Evaluating the effects of metronidazole, metformin, and onion extract on GAPDH activity and nitric oxide production in Blastocystis sp. cultures: a biochemical and therapeutic approach.

BACKGROUND: Blastocystis sp. is a highly prevalent anaerobic intestinal protozoan increasingly associated with gastrointestinal disorders. Its survival within the gut microenvironment depends on metabolic pathways such as glycolysis, where glyceraldehyde-3-phosphate dehydrogenase (GAPDH) plays a central role, alongside interactions with nitric oxide (NO) derived from host defenses and surrounding microbiota. The emergence of metronidazole resistance necessitates exploration of alternative therapeutic strategies. OBJECTIVE: To evaluate the in vitro effects of metronidazole, metformin, and Alli

Development of a novel organoid-based two-dimensional system to model the intestinal host-parasite interface in fasciolosis.

BACKGROUND: The earliest interactions between Fasciola hepatica newly excysted juveniles (FhNEJ) and the intestinal epithelium remain poorly understood, partly due to the limited availability of physiologically relevant in vitro models. Here, we establish a human duodenum organoid-derived epithelial monolayer (ODM) co-culture system to investigate the early host-parasite interface, providing a controlled experimental platform for analysing interactions between FhNEJ and the intestinal epithelium. METHODS: ODMs representing the human duodenum were co-cultured with either live FhNEJ or a tegume

Exploring Blastocystis: hidden complexity in a common intestinal microbe.

Blastocystis is an anaerobic stramenopile that colonizes the large intestine of humans and many animals worldwide. Small-subunit rRNA-based typing reveals deep genetic diversity: at least 44 genetic variants, namely, subtypes (STs), are recognized, with ST1-ST4 accounting for most human carriage. Its medical relevance remains contested. Blastocystis carriage is frequently asymptomatic, and while associations with non-specific gastrointestinal symptoms (e.g. diarrhoea, abdominal pain and bloating), irritable bowel syndrome and extraintestinal manifestations such as urticaria have been proposed.

Giardia duodenalis in domestic cats from the US and Canada: Prevalence and zoonotic potential.

Giardia duodenalis is a flagellated commensal protozoan that can infect both humans and animals. Infection can result in clinical signs such as diarrhea or, more often, the infection can be subclinical. Within the G. duodenalis species, there are several major genetic assemblages, A - H. Certain assemblages are species specific (C and D in dogs, F in cats) while assemblages A and B can infect humans as well as other mammals. This retrospective study used data collected from a commercial reference laboratory (Antech Diagnostics, Mars Petcare Science & Diagnostics). Real-time PCR data using feca

Trophic niche segregation in small pelagic fishes suggests mesopelagic pearlsides as the main transmission pathway of Anisakis larvae in a Patagonian food web.

Many parasites are transmitted through trophic interactions, yet the specific pathways connecting intermediate and definitive hosts often remain poorly understood in marine food webs. In the Southwestern Atlantic, the Argentine anchovy (Engraulis anchoita) has been considered a potential vector of larval Anisakis to higher trophic levels, but low infection levels in San Matías Gulf suggest alternative transmission routes in this gulf. We investigated the role of pearlsides (Maurolicus spp.), abundant mesopelagic fishes in the gulf, in the transmission of anisakid nematodes. Examination of 229

Pathogenicity and virulence of Giardia intestinalis.

Giardiasis, caused by Giardia intestinalis, is a prevalent intestinal protozoan infection worldwide. Clinical manifestations are variable, and many infections remain asymptomatic. Disease pathogenesis is multifactorial, involving brush border enzyme deficiency, apoptosis, dysbiosis and increased intestinal permeability. Advances in genomics, genetic tools, and model systems have substantially improved our understanding of the parasite's virulence mechanisms. Giardia exhibits considerable genetic diversity, with distinct assemblages associated with differences in parasite biology. Attachment is

Novel inhibitors against Cryptosporidium parvum through blocking its energy metabolism.

Through systematic structural optimization and modification of anti-Cryptosporidium parvum compound NSC158011 (N-1-naphthalenyl-2-(phenylthiol) ethane thioamide), we constructed a virtual compound library comprising 1,440 derivatives, and 29 derivatives were selected via molecular docking for chemical synthesis. Among them, eight derivatives exhibited significant inhibitory effects on LDH compared with NSC158011. In vitro examination found that two derivatives could markedly reduce the number of C. parvum in cells while exhibiting reduced toxicity. In vivo studies in model animals (IFN-γ knock

Differential Effect of Ivermectin on Intestinal Helminths in Multimmate Mouse (Mastomys natalensis).

Mastomys natalensis is a widespread rodent species in sub-Saharan Africa and is increasingly recognised as an important component of food systems in some communities. However, rodents may harbour gastrointestinal helminths with potential implications for animal health, food safety, and zoonotic disease transmission. Information on gastrointestinal helminth infections in M. natalensis and the response of these parasites to anthelmintic treatment is limited. This study investigated the prevalence and diversity of gastrointestinal helminths in M. natalensis and evaluated changes in helminth egg b

Automated detection and classification of intestinal protozoan cysts using a nuclei structure-based deep learning framework for epidemiological surveillance.

Intestinal parasitic infections (IPIs) remain a critical global public health concern, particularly for epidemiological surveillance in resource-limited tropical regions. Standard laboratory diagnostics heavily rely on the manual microscopic examination of stool samples, a process that is notoriously time-consuming, labor-intensive, and highly dependent on the scarce availability of experienced parasitologists. To alleviate medical workloads and enhance community-wide screening efficiency, reliable, automated diagnostic tools are urgently needed. From an epidemiological control perspective, su
