Disseminated ascariasis infection with encephalopathy: Molecular confirmation and phylogenetic insights.

Ascariasis remains a major parasitic infection in low-resource regions, with children at highest risk. We describe a rare and severe pediatric case from rural Colombia involving intestinal obstruction, peritonitis, cholecystitis, and encephalopathy. A recovered helminth underwent stereoscopic examination and molecular characterization through amplification of 18S-rRNA and COII genes, followed by long-read sequencing using Oxford Nanopore technology. Taxonomic analysis with Centrifuge and BLAST confirmed the specimen as Ascaris, showing high similarity to Ascaris suum, while COII phylogenetics

The photodynamic inactivation in the control of the seafood zoonotic parasite, Anisakis sp.

Anisakis spp. L3 larvae commonly found in marine fish species are an important biological hazard when raw or inadequately thermally processed fishery products are consumed with live larvae, highlighting the need for effective control strategies. This study evaluated the efficacy and mechanism of photodynamic inactivation (PDI) of Anisakis L3 larvae, using the cationic photosensitizer (PS) 5,10,15,20-tetrakis(1-methylpyridinium-4-yl)porphyrin tetraiodide (TMPyP). Larvae were collected from Atlantic horse mackerel and grouped by size: batch A (larvae ≈ 1.5 cm) and batch B (larvae ≈ 2.0 cm). Inac

Controlled human helminth infection models: insights into type 2 immunity and therapeutic development.

Controlled human infection models are powerful tools for evaluating pathogenesis, immune responses, vaccines, and therapeutics, including parasitic helminths. Helminths have also been tested as inflammatory disease therapeutics. This review focuses on immune responses during controlled infections with Trichuris species, Necator americanus, and Schistosoma mansoni. Over 700 participants have been experimentally infected across 33 studies to date. These models reveal significant heterogeneity in type 2-associated responses among individuals for all infections. While eosinophilia is most frequent

Gut microbiota and sarcoidosis: a concise review.

Microbial involvement in sarcoidosis pathogenesis is suggested by the observation that histological findings in sarcoid granulomas are like those of leprosy, tuberculosis and parasitic infection. Some studies have shown that the lung microbiome in patients with sarcoidosis is different from healthy individuals. Results are conflicting, reporting an abundance or decrease in bacterial and fungal species. The altered composition of the microbiome in sarcoidosis can contribute to the formation of granulomas, typical lesions of the disease, through interactions with the host immune system. However,

Giardia intestinalis trophozoites activate human PMN and induce NET formation but dampen neutrophil ROS production.

INTRODUCTION: Giardia intestinalis is a zoonotic enteric protozoan parasite causing giardiasis in humans, domestic animals and wildlife. More than 300 million human cases of diarrhea due to giardiasis have annually been reported. Despite its high global prevalence, human polymorphonuclear neutrophil (PMN)-mediated early innate immune responses against G. intestinalis remain poorly investigated. This study aimed to evaluate whether vital G. intestinalis trophozoites activate PMN and foster neutrophil metabolic responses, thereby eventually driving NET formation. METHODS: Human PMN were exposed

Functionally-derivatized Nanoantibodies as novel tools for Giardia lamblia cyst detection.

Giardia lamblia (syn. intestinalis, duodenalis) is the causative agent of Giardiasis, a diarrheal disease of global medical importance, especially problematic in young children living in unhygienic, resource-constrained settings. Diagnostics of potential Giardia infections are generally done through classic light-microscopy stool examination. This is often insufficient, and ELISA-based fluorescence detection using costly proprietary reagents is employed. These reagents are often not affordable in contexts where they are needed the most, and this limits their use to resource-rich settings where

Inflammatory ILC2s migrate to distal tissues during infection using stage-specific S1P receptors.

Tissue-resident lymphocytes can recirculate, but the underlying molecular mechanism is poorly understood. During helminth infection, intestinal group 2 innate lymphoid cells (ILC2s) rapidly proliferate and give rise to inflammatory ILC2s (iILC2s), which migrate from the intestine to distal tissues. Here, we show in mice that the redistribution of iILC2s requires access to lymphatic vessels. Interleukin-25 (IL-25) induces a substantial change in the epigenetic landscape of iILC2s, with transcription factors KLF2 and ZEB2 driving increased expression of sphingosine-1-phosphate receptor 1 (S1PR1)

Treatment-refractory giardiasis is predominantly an imported infection from South Asia associated with Giardia sub-assemblage BIII: a five-year study from the Czech Republic.

Giardiasis is a cosmopolitan parasitic infection caused by a protozoan pathogen Giardia intestinalis. A high number of failures complicates the metronidazole (MTZ) treatment. However, long-term studies characterising the parasites associated with MTZ treatment failures are lacking. This study was designed to map Giardia genotypes, clinical manifestations and geographical origin of giardiasis in a patient cohort from the Czech Republic. A multilocus genotyping was applied to 93 Giardia DNA isolates collected over five years in a tertiary care hospital diagnostic facility, and corresponding pati

Hookworm genes encoding intestinal excreted-secreted proteins are transcriptionally upregulated in response to the host's immune system.

Hookworms are intestinal parasitic nematodes that chronically infect ~500 million people. How hookworms successfully overcome host protective mechanisms is unclear, but it may involve hookworm proteins that digest host tissues, or counteract the host's immune system, or both. To find such proteins in the zoonotic hookworm Ancylostoma ceylanicum, we used mass spectrometry to identify 565 genes encoding excreted-secreted (ES) proteins from adults, and used RNA-seq to identify genes expressed both in young adults (12 days post-infection) and in intestinal and non-intestinal tissues dissected from

Comparative evaluation of four genetic markers for molecular detection of Giardia duodenalis in human fecal samples.

BACKGROUND: Giardia duodenalis is globally distributed intestinal protozoan parasite responsible for giardiasis causing various clinical manifestations ranging from asymptomatic to chronic diarrhea and malabsorption. Accurate diagnosis is crucial for effective clinical management and prevention of transmission especially in cases with low-parasite burden where false-negative results remain a challenge. METHODS: In this study, the diagnostic performance of four commonly used molecular markers: triosephosphate isomerase (tpi), small subunit ribosomal RNA (ssu-rRNA), glutamate dehydrogenase (gdh
