Cynaroside Attenuates Ethinylestradiol-Induced Cholestatic Liver Injury by Modulating the FXR/BSEP/CYP7A1/SHP Axis.

Cholestatic liver diseases are characterized by disrupted bile acid homeostasis, oxidative stress, and inflammation, ultimately leading to progressive hepatic injury. Although ursodeoxycholic acid (UDCA) remains the standard pharmacological therapy, its therapeutic efficacy is limited in several cholestatic conditions. This study investigated the hepatoprotective effects of cynaroside (CYN), a naturally occurring flavonoid glycoside, against ethinylestradiol (EE)-induced cholestatic liver injury and explored its underlying molecular mechanisms. Female Wistar rats were randomly allocated into f

Nephroprotective Effect of the Methanolic Extract of Flourensia cernua Against Renal Ischemia-Reperfusion Injury Associated with Modulation of Oxidative Stress in Wistar Rats.

Renal ischemia-reperfusion (I/R) injury is a major cause of graft dysfunction. Flourensia cernua possesses antioxidant properties that may protect against this injury. This study evaluated its antioxidant capacity and nephroprotective effects in a renal I/R model in Wistar rats. The extract was characterized by phytochemical analysis, total phenolic content, and ABTS, DPPH, and FRAP assays. Hemolytic activity was assessed, and rats were assigned to Sham, Non-Tox, I/R, and I/R + F. cernua groups (200 mg/kg/day for 7 days before I/R). Biochemical parameters, renal oxidative stress markers, and G

Tartrazine-Induced Toxicity Is Attenuated by Naringenin in Wistar Rats: A Biochemical, Histological, and Behavioral Study.

Background: Tartrazine (TZ) has been linked to hyperactivity in children, as well as to kidney and liver damage. Furthermore, naringenin (NG) is a flavonoid with antioxidant and anti-inflammatory properties that may mitigate the damage caused by TZ. Methods: Four independent groups were used, each consisting of seven male Wistar rats-vehicle VEH (saline solution/DMSO, 0.600 mL; IP), TZ (150 mg/kg; IP), NG (100 mg/kg; IP), and TZ + NG (150 mg/kg; 100 mg/kg; IP). For 28 days, treatments were administered every 24 h, and the behavioral effects were evaluated via the open field and Rotarod tests.

Sulforaphane Alleviates α-Amanitin-Induced Liver Injury by Modulating Pink1-Parkin Pathway.

BACKGROUND: α-Amanitin (α-AMA) is the primary lethal toxin found in poisonous mushrooms of the genus Amanita; accidental ingestion can lead to severe liver injury, and there is currently no specific antidote. Due to its significant anti-inflammatory and antioxidant activity, Sulforaphane (SFN) has attracted increasing attention for its role in preventing liver injury. However, it remains unclear whether SFN can alleviate α-AMA-induced liver injury. This study aimed to elucidate the protective effects of SFN against α-AMA-induced liver injury and its potential molecular mechanisms. METHODS AND

Physiological and Pathological Responses of Grass Carp (Ctenopharyngodon idella) to Total Dissolved Gas Supersaturation During Exposure and Subsequent Recovery.

The total dissolved gas (TDG) supersaturation induced by discharged floods severely threatens fish survival during the flood season. However, investigations into the physiological and pathological effects of TDG supersaturation on fish remain limited, and even less is known about the recovery from TDG-induced injuries at the physiological and pathological levels. The biochemical and histopathological responses of fish injured by TDG exposure, and whether injured fish can return to normal conditions, remain unclear. In this study, on the basis of a combination of in situ investigations and labo

Hepatoprotective Effects of Flavonoids Against Acetaminophen Toxicity.

Acetaminophen (APAP) is a common analgesic and antipyretic agent; however, overdose can cause severe liver injury requiring transplantation or resulting in death. Currently, N-acetylcysteine (NAC) is the standard first-line treatment for APAP-induced liver injury (AILI). There is ongoing interest in exploring complementary hepatoprotective compounds, such as plant-derived flavonoids and their mechanisms of action to improve our understanding and management of AILI. This review aims to identify key flavonoids studied to date in the setting of AILI and their proposed mechanisms. A systematic sea

Pharmacological TLR4 Activation Promotes Hepatocyte Proliferation and Liver Repair Through Coordinated IL-6 and TCDCA Signaling.

The regenerative capacity of the liver is frequently insufficient following severe injury, and effective therapeutic strategies remain limited. Here, we identify CRX-527, a TLR4 agonist, as a novel inducer of hepatocyte proliferation in both healthy mice and a model of acetaminophen (APAP)-induced acute liver injury. Immunofluorescence analysis revealed that CRX-527-induced proliferative activity was predominantly observed in zone 2. Mechanistically, CRX-527 acts via TLR4 to induce transient IL-6 secretion, which subsequently activates the JAK2/STAT3 signaling pathway. In addition, metabolomic

Economic Evaluation of Prolonged Hypothermic Oxygenated Machine Perfusion Enabling Daytime Liver Transplantation.

BACKGROUND: Short-duration (1-2 h) dual hypothermic oxygenated machine perfusion (DHOPE) reduces ischemia-reperfusion injury-related complications. Prolonged DHOPE preservation (DHOPE-PRO) may facilitate logistics. We assessed the economic impact of DHOPE-PRO versus conventional short-duration DHOPE, hypothesizing that DHOPE-PRO would not be associated with increased one-year transplantation-related costs. METHODS: This post hoc economic evaluation of the prospective DHOPE-PRO trial (NL8740) included adult recipients of livers donated after brain death. All transplantation-related medical exp

Polystyrene nanoplastics potentiate cadmium-induced pyroptosis and apoptosis in primary duck hepatocytes: differential responses to N-acetylcysteine.

INTRODUCTION: Nanoplastics (NPs) can act as carriers for heavy metals, and co-exposure to NPs and co-pollutants may produce greater toxicity than single exposure. However, the combined toxicity of nanoplastics and heavy metals in poultry liver cells remains poorly understood. METHODS: Primary duck embryonic hepatocytes (PDEH) were used as an in vitro model. Cells were exposed to polystyrene nanoplastics (PS-NPs, 80 nm, 10 μg/mL) and/or cadmium chloride (CdCl2, 4 μg/mL) for 12 h. Pyroptosis- and apoptosis-associated endpoints were assessed by brightfield microscopy, PI staining, SEM, flow cyto

Phenotype-Anchored Multi-Omics Reveals Mechanisms Underlying Starvation-Induced Body, Liver, and Gallbladder Discoloration in Rock Carp (Procypris rabaudi).

Visible changes in body and internal organ coloration can serve as practical indicators of nutritional and metabolic stress in aquaculture, but their physiological meaning remains poorly understood. Here, we used starvation as a controlled nutritional stress model to investigate the physiological and molecular changes associated with body color fading and liver and gallbladder discoloration in rock carp (Procypris rabaudi). To assess the effects of food deprivation (0-35 days), fish were examined using growth and biochemical assays, intestinal histology, as well as integrated hepatic transcrip
