic infections in wildlife is essential for assessing ecosystem health and pathogen dynamics, particularly in apex predators like the wolf (Canis lupus). As top-level carnivores with wide-ranging habitats and diverse interactions with prey, wolves can serve as effective sentinels for the circulation of parasitic agents within ecosystems. The present study aimed at monitoring parasitic infections, particularly those of zoonotic importance, in wolves from central Italy. METHODS: The study was conducted on 169 wolf carcasses recovered between 2018 and 2024. Leishmani

Determinants of Ascaridoid Nematode Infection and Anisakis-Related Zoonotic Exposure Risk in Eastern Mediterranean Gadiformes Fishes.

Larvae of ascaridoid nematodes, particularly Anisakis spp., are common parasites of commercially important marine fishes and may represent a zoonotic hazard following ingestion of raw or undercooked seafood. We investigated the ascaridoid fauna of the sympatric European hake (Merluccius merluccius) and greater forkbeard (Phycis blennoides) from the Ionian Sea (Eastern Mediterranean), integrating host biometric and seasonal drivers with molecular identification and quantitative risk assessment (QRA) for the zoonotic Anisakis pegreffii. Anisakis pegreffii was the dominant species in both hosts,

Excretory-secretory products of the fish-borne parasite Anisakis simplex L3 larvae possess allergens and unusual glycan modifications.

Anisakis simplex is a parasitic aquatic nematode, which may cause mild-to-severe gastrointestinal allergic reactions (anisakiasis) with clinical symptoms, such as rhinitis and urticaria, in humans who accidentally consume raw or undercooked marine products contaminated with infective L3 Anisakis larvae. Several Anisakis excretory-secretory (E/S) products and somatic proteins are known to be involved in IgE-mediated allergic reactions. In comparison with vertebrates, nematodes have a distinct machinery to glycosylate their proteins, and unusual glycan structures have been reported previously, m

Current Applications and Immunological Considerations of Salmonella enterica Serovar Typhimurium as a Vaccine Vector.

Live attenuated Salmonella enterica serovar Typhimurium has been investigated for decades as an orally delivered vaccine vector due to its ability to target the intestinal mucosa and engage both innate and adaptive immune responses. In humans, S. Typhimurium infection is largely restricted to the gastrointestinal tract, distinguishing it from Salmonella Typhi and providing a rationale for its use in mucosal vaccine strategies. In this review, we discuss the biological features of S. Typhimurium that support its use as a vaccine vector and summarize current understanding of the immune responses

Schistosoma Species and Hybrid Genotyping With a Field Deployable Multi-Marker Amplicon Panel.

Schistosomes are parasitic trematode worms of the genus Schistosoma, responsible for causing urogenital and intestinal schistosomiasis. Six primary species infect humans: S. mansoni, S. japonicum, S. mekongi, S. intercalatum, S. guineensis, and S. haematobium. In addition, several species including S. bovis, S. curassoni, and S. mattheei primarily infect animals, particularly cattle. Hybridization events have been documented both between human-infecting species and between human- and animal-infecting species of Schistosoma. Current methods for detecting hybrids rely on genotyping mitochondrial

Virulence attenuation of intestinal pathogenicity via combined gene deletion in duck enteritis vaccine strain restores gut microbiota balance and enhances safety.

There is an increasing need for a new generation of effective and safe vaccines, in the context of large-scale poultry farming and the prevalence of infectious diseases. With this in mind, we developed, for the first time, a duck enteritis virus (DEV) mutant, ΔTK-ΔgI/gE-ΔgG/gJ, through the deletion of multiple virulence genes. The resulting gene-deletion strain exhibited replication kinetics similar to those of the parent strain and was found to be safe in various animal models, offering a strategy for rapidly generating attenuated DEV strains. Previously, our team reported that DEV infection

Prevalence, risk factors, and zoonotic potential of gastrointestinal helminths in cats: A participatory epidemiological study.

Domestic cats are the only felines living exclusively with humans, making them ecologically invasive and widespread across terrestrial habitats. Their domestication stems from their rodent control abilities and companionship. However, they are susceptible to gastrointestinal parasites such as Ancylostoma spp., Toxocara cati, and Dipylidium caninum, causing issues like anaemia, vomiting, and sometimes death, especially in kittens. These parasites can also pose zoonotic risks, transmitting diseases like visceral and ocular larva migrants to humans. A total of 100 cats with no deworming history w

Analysis of MIR155HG gene polymorphisms and ulcerative colitis susceptibility in the Chinese Han Population.

Aberrant expression of miR-155 has been implicated in the pathogenesis of inflammatory bowel disease (IBD), where its dysregulation may contribute to impaired intestinal barrier integrity and sustained inflammation. However, the precise role of miR-155 in the development of ulcerative colitis (UC) remains incompletely understood. This case-control study aimed to evaluate the association between polymorphisms of MIR155HG gene and susceptibility to UC in a Chinese Han population. The study included 84 UC patients and 216 matched healthy controls. Four single nucleotide polymorphisms (SNPs) of MI

The dual role of intestinal parasites in shaping human gut microbiota: distinguishing helminthic and protozoan dynamics.

The human gut microbiota is essential for supporting the host's health and immune system. Imbalances in this microbiota, known as dysbiosis, are associated with numerous inflammatory and autoimmune diseases. Despite extensive research on bacterial components, the significant modulatory role of eukaryotic gut inhabitants, particularly intestinal parasites, remains largely overlooked. While traditionally viewed as pathogens, emerging evidence highlights a profound duality in their role, suggesting they are integral and active modulators of the gut microbiota and challenging the conventional view
