Establishment of In Vivo Mouse Models to Study Salmonella-Induced Chronic Infection and Gastrointestinal Cancer.

Salmonella infection increases the risk of chronic diseases, e.g., inflammatory bowel diseases, colon cancer, and gallbladder cancer. However, the mechanisms leading to the development of these conditions are still not completely understood. Here, we describe the methods to establish in vivo mouse models to study Salmonella-induced chronic infection and colon cancer. These models will allow us to investigate the initiation and progression of diseases.

Global, regional, and national burden of exposure to chewing tobacco, 1990-2023: a systematic analysis of the Global Burden of Disease Study 2023.

BACKGROUND: Exposure to chewing tobacco is a known health risk factor but has received less attention than smoking in global tobacco control efforts. As the evidence base linking chewing tobacco use to poor health grows, it is essential to closely track the changing use of chewing tobacco distinct from other forms of smokeless tobacco, as well as the estimated burden associated with its use. These data are essential to inform policy makers who seek to identify and avert the health burden associated with these dangerous products. METHODS: We estimated the prevalence of chewing tobacco use and

Decompressive Cholecystocentesis in 14 Dogs with Gallbladder Distension and Persistent Primary Digestive Symptoms.

Decompressive cholecystocentesis (DCC) was performed in 14 dogs with gallbladder distension (GBD) and primary digestive symptoms non-responsive to conventional symptomatic therapy. All dogs in this uncontrolled, retrospective case series had their fasting estimated gallbladder volume (eGBV) measured by the ellipsoid method. Aspirated volumes were compared to predicted volumes. Common symptomatic characteristics included diurnal inappetence (DI) and bilious vomiting (BV). Total magnesium insufficiency was common in the general population. In all dogs treated, symptom relief was reported within

FGF21-NR1D1 signaling restrains the CYP4A/20-HETE/GPR75 axis to counteract integrated renal-vascular injury in hypertension.

Hypertension leads to concomitant injuries in the kidney and vasculature, yet the molecular mechanisms underlying this coordinated pathology remain incompletely understood. Here, we identify activation of the CYP4A/20-HETE/GPR75 axis as being associated with coordinated renal and vascular injury in hypertension. In hypertension, this pathway was concurrently activated in the liver, kidney, and vasculature and was associated with renal fibrosis, endothelial dysfunction, and oxidative stress. Pharmacological inhibition of CYP4A attenuated these pathological changes, supporting a functional contr

Comparative hepatoprotective effects of Ginkgo biloba extract and N-Acetylcysteine against methotrexate-induced liver injury in rats: modulation of IκB-α, p38 MAPK, and p53 phosphorylation.

Methotrexate (MTX) is an effective chemotherapeutic agent whose use can be limited by hepatotoxicity. This study comparatively evaluated the effects of Ginkgo biloba extract (GbE) and N-acetylcysteine (NAC) on MTX-induced liver injury, focusing on oxidative stress, inflammatory and apoptotic responses, and associated hepatic phosphorylation of Inhibitor of nuclear factor kappa-B alpha (IκB-α), p38 mitogen-activated protein kinase (p38 MAPK), and tumor protein p53 (p53), key components of stress-responsive signaling pathways. Thirty-two adult male rats were randomly assigned to four groups (n =

Neuroimaging findings in hyperammonemic encephalopathy associated with sepsis-induced liver injury: a pilot case series.

BACKGROUND: Hyperammonemic encephalopathy (HE) is a severe complication of sepsis-induced liver injury (SILI), often associated with sepsis. However, its neuroimaging manifestations remain poorly understood. The aim of this pilot case series is to describe the brain MRI findings in patients with sepsis and HE. METHODS: We analysed brain MRI data from 15 consecutive patients with sepsis and clinically confirmed HE. Assessment of cerebral changes was performed using a standard MRI protocol. The following parameters were evaluated during the analysis: localization and signal intensity characteri

ATF6α mediates Cr(VI)-induced acute hepatotoxicity via GRP78-dependent mitochondrial dysfunction under oxidative stress.

Acute exposure to the common environmental toxin, hexavalent chromium (Cr(VI)), can cause severe chemical hepatotoxicity. Although endoplasmic reticulum (ER) stress is closely associated with Cr(VI)-induced liver damage, the underlying mechanism requires further investigation. In this study, mice and hepatocytes exposed to Cr(VI) were used to analyze the effects of reactive oxygen species (ROS)-mediated ER stress on acute liver injury, with a particular focus on the activating transcription factor 6 (ATF6) branch. It was found that Cr(VI) stimulated the expression and cleavage of ATF6α, while

A near-infrared fluorescent probe for imaging hydrogen peroxide in living cells and acute liver injury.

Acute liver injury (ALI) is linked to oxidative stress, with hydrogen peroxide H2O2 as a key mediator. We report a near-infrared fluorescent probe, PhCy-H2O2, based on an ICT mechanism. The probe shows high sensitivity (detection limit in the low micromolar range) and excellent selectivity, producing a significant fluorescence turn-on at 796 nm upon H2O2 recognition. This reaction has the advantages of high sensitivity, high selectivity, fast reaction and stability under physiological pH conditions. In both cellular and mice models, the probe clearly visualized dose-dependent H2O2 generation i

Nonlinear Dose-Response Association Between Exercise Intensity and Urinary Liver-Type Fatty Acid-Binding Protein in Young Men: A Piecewise Threshold Analysis.

High-intensity physical exercise can induce temporary renal hemodynamic strain and subclinical tubular injury, yet the exact dose-response relationship remains poorly defined. This study investigated the relationship between exercise intensity and urinary liver-type fatty acid-binding protein (L-FABP) concentrations, a sensitive marker of proximal tubular stress, to identify an exploratory intensity threshold. We conducted an observational cohort study of 285 healthy young men undergoing a routine 20 min physical training session. Exercise intensity was quantified as metabolic equivalents of t

Reduced hepatic GLUL expression accompanies altered RIP3/MLKL-pathway protein abundance in early cholestatic liver injury following bile duct ligation.

BACKGROUND: Obstructive cholestasis is characterized by disturbances in bile acid homeostasis, amino acid metabolism, inflammatory responses, and hepatocellular injury. Although RIP3/MLKL-pathway proteins have been implicated in cholestatic liver damage, their relationship with metabolic alterations remains incompletely understood. This study aimed to identify necroptosis-related genes and associated metabolic-pathway alterations in early bile duct ligation (BDL)-induced cholestatic liver injury through integrated bioinformatics analyses and experimental validation. METHODS: Gene expression d

Integrating gut microbiome and serum metabolomes to investigate the hepatoprotective effects of Lactiplantibacillus Plantarum PA01 against deoxynivalenol-induced liver injury in broilers.

Deoxynivalenol (DON) contamination is a significant challenge in broilers, which has a negative impact on liver function and health, endangering broiler production. This study aimed to evaluate the protective effects of L. plantarum PA01 in mitigating DON-induced liver injury and to elucidate the underlying mechanisms. A total of 144 one-day-old Arbor Acres broilers were divided into three groups: the control group receiving only a basal diet, the DON group (5 mg DON/kg diet), and the DP group (1 × 109 cfu L. plantarum PA01/g diet + 5 mg DON/kg diet). The results showed that L. plantarum PA01

Cardio-hepatic injury phenotypes and short-term outcomes in acute decompensated heart failure.

BACKGROUND: Abnormal liver biochemistry is common in acute decompensated heart failure (ADHF), yet most studies evaluate individual markers rather than clinically meaningful patterns of hepatic involvement. We examined whether cardiohepatic injury phenotypes derived from routine liver tests identify distinct short-term prognostic profiles in patients hospitalized with ADHF. METHODS: In this retrospective cohort study, we included consecutive adults hospitalized with ADHF between December 2006 and 2017. Using peak in-hospital values of aspartate aminotransferase, alanine aminotransferase, alka

Hybrid lesions of hepatocellular adenoma and focal nodular hyperplasia: a clinicopathological and molecular study of two cases.

AIMS: Hybrid lesions containing distinct components of hepatocellular adenoma (HCA) and focal nodular hyperplasia (FNH) within a single nodule are exceptionally rare. This study aimed to investigate the biological nature of these unusual lesions. METHODS AND RESULTS: The two cases identified to date were examined by histopathological, immunohistochemical and targeted next-generation sequencing analyses. In case 1, the hybrid tumour (6.0 cm) consisted of approximately equal proportions of HNF1α-inactivated HCA and FNH, which were intimately admixed but remained histologically distinct. In case

The maternal-placental-fetal liver axis: A mechanistic framework for understanding benzene developmental hematotoxicity.

Benzene is a ubiquitous environmental pollutant and a human hematotoxicant and carcinogen. Increasing epidemiological and experimental evidence links in utero benzene exposure to adverse pregnancy outcomes, altered fetal development, and increased childhood leukemia risk. Benzene toxicity is initiated through metabolism into reactive intermediates, primarily by cytochrome P450 2E1 (CYP2E1) and myeloperoxidase. These enzymes are expressed not only in the maternal liver but also in the placenta and fetal liver, indicating that multiple compartments may contribute to benzene toxicity during pregn

Blood-transforming coating enables hemostasis and vascularized bone re-generation via bioactive gelation.

Conventional bone repair materials often treat blood as an obstacle through physical isolation or passive hemostasis, overlooking its physiological role in the regenerative cascade. Here, we propose a "Blood-Transforming Coating" concept and engineer a functionalized microsphere system (GM/TCP@PEG). The NHS ester-coated microspheres rapidly crosslink with amino-rich blood components to form a robust hydrogel while covalently anchoring to tissue surfaces, transforming blood into a synergistic component that enhances material function. Structural characterization confirmed the core-shell archite

Silybum marianum peptide-rich fraction alleviates acute liver injury by inhibiting TRAF3/NIK/NF-κB and is associated with changes in gut microbiota composition.

Acute liver injury (ALI) is a severe and prevalent condition that can progress to irreversible liver damage and failure. This study aimed to investigate the prophylactic hepatoprotective potential of Silybum marianum peptides (SMP) in ALI and elucidate its underlying mechanisms. Using a carbon tetrachloride (CCl4)-induced ALI mouse model, we evaluated the dose-dependent effects of SMP. Additionally, we explored the modulatory effects of SMP on the NF-κB pathway, gut microbiota composition, and metabolite profiles. The role of SMP in the TRAF3/NIK/NF-κB signaling axis was further validated in a

Glycochenodeoxycholate promotes IL-8 expression through ER-mitochondrial calcium signaling in human hepatocytes.

Accumulation of excessive bile acids in the liver contributes to the progression of liver injury; thus, elucidating the bile acid-induced responses remains a critical challenge. Although conjugated bile acids have recently been reported to induce inflammatory responses at non-cytotoxic concentrations, the precise molecular mechanisms involved are not fully understood. The present study investigated the role of intracellular calcium dynamics-focusing particularly on mitochondria-in bile acid-induced inflammatory signaling. Exposure of human hepatocytes isolated from humanized liver chimeric mic
